The design of enzymes with complex active sites that mediate multistep reactions remains an outstanding challenge. With serine hydrolases as a model system, we combined the generative capabilities of RFdiffusion with an ensemble generation method for assessing active site preorganization at each step in the reaction to design enzymes starting from minimal active site descriptions. Experimental characterization revealed catalytic efficiencies (kcat/Km) up to 2.2 × 105 M-1 s-1 and crystal structures that closely match the design models (Cα root mean square deviations <1 angstrom). Selection for structural compatibility across the reaction coordinate enabled identification of new catalysts remove with five different folds distinct from those of natural serine hydrolases. Our de novo approach provides insight into the geometric basis of catalysis and a roadmap for designing enzymes that catalyze multistep transformations.

We present the task of Automated Punishment Extraction (APE) in sentencing decisions from criminal court cases in Hebrew. Addressing APE will enable the identification of sentencing patterns and constitute an important stepping stone for many follow up legal NLP applications in Hebrew, including the prediction of sentencing decisions. We curate a dataset of sexual assault sentencing decisions and a manuallyannotated evaluation dataset, and implement rulebased and supervised models. We find that while supervised models can identify the sentence containing the punishment with good accuracy, rulebased approaches outperform them on the full APE task. We conclude by presenting a first analysis of sentencing patterns in our dataset and analyze common models' errors, indicating avenues for future work, such as distinguishing between probation and actual imprisonment punishment. We will make all our resources available upon request, including data, annotation, and first benchmark models. © 2021 Association for Computational Linguistics.

Fano resonances in photonics arise from the coupling and interference between two resonant modes in structures with broken symmetry. They feature an uneven and narrow and tunable lineshape, and are ideally suited for optical spectroscopy. Many Fano resonance structures have been suggested in nanophotonics over the last ten years, but reconfigurability and tailored design remain challenging. Herein, we propose an all-optical "pick-and-place" approach aimed at assemble Fano metamolecules of various geometries and compositions in a reconfigurable manner. We study their coupling behavior by in-situ dark-field scattering spectroscopy. Driven by a light-directed opto-thermoelectric field, silicon nanoparticles with high quality-factor Mie resonances (discrete states) and low-loss BaTiO3 nanoparticles (continuum states) are assembled into all-dielectric heterodimers, where distinct Fano resonances are observed. The Fano parameter can be adjusted by changing the resonant frequency of the discrete states or the light polarization. We also show tunable coupling strength and multiple Fano resonances by altering the number of continuum states and discrete states in dielectric heterooligomers. Our work offers a general design rule for Fano resonance and an all-optical platform for controlling Fano coupling on demand. Copyright © 2024, The Authors. All rights reserved.

Lymphomas, a diverse group of blood cancers originating from the lymphatic system, present significant challenges for diagnosis and treatment due to their morphological and genetic differences. These variability obscures the development of personalized therapies and impacts disease outcomes. The scarcity of high-quality, diverse images of lymphoma cells is further hindered by patient privacy concerns, the rarity of certain types, and logistical challenges of collecting and processing biomedical images. Additionally, the costs and technical difficulties of microscopic imaging limit the scope of research. This paper addresses these challenges by utilizing generative adversarial networks (GANs) to generate synthetic yet realistic images of lymphoma histopathology and classify them in a low resource setting. Focusing specifically on chronic lymphocytic leukemia (CLL), follicular lymphoma (FL), and mantle cell lymphoma (MCL), we employed GANs to enhance the understanding of lymphoma variability and support advancements in digital cancer biology and pathology. This dual approach of image generation and classification aims to improve disease diagnosis and prognosis, ultimately contributing to better patient outcomes. © 2024, The Authors. Semantic communication technologies enable wireless edge devices to communicate effectively by transmitting semantic meaning of data. Edge components, such as vehicles in next-generation intelligent transport systems, use well-trained semantic models to encode and decode semantic information extracted from raw and sensor data. However, the limitation in computing resources makes it difficult to support the training process of accurate semantic models on edge devices. As such, edge devices can buy the pretrained semantic models from semantic model providers, which is called "semantic model trading". Upon collecting semantic information with the semantic models, the edge devices can then sell the extracted semantic information, e.g., information about urban road conditions or traffic signs, to the interested buyers for profit, which is called "semantic information trading". To facilitate both types of the trades, effective incentive mechanisms should be designed. Thus, in this paper, we propose a hierarchical trading system to support both semantic model trading and semantic information trading jointly. The proposed incentive mechanism helps to maximize the revenue of semantic model providers in the semantic model trading, and effectively incentivizes model providers to participate in the development of semantic communication systems. For semantic information trading, our designed auction approach can support the trading between multiple semantic information sellers and buyers, while ensuring individual rationality, incentive compatibility, and budget balance, and moreover, allowing them achieve higher utilities than the baseline method. Personnel and education data very important for organizations, including the military. All data related to military personnel will give effect positions and careers in the military field. The integration of personnel and education data can assist in processing and decision making. An analysis is needed so that the developed system can be useful for the long term. Analysis with IT balanced scorecard and COBIT® 2019, will be able to help and fill the gaps in the existing system based on the objectives of the military organization itself. It is shown that the domains BAI05 (Managed Organizational Change) and DSS02 (Managed Service Requests and Incidents) are still not being implemented well. The system developed will have the features and capabilities to support the organization in achieving its goals, including in the field of staffing and education. Besides, it provides convenience in making decisions in terms of positions related to personnel careers. © 2020 IEEE.

When implementing Markov Chain Monte Carlo (MCMC) algorithms, perturbation caused by numerical errors is sometimes inevitable. This paper studies how perturbation of MCMC affects the convergence speed and Monte Carlo estimation accuracy. Our results show that when the original Markov chain converges to stationarity fast enough and the perturbed transition kernel is a good approximation to the original transition kernel, the corresponding perturbed sampler has similar convergence speed and high approximation accuracy as well. We discuss two different analysis frameworks: ergodicity and spectral gap, both are widely used in the literature. Our results can be easily extended to obtain non-asymptotic error bounds for MCMC estimators. We also demonstrate how to apply our convergence and approximation results to the analysis of specific sampling algorithms, including Random walk Metropolis and Metropolis adjusted Langevin algorithm with perturbed target densities, and parallel tempering Monte Carlo with perturbed densities. To model the periodicity of beats, state-of-the-art beat tracking systems use "post-processing trackers" (PPTs) that rely on several empirically determined global assumptions for tempo transition, which work well for music with a steady tempo. For expressive classical music, however, these assumptions can be too rigid. With two large datasets of Western classical piano music, namely the Aligned Scores and Performances (ASAP) dataset and a dataset of Chopin's Mazurkas (Maz-5), we report on experiments showing the failure of existing PPTs to cope with local tempo changes, thus calling for new methods. In this paper, we propose a new local periodicity-based PPT, called predominant local pulse-based dynamic programming (PLPDP) tracking, that allows for more flexible tempo transitions. Specifically, the new PPT incorporates a method called "predominant local pulses" (PLP) in combination with a dynamic programming (DP) component to jointly consider the locally detected periodicity and beat activation strength at each time instant. Accordingly, PLPDP accounts for the local periodicity, rather than relying on a global tempo assumption. Compared to existing PPTs, PLPDP particularly enhances the recall values at the cost of a lower precision, resulting in an overall improvement of F1-score for beat tracking in ASAP (from 0.473 to 0.493) and Maz-5 (from 0.595 to 0.838). © 2023, CC BY.
