The growing discovery and development of novel radiopharmaceuticals and radiolabeling methods requires an increasing capacity for radiochemistry experiments. However, such studies typically rely on radiosynthesizers designed for clinical batch production rather than research, greatly limiting throughput. Two general solutions are being pursued to address this: developing new synthesis optimization algorithms to minimize how many experiments are needed, and developing apparatus with enhanced experiment throughput. We describe here a novel high-throughput system based on performing arrays of droplet-based reactions at 10 μL volume scale in parallel. The automatic robotic platform can perform a set of 64 experiments in ~3 h (from isotope loading to crude product, plus sampling onto TLC plates), plus ~1 h for off-line radio-TLC analysis and radioactivity measurements, rather than the weeks or months that would be needed using a conventional system. We show the high repeatability and low crosstalk of the platform and demonstrate optimization studies for two 18F-labeled tracers. This novel automated platform greatly increases the practicality of performing arrays of droplet reactions by eliminating human error, vastly reducing tedium and fatigue, minimizing radiation exposure, and freeing up radiochemist time for more intellectually valuable pursuits. © 2023, The Authors. Evolution of “Michigan Probes” has enabled unraveling the connectivity of neurons at cellular resolution and their dynamic interaction within and across brain areas that underlie behavioral and cognitive functions. Recently, micro-LEDs were monolithically integrated for optogenetic neuromodulation at scale without requiring external optical fiber connection. The ‘hectoSTAR’ micro-LED optoelectrode features 256 recording electrodes and 128 stimulation micro-LEDs in four silicon micro-needle shanks, covering a large volume with 1.3-mm × 0.9-mm cross-sectional area located as deep as 6 mm inside the brain. Technical challenges to overcome in scaling of optogenetic probes will be discussed, followed by future direction.

Absorption rates for traditional adsorbents were predominately determined by the concentration gradient from bulk to absorbents and surface area. A novel sandwich-structured capacitor (SSC) containing adsorbents is designed to reduce the time consuming of absorption of pollutants by dielectrophoresis force (FDEP). Graphene oxide (GO) are coated on polystyrene microspheres (PSs) as the core/shell PS@GO adsorbents that were encapsulated within the SSC to treat the solution of dye-emulsified micells (DEM). The FDEP results the assembling of PS@GOs and DEMs under polarization and enhance the adsorption rate ca. 1 ∼ 2 times with polarization at 3 V and 200 kHz. Additionally, most adsorbents are recycled with secondary pollution. Only two-fifth pure water volume of the wastewater through the SSC could achieve the recycling of the adsorbents at 900 kHz. The high complex permittivity of GO coating at high frequency allows ultrafast alternative spinning of the induced dipoles in the PS@GOs facilitates desorption and hinders the re-adsorption of the DEM during the recycling of PS@GOs. The PS@GOs was reused ca. 10 times at 20 mL min−1 of flow rate through the SSC without damages between 200 and 900 kHz. The designed SSC containing PS@GOs accomplishes either reusing of adsorbents or concentrating of dyes with low secondary pollution in practice through frequency manipulation. © 2023 Elsevier B.V.

Interpretable ECG Analysis for Myocardial Infarction Detection through Counterfactuals

In the evolving landscape of ECG signal analysis, the challenge of limited transparency in machine learning models remains a significant barrier to their effective integration into clinical practice. This study addresses this issue by investigating the use of counterfactual explanations to improve model interpretability for clinicians, particularly in differentiating healthy subjects from myocardial infarction patients. Utilizing the PTB-XL dataset, we developed a methodology for systematic feature extraction and refinement to prepare for counterfactual analysis. This led to the creation of the Visualizing Counterfactual Clues on Electrocardiograms (VCCE) method, designed to improve the practicality of counterfactual explanations in a clinical setting. The validity of our approach was assessed using custom metrics that reflect the diagnostic relevance of counterfactuals, evaluated with the help of two cardiologists. Our findings suggest that this approach could support future efforts in using ECGs to predict patient outcomes for cardiac conditions, achieving interpretation validity scores of 23.29 ± 1.04 and 20.28 ± 0.99 out of 25 for high and moderate-quality interpretations, respectively. Clinical alignment scores of 0.83 ± 0.12 for high-quality and 0.57 ± 0.10 for moderate-quality interpretations underscore the potential clinical applicability of our method. The methodology and findings of this study contribute to the ongoing discussion on enhancing the interpretability of machine learning models in cardiology, offering a concept that bridges the gap between advanced data analysis techniques and clinical decision-making. The purpose of this letter is to study the design and explore vertically stacked complementary tunneling field-effect transistors (CTFETs) using CFET technology for emerging technology nodes. As a prior work, the CTFET’s device-level simulations are implemented and deliberated in strict compliance with the experimental settings. This work comprises the study of physical and DC analyses by scaling the p- to n-CTFET separation (D pn), being a significant factor in CFET/CTFET design for its process difficulty. By utilizing the 50% benefit in footprint, the work is further extended to CTFET static random access memory implementation and characterization with hold/read noise margin analysis. © 2023 The Author(s). Diabetes mellitus is one of the most pressing health concerns because so many people are afflicted by its disabling symptoms. Factors such as age, excess body fat, insufficient physical activity, a history of diabetes in one's family, a sedentary lifestyle, an unhealthy diet, hypertension, etc., all increase the likelihood of developing diabetes mellitus. Health complications are more common in people with diabetes, including cardiovascular disease, renal failure, stroke, blindness, and nerve injury. To validate a diagnosis of diabetes, hospitals typically perform a battery of procedures on the patient. Big data analytics has many vital applications in the healthcare sector. Numerous large computer systems are used in the healthcare sector. With the help of big data analytics, researchers can sift through mountains of data in search of previously unseen patterns and insights. Current techniques have a poor degree of precision in classification and forecast. While previous research has focused on factors such as glucose, body mass index, age, insulin, etc., the proposed model takes these into account and also the other factors that may be more relevant to the development of diabetes. The newer sample is superior to the older one based on categorization accuracy. A workflow algorithm for diabetes prognosis is also required to improve the accuracy.

