The value of preclinical diffusion MRI (dMRI) is substantial. While dMRI enables in vivo non-invasive characterization of tissue, ex vivo dMRI is increasingly being used to probe tissue microstructure and brain connectivity. Ex vivo dMRI has several experimental advantages including higher signal-to-noise ratio (SNR) and spatial resolution compared to in vivo studies, and enabling more advanced diffusion contrasts for improved microstructure and connectivity characterization. Another major advantage of ex vivo dMRI is the direct comparison with histological data, as a crucial methodological validation. However, there are a number of considerations that must be made when performing ex vivo experiments. The steps from tissue preparation, image acquisition and processing, and interpretation of results are complex, with many decisions that not only differ dramatically from in vivo imaging of small animals, but ultimately affect what questions can be answered using the data. This work represents "Part 2" of a 3-part series of recommendations and considerations for preclinical dMRI. We describe best practices for dMRI of ex vivo tissue, with a focus on the value that ex vivo imaging adds to the field of dMRI and considerations in ex vivo image acquisition. We first give general considerations and foundational knowledge that must be considered when designing experiments. We briefly describe differences in specimens and models and discuss why some may be more or less appropriate for different studies. We then give guidelines for ex vivo protocols, including tissue fixation, sample preparation, and MR scanning. In each section, we attempt to provide guidelines and recommendations, but also highlight areas for which no guidelines exist (and why), and where future work should lie. An overarching goal herein is to enhance the rigor and reproducibility of ex vivo dMRI acquisitions and analyses, and thereby advance biomedical knowledge. © 2022, CC BY.

Spam filters have become an important tool for ISP(Internet service Provider) to address the growing spam in cyberspace. Spam filtering is a program used to detect spam, prevent it and enter users' mailboxes. With the rapid growth of Internet technology, email communication is becoming increasingly important in people's daily lives. In addition, the amount of spam contained in emails has increased considerably in recent years. In this study, the authors will compare the performance of the decision tree classification models using or not using Recursive Feature Elimination (RFE) in spam filtering. The results of this test will be compared with the results of level accuracy, recall, precision, and F 1 Score using or not using the RFE method. where in the results of this level measurement, the model used can be a reference or solution to filter spam correctly, especially through the use of supervised learning data.

Friedreich's ataxia (FRDA) is a rare autosomal recessive inherited neurodegenerative disease which is the result of a triplet repeat expansion in the intronic region of the frataxin FXN gene resulting in depleted frataxin protein expression. Disease onset is usually in childhood and causes progressive damage to the nervous system resulting in progressive disability. This work uses computer aided classification techniques to identify which measures of the disease progression, including accurate determination of the shortest allele repeat length, are the most informative when trying to predict likely disease progression and prognosis. Further we investigate the possibility of a gender difference in the progression of the disease. Our results highlight the importance of accurate determination GAA repeat length in any clinical predictions showing that the number of repeats is the best prognostic tool in FRDA and is strongly linked to the age at onset disease. Further that there are possible gender dependent differences in cardiac measurements recorded from patients of similar age of onset and GAA repeat length. © 2019

