E3 ubiquitin ligase UBR5 modulates circadian rhythm by facilitating the ubiquitination and degradation of the key clock transcription factor BMAL1

The circadian clock is the inner rhythm of life activities and is controlled by a self-sustained and endogenous molecular clock,which maintains a～24 h internal *** the core element of the circadian clock,BMAL1 is susceptible to degradation through the ubiquitin-proteasome system(UPS).Nevertheless,scant information is available regarding the UPS enzymes that intricately modulate both the stability and transcriptional activity of BMAL1,affecting the cellular circadian *** this work,we identify and validate UBR5 as a new E3 ubiquitin ligase that interacts with BMAL1 by using affinity purification,mass spectrometry,and biochemical ***5 overexpression induced BMAL1 ubiquitination,leading to diminished stability and reduced protein level of BMAL1,thereby attenuating its transcriptional *** with this,UBR5 knockdown increases the BMAL1 *** mapping discloses that the C-terminus of BMAL1 interacts with the N-terminal domains of ***,cell-line-based experiments discover that HYD,the UBR5 homolog in Drosophila,could interact with and downregulate CYCLE,the BMAL1 homolog in ***2-luciferase bioluminescence real-time reporting assay in a mammalian cell line and behavioral experiments in Drosophila reveal that UBR5 or hyd knockdown significantly reduces the period of the circadian ***,our work discovers a new ubiquitin ligase UBR5 that regulates BMAL1 stability and circadian rhythm and elucidates the underlying molecular *** work provides an additional layer of complexity to the regulatory network of the circadian clock at the post-translational modification level,offering potential insights into the modulation of the dysregulated circadian rhythm.

Understanding the research landscape of any domain is of primary importance at all stages of a concept's evolution. Research in the domain of analytics has increased exponentially in the past decade, analytics is a tool being applied to increase effectiveness and efficiency within all domains. Similarly, there are numerous research studies that have applied analytics to the domain of higher education. Assessing research activity is important for planning future research and identifying the way forward to make meaningful contributions. The objective of this paper is to examine the current research in this domain and to identify emerging areas of research to make meaningful contributions. Bibliometric analysis was applied using Scopus. Documents on analytics and higher education keywords in business, accounting and management and published in journals were considered for the analysis. Vos Viewer was used for bibliometric analysis in the area, to identify emerging research themes and to understand the scope of applying analytics in higher education. The search query found 1635 documents when using the keywords of “analytics” and “higher education”. This was further narrowed down to research in Business Management and Accounting and using Journal articles and conference articles, finally yielding 103 articles. After a primary read to remove articles misplaced in the group the final number of articles include for the bibliometric analysis came down to 98. There has been an increase in publications since 2018 with interest increasing post pandemic in the area. The research categories are Student outcomes and perspective, academician outcomes and perspective, design of learning systems, etc. Based on this, it is recommended that research should focus on theoretical aspects of applying analytics to HEIs, ethical issues in data collection, etc..

Significant privacy and security concerns apply to the Internet of Things. IoT issues might include driver ignorance, inadequate device updates, a lack of robust and effective safeguards, and unreliable dynamic instrument management. The study’s objectives include identifying (a) various cybersecurity concern regarding, (b) techniques for attempting to secure IoT-enabled options and buildings, (c) available at the moment security remedies, and (d) the much more acceptable privacy depictions for the differing stages of technology apps. Researchers proposed a new, generally expanded layered paradigm that included security and privacy tools as well as layer documenting. the recommended virtualized Internet of Things system’s deployment and evaluation. Internet of Things (IoT) units built as virtualized using Amazon Web Services make the bottom layer (AWS). The topcoat was developed by researchers utilising AWS’s cloud-based IoT platform (the cloud). To allow data transfer between the envisioned cloud/edge authorised IoT representation’s levels, researchers have created ssl certificates.

Unlike healthy, non-transformed cells, the proteostasis network of cancer cells is taxed to produce proteins involved in tumor development. Cancer cells have a higher dependency on molecular chaperones to maintain proteostasis. The chaperonin T-complex protein ring complex(TRiC) contains eight paralogous subunits(CCT1-8), and assists the folding of as many as 10% of cytosolic *** is essential for the progression of some cancers, but the roles of TRiC subunits in osteosarcoma remain to be explored. Here, we show that CCT4/TRiC is significantly correlated in human osteosarcoma,and plays a critical role in osteosarcoma cell survival. We identify a compound anticarin-β that can specifically bind to and inhibit CCT4. Anticarin-β shows higher selectivity in cancer cells than in normal cells. Mechanistically, anticarin-β potently impedes CCT4-mediated STAT3 maturation. Anticarin-β displays remarkable antitumor efficacy in orthotopic and patient-derived xenograft models of ***, our data uncover a key role of CCT4 in osteosarcoma, and propose a promising treatment strategy for osteosarcoma by disrupting CCT4 and proteostasis.

