The bibliography includes articles dealing with the simultaneous independent shared use of computers. Real time, on-line, memory or facility allocation, multi-programming, and multi-processing terminology is evolved. Though a number of computer applications are reported, the requirements of special or dedicated time-sharing systems seem to be within the capability of most of the general-purpose time-sharing systems which are beginning to emerge. The systems are being designed for 100 percent availability, memory and facility allocation among users, a common file or data base, and direct access by the users via consoles (for example, typewriter, display, etc.).

The optimization of memory hierarchy involves the selection of types and sizes of memory devices such that the average access time to an information block is a minimum for a particular cost constraint. It is assumed that the frequency of usage of the information is known a priori. In this paper the optimization theory for a single task or program is reviewed and it is extended to a general case in multiprogramming when a number of tasks are executed concurrently. Another important extension treats the case when memories are available only in indivisible modules. Comparisons with conventional methods of solution as well as computational experience on the multiprogrammed and modular cases are given.

I have been using the first edition of this book as a text for a number of years. This was in a Stanford University first-year graduate course that is taken by students from electricalengineering or computerscience who are interested in computer organization. Because computer tech nology has been changing so rapidly, it became necessary to supplement the text with additional readings. My colleagues and I examined many newly-published books for possible use as texts. We found no book with the same excellent choice of topics and thorough coverage as Dr. Gschwind's first edition. Springer-Verlag's request that I prepare a second edition of this book came at a time when I had many other projects underway. Before I de cided whether to take on the project of preparing a revision, I asked many of my students for their opinions of Dr. Gschwind's first edition. Even I was surprised by the enthusiasm that this rather skeptical and critical group of students displayed for the book. It was this enthusiasm that convinced me of the value and importance of preparing the revision.

Proteolysis plays critical roles in normal and pathologic physiology; these enzymes are intricately involved in cancer progression and spread. Our understanding of protease function has advanced from nonspecific degrading enzymes to a modern appreciation of their diverse roles in posttranslational modification and signaling in a complex microenvironment. This new understanding has led to next-generation diagnostics and therapeutics that exploit protease activity in cancer. For diagnostics, protease activity may be measured as a biomarker of cancer, with wide-ranging utility from early detection to monitoring therapeutic response. Therapeutically, while broad inhibition of protease activity proved disappointing, new approaches that more specifically modulate proteases in concert with secondary targets might enable potent combination therapies. In addition, clinical evaluation is underway for tools that leverage protease activity to activate therapeutics, ranging from imaging agents that monitor surgical margins to immunotherapies with improved specificity. Technologies that interact with, measure, or modulate proteases are poised to improve cancer management on diagnostic and therapeutic fronts to realize the promise of precision medicine.