Background: Despite major advances in artificial intelligence (AI) research for healthcare, the deployment and adoption of AI technologies remain limited in clinical practice. In recent years, concerns have been raised about the clinical, technical, ethical and legal risks associated with healthcare AI. To increase adoption in the real world, it is essential that AI technologies are trusted and accepted by patients, clinicians, health organisations and authorities. This paper describes the FUTURE-AI framework as the first international consensus guideline for trustworthy AI in healthcare. Methods: The FUTURE-AI consortium was founded in 2021 and now comprises 117 interdisciplinary experts from 50 countries representing all continents, including AI scientists, clinical researchers, biomedical ethicists, and social scientists. Over a two-year period, the consortium established guiding principles and best practices for trustworthy and deployable AI through an iterative process comprising an in-dept. literature review, a modified Delphi survey, and online consensus meetings. Findings: The FUTURE-AI framework was established based on six guiding principles for trustworthy AI in healthcare, i.e. Fairness, Universality, Traceability, Usability, Robustness and Explainability. Through consensus, a set of 30 best practices were defined, addressing technical, clinical, socio-ethical and legal dimensions of trustworthy AI. The recommendations cover the entire lifecycle of healthcare AI, from design, development and validation to regulation, deployment, and monitoring. Interpretation: FUTURE-AI is a structured, risk-informed framework which provides guidance for constructing healthcare AI tools that will be trusted, deployed and adopted in real-world clinical practice. Big data often has emergent structure that exists at multiple levels of abstraction, which are useful for characterizing complex interactions and dynamics of the observations. Here, we consider multiple levels of abstraction via a multiresolution geometry of data points at different granularities. To construct this geometry we define a time-inhomogeneous diffusion process that effectively condenses data points together to uncover nested groupings at larger and larger granularities. This inhomogeneous process creates a deep cascade of intrinsic low pass filters on the data affinity graph that are applied in sequence to gradually eliminate local variability while adjusting the learned data geometry to increasingly coarser resolutions. We provide visualizations to exhibit our method as a "continuously-hierarchical" clustering with directions of eliminated variation highlighted at each step. The utility of our algorithm is demonstrated via neuronal data condensation, where the constructed multiresolution data geometry uncovers the organization, grouping, and connectivity between neurons. Cluster analysis is an important exploratory tool for a broad range of applications including data analysis of biomedical datasets to uncover meaningful subgroups such as in autism spectrum disorder (ASD). For a given clustering algorithm, multiple results can be obtained on the same dataset by varying the algorithm parameters. In biomedical applications, discovering meaningful subgroups, not just the optimal number of clusters, is expedient. It is imperative to develop quality measures capable of identifying optimal partitions for a given dataset. In this paper, we apply varied clustering methods to subgroup an ASD simplex sample based on relevant phenotype features that may uncover meaningful subtypes. We present a detailed cluster validation analysis using an ensemble validation paradigm and visualization techniques. We present a rigorous clinical/behavioral analysis of the top highly ranked results. The evaluation demonstrated that both configurations yielded similar clinical significance results: 2-subgroups configuration with distinct clinical profile. © 2018 IEEE.

Small-animal diffusion MRI (dMRI) has been used for methodological development and validation, characterizing the biological basis of diffusion phenomena, and comparative anatomy. The steps from animal setup and monitoring, to acquisition, analysis, and interpretation are complex, with many decisions that may ultimately affect what questions can be answered using the resultant data. This work aims to present selected recommendations and guidelines from the diffusion community, on best practices for preclinical dMRI of in vivo animals. We describe the general considerations and foundational knowledge that must be considered when designing experiments. We briefly describe differences in animal species and disease models and discuss why some may be more or less appropriate for different studies. We then give guidelines for in vivo acquisition protocols, including decisions on hardware, animal preparation, and imaging sequences, followed by advice for data processing including pre-processing, model-fitting, and tractography. Finally, we provide an online resource which lists publicly available preclinical dMRI datasets and software packages, to promote responsible and reproducible research. In each section, we attempt to provide guides and recommendations, but also highlight areas for which no guidelines exist (and why), and where future work should focus. While we mainly cover the central nervous system (on which most preclinical dMRI studies are focused), we also provide, where possible and applicable, recommendations for other organs of interest. An overarching goal herein is to enhance the rigor and reproducibility of small animal dMRI acquisitions and analyses, and thereby advance biomedical knowledge. © 2022, CC BY-NC-ND.

We present dialogue management routines for a system to engage in multiparty agent-infant interaction. The ultimate purpose of this research is to help infants learn a visual sign language by engaging them in naturalistic and socially contingent conversations during an early-life critical period for language development (ages 6 to 12 months) as initiated by an artificial agent. As a first step, we focus on creating and maintaining agent-infant engagement that elicits appropriate and socially contingent responses from the baby. Our system includes two agents, a physical robot and an animated virtual human. The system's multimodal perception includes an eye-tracker (measures attention) and a thermal infrared imaging camera (measures patterns of emotional arousal). A dialogue policy is presented that selects individual actions and planned multiparty sequences based on perceptual inputs about the baby's internal changing states of emotional engagement. The present version of the system was evaluated in interaction with 8 babies. All babies demonstrated spontaneous and sustained engagement with the agents for several minutes, with patterns of conversationally relevant and socially contingent behaviors. We further performed a detailed case-study analysis with annotation of all agent and baby behaviors. Results show that the baby's behaviors were generally relevant to agent conversations and contained direct evidence for socially contingent responses by the baby to specific linguistic samples produced by the avatar. This work demonstrates the potential for language learning from agents in very young babies and has especially broad implications regarding the use of artificial agents with babies who have minimal language exposure in early life.