Enterprises must use human resource management to create the difference in the fierce competition of today’s business environment. Employees are an enterprise’s most important assets, and it is important for administrators to grasp the psychology of their staff. Modern technology provided by mobile instant messaging (IM) has created a communication revolution, creating a culture of "texting rather than talking" through the use of smart phone mobile applications, such as WhatsApp, Line, and WeChat. Therefore, it is important for researchers to consider the influence of IM on work relationships and communication. This study explores the relationships between organizational trust, organizational justice, and organizational spirituality in the context of IM communication. Structural equation modeling was used to analyze questionnaire data, showing that organizational spirituality predicted organizational trust and organizational justice. The findings suggested that organizations should utilize the benefits of IM, create an organizational justice strategy, offer better human resource management, and create an environment in which employees’ organizational spirituality can be enhanced. © 2019, ICST Institute for Computer Sciences, Social Informatics and Telecommunications Engineering.

Institutional networking capability as a catalyst for sustainable supply chains in the manufacturing sector of Ghana: The role of green human resource management strategy and green dynamic capability

This study explores the influence of Green human resource Management Strategy (GHRMS) on supply chain environmental sustainability in manufacturing SMEs, emphasising the mediating role of green dynamic capability and the moderating effect of institutional networking capability. Quantitative data was gathered from 263 owners and managers in Ghana's manufacturing sector. The Covariant-Based Structural Equation Modelling (CB-SEM) approach within Amos (v.23) was employed to test the proposed hypotheses. The findings revealed that GHRMS significantly enhances supply chain environmental sustainability by fostering eco-conscious HR practices. Green dynamic capability partially mediates this relationship, enabling firms to develop and adapt sustainable competencies. Additionally, institutional networking capability strengthens the indirect effect of GHRMS on supply chain sustainability, demonstrating the crucial role of external networks in achieving environmental goals. These findings underscore the importance of both internal organisational capabilities and external collaborations in driving sustainable supply chain practices in emerging economies. This study provides strategic insights for firms aiming to integrate GHRMS into their environmental sustainability initiatives, fostering a greener and more resilient supply chain ecosystem. © 2025 Elsevier Ltd

We report the first laboratory and interstellar detection of the α-cyano vinyl radical (H2CCCN). This species was produced in the laboratory by an electric discharge of a gas mixture of vinyl cyanide, CH2CHCN, and Ne. Its rotational spectrum was characterized using a Balle-Flygare narrowband-type Fourier-transform microwave spectrometer operating in the frequency region of 8-40 GHz. The observed spectrum shows a complex structure due to tunneling splittings between two torsional sublevels of the ground vibronic state, 0+ and 0-, derived from a large-amplitude inversion motion. In addition, the presence of two equivalent hydrogen nuclei makes it necessary to discern between ortho- and para-H2CCCN. A least-squares analysis reproduces the observed transition frequencies with a standard deviation of ca. 3 kHz. Using the laboratory predictions, this radical was detected in the cold dark cloud TMC-1 using the Yebes 40 m telescope and the QUIJOTE1 line survey. The 40,4-30,3 and 50,5-40,4 rotational transitions, composed of several hyperfine components, were observed in the 31.0-50.4 GHz range. Adopting a rotational temperature of 6 K, we derived a column density of (1.4±0.2)*1011 cm-2 and (1.1±0.2)s*1011 cm-2 for ortho-H2CCCN and para-H2CCCN, respectively. The reaction of C + CH3CN emerges as the most likely route to H2CCCN in TMC-1, and possibly that of N + CH2CCH as well. © The Authors 2023.

The STEAM interdisciplinary integrated-thematic curriculum with engineering design is the main concept of the 12-year basic education curricula of technology. This research is based on the engineering design orientation as a teaching strategy, developing project for paper mechanism, and implementing Living technology courses for gifted high school students in Taiwan to test the teaching effectiveness. In the project, students need to integrate the theory, using engineering design process methods, performing work prediction and evaluation, prototyping experiments and trial work, and finally optimizing the work and improving the design. The results of the present study facilitate us get deeper understanding of the practical implementation and engineering thinking skills of gifted students. Suggestions based on the results were generated to improve the future Living technology curriculum. The findings in this study serve as a reference for the future design, development, and implementation of STEAM curricula.

Evidence for the existence of monkeys with spontaneous Parkinson’s disease (PD) has been lacking. Here,we screened macaque colonies at the Kunming Primate Research Center(Association for Assessment and Accreditation of Laboratory Animal Care(AAALAC) accredited), Kunming Institute of Zoology, Chinese Academy of Sciences, which has a large population of monkeys (>2400), to identify naturally occurring PD monkeys.

In recent years, the Internet Celebrities (ICs) have emerged as the most influential factor on adolescents. Although ICs are simply other users uploading their user-generated content (UGC), they attract followers through their unique combination of similarity, sociability, and a sense of being different from the audience. While ICs were widely discussed in fields like marketing and youth culture, few researches studied their education value. Our study utilize Uses and Gratifications Theory to extract students’ perceptive motivation to learn from ICs’ videos. We surveyed 101 high school students about their motivations to use ICs’ video to learn enterprise finance. These students considered Entertainment, Convenience, and Information were major gratifications they would get by learning from ICs’ videos. We also found past experience with ICs’ videos did influence student’s perceptive Escapism gratification. © Springer Nature Switzerland AG 2019.