The clinical utility of chemotherapy is often compromised by its limited efficacy and significant side effects. Addressing these concerns, we develop a self-assembled nanomicelle, namely SANTA FE OXA, which is composed of hyaluronic acid (HA) conjugated with ferrocene methanol (FC-OH), oxaliplatin prodrug (OXA(Ⅳ)) and glycol-coupled linoleic acid (EG-LA). Targeted delivery is achieved as HA binds to the CD44 receptors that are overexpressed on tumor cells, facilitating drug uptake. Once internalized, hyaluronidase (HAase) catalyzes the digestions of the SANTA FE OXA, releasing FC and reducing OXA(Ⅳ) into an active form. Active OXA induces DNA damage, while simultaneously promoting intracellular hydrogen peroxide levels via cascade reactions. In parallel, FC disrupts the redox balance within tumor cells, inducing ferroptosis. The synergistic combination of cascade chemotherapy and self-sensitized ferroptosis therapy has demonstrated remarkable anti-cancer efficacy in both in vitro and in vivo models. Moreover, this SANTA FE OXA significantly mitigates the systemic toxicity commonly associated with platinum-based chemotherapeutics. Our findings suggest a compelling advancement in nanomedicine for enhanced cascade cancer therapy. © 2024, The Authors. All rights reserved.

Influence of shear waves on transcranial ultrasound propagation in cortical brain regions

Transcranial ultrasound applications require accurate simulations to predict intracranial acoustic pressure fields. The current gold standard typically consists of calculating a longitudinal ultrasound wave propagation using a fluid skull model, which is based on full head CT images for retrieving the skull’s geometry and elastic constants. Although this approach has extensively been validated for deep brain targets and routinely used in transcranial ultrasound ablation procedures, its accuracy in shallow cortical regions remains unexplored. In this study, we explore the shear wave effects associated with transcranial focused ultrasound propagation, both numerically and experimentally. The intracranial acoustic pressure was measured at different incidence angles at the parietal and frontal regions in an ex vivo human skull. The fluid-like skull model was then compared to the solid model comprising both longitudinal and shear waves. The results consistently show a larger error and variability for both models when considering an oblique incidence, reaching a maximum of 125% mean deviation of the focal area when employing the fluid skull model. Statistical assessments further revealed that ignoring shear waves results in an average ~40% overestimation of the intracranial acoustic pressure and inability to obtain an accurate intracranial acoustic pressure distribution. Moreover, the solid model has a more stable performance, even when small variations in the skull-transducer relative position are introduced. Our results could contribute to the refinement of the transcranial ultrasound propagation modeling methods thus help improving the safety and outcome of transcranial ultrasound therapy in the cortical brain areas. The Pull of Tissue engineering: A STEM Outreach program with a Modular Cyclic Stretch Device to Engage High School Students

In this paper we designed a STEM outreach program with a modular tissue engineering education tool (we refer to as “modular cyclic-stretch device”) to engage high school students in STEM hands-on activities. Using simple machines such as gears and rotating cams, students were able to build a custom device to apply cyclic stretch to biomaterials. With the help of this hands-on activity, our outreach program helped students grasp tendon tissue biomechanics and understand the importance of applying biomechanical force to regenerate tendon tissue in a laboratory setting. The two-day outreach program comprised: 1. pre- and post-tests; 2. lectures; 3. laboratory sessions, including the microscopic examination of stained tissue sections and a hands-on group activity employing the modular cyclic-stretch device; and 4. homework. Assessment results suggest that our program supports improved student awareness and interest in tissue engineering as a future profession. The program elevated students’ confidence in their ability to apply engineering principles to tasks such as building a modular cyclic-stretch device and measuring the mechanical properties of biological tissues. Building an educational bioreactor improved students’ understanding of the dynamic nature of the human body and the importance of tissue engineering as an emerging discipline towards replacing or regenerating damaged organs. We propose that our modular device has great outreach potential to introduce tissue engineering concepts to high school and potentially college freshmen engineering students.
