Optimalization of enzymatic degradation on oil palm leaves hemicellulose

Indonesia is the largest country of oil palm production in the world. The oil palm by-product is still containing almost 80% of carbohydrates, such as cellulose and hemicellulose. This study aimed to optimize the enzymatic degradation of oil palm leaves in order to achieve zero waste of oil palm biomass by-product. Sodium hydroxide (NaOH) solution was used to isolate hemicellulose extraction from oil palm leaves. Various concentrations of NaOH such as 2 M, 2.5 M, 3 M, 3.5 M and 4 M and the reflux times of 2 hours, 4 hours, and 6 hours were used to optimize hemicellulose extraction. The optimum condition for hemicellulose extraction was 3 M NaOH and reflux time was 6 hours, while the yield at that condition was 48.94 % (w/w). The extracted hemicellulose is then hydrolyzed by a xylanolytic enzyme from recombinant Escherichia coli DH5α (namely pTP510). Temperatures of incubation were varied 50°C, 60°C, 70°C, and 80°C for 24 hours at pH 6. The hydrolysis result was analyzed using High Performance Liquid Chromatography (HPLC). Xylose was produced at an optimum temperature of 60°C (120.231 ppm), arabinose at 50°C (26.265 ppm) whereas xylo-oligosaccharide at 50°C (280.465 ppm).

It is essential to examine the physical or chemical properties of molecular gas in starburst galaxies to reveal the underlying mechanisms characterizing starbursts. We used non-negative matrix factorization (NMF) to extract individual molecular or physical components involved in the star formation process in NGC 253. We used images of 148 transitions from 44 different species of the ALMA large program ALCHEMI. Additionally, we included the continuum images at ALMA Bands 3 and 7 from the same dataset. For the five NMF components (NF1–5), we obtained that their distributions correspond to various basic phenomena related to star formation: i) low-density gas extended through the galactic central molecular zone (NF2), ii) shocks (NF3), iii) starburst regions (NF4), and iv) young star-forming regions (NF5). The other component (NF1) is related to excitation;three components obtained by NMF (NF3, 1, and 5) show a strong dependence upon the upper state energies of transitions, and represent low-, intermediate-, and high-excitation, respectively. We also compared our results using principal component analysis (PCA) previously applied to the same dataset. Molecular components extracted from NMF are similar to the ones obtained from PCA. However, NMF is better at extracting components associated with a single physical component, while a single component in PCA usually contains information on multiple physical components. This is especially true for features with weak intensities like emission from outflows. Our results suggest that NMF can be one of promising methods interpreting molecular line survey data, especially in the upcoming era of wide-band receivers. © 2024, CC BY.

An Integrated Method for the Selection of Optimum Locations for Landfilling Utilizing GIS: A Case Study of the State of Kuwait

With the rise of urbanization, change in lifestyles, and food habits, the amount of Municipal Solid Waste (MSW) has been rapidly increasing with the composition constantly changing. Landfilling is the common approach for waste management in Kuwait, and around 90% of municipal wastes are disposed in landfill sites. Therefore, this study focused on the integration of different methods for the process of allocating optimum sites for MSW landfills, in four main stages. The first stage focused on utilizing the gap analysis method for the identification of effective criteria for landfill site selection, by comparing the criteria in Kuwait with the World Bank guidelines, US EPA, and Australia EPA, to suggest the best criteria for landfill site selection. These criteria were applied in the second and third stages to determine suitable locations for landfill sites using the Geographical Information System (GIS). In the last stage, the suitable areas were evaluated by studying the socioeconomic aspects such as distance to waste generation center, the proximity of existing roads, and lifespan to select optimum locations for landfilling. The results indicated that only one site with a total area of 12 km2 was considered optimum for MSW landfill sites in Kuwait. Therefore, moderate suitability areas were indicated to support and provide decision makers with more options. The moderately suitable sites are also suitable for landfilling, but they need to be designed carefully in order to avoid negative impacts on the environment and human health. Therefore, we conclude that there is an urgent need to apply the integrated waste management system in Kuwait, as well as to develop a long-term strategic plan.

Leveraging the high resolution, sensitivity, and wide frequency coverage of the Atacama Large Millimeter/submillimeter Array (ALMA), the QUARKS survey, standing for 'Querying Underlying mechanisms of massive star formation with ALMA-Resolved gas Kinematics and Structures', is observing 139 massive star-forming clumps at ALMA Band 6 (λ ∼ 1.3 mm). This paper introduces the Atacama Compact Array (ACA) 7-m data of the QUARKS survey, describing the ACA observations and data reduction. Combining multi-wavelength data, we provide the first edition of QUARKS atlas, offering insights into the multiscale and multiphase interstellar medium (ISM) in high-mass star formation. The ACA 1.3 mm catalog includes 207 continuum sources that are called ACA sources. Their gas kinetic temperatures are estimated using three formaldehyde transitions with a non-LTE radiation transfer model, and the mass and density are derived from a dust emission model. The ACA sources are massive (16-84 percentile values of 6-160 M☉), gravity-dominated (M ∝ R1.1) fragments within massive clumps, with supersonic turbulence (M > 1) and embedded star-forming protoclusters. We find a linear correlation between the masses of the fragments and the massive clumps, with a ratio of 6% between the two. When considering the fragments as representative of dense gas, the ratio indicates a dense gas fraction (DGF) of 6%, although with a wide scatter ranging from 1% to 10%. If we consider the QUARKS massive clumps to be what is observed at various scales, then the size-independent DGF indicates a self-similar fragmentation or collapsing mode in protocluster formation. With the ACA data over four orders of magnitude of luminosity-to-mass ratio (L/M), we find that the DGF increases significantly with L/M, which indicates clump evolutionary stage. We observed a limited fragmentation at the subclump scale, which can be explained by dynamic global collapse process. © 2024, CC BY.

KAGRA, the underground and cryogenic gravitational-wave detector, was operated for its solo observation from February 25 to March 10, 2020, and its first joint observation with the GEO 600 detector from April 7 to April 21, 2020 (O3GK). This study presents an overview of the input optics systems of the KAGRA detector, which consist of various optical systems, such as a laser source, its intensity and frequency stabilization systems, modulators, a Faraday isolator, mode-matching telescopes, and a high-power beam dump. These optics were successfully delivered to the KAGRA interferometer and operated stably during the observations. The laser frequency noise was observed to limit the detector sensitivity above a few kilohertz, whereas the laser intensity did not significantly limit the detector sensitivity.

The rhKU_Sej_LRR_2271 protein is introduced as one of the leptospiral vaccine candidates since it contains the predicted immunogenic epitopes. The in-silico sequence and structural based analysis have been used in this study to analyze MHC class I and class II-restricted epitopes of the protein. Six epitope prediction programs were employed, which the epitopes with high prediction scores from each program were aligned. The 21 of potentially unrestrained epitopes with prediction scores above the cut-off value from at least two prediction programs were selected. The 3D-molecular modeling, docking, and molecular dynamic simulation were performed to evaluate the affinity binding between peptide-MHC complex and T-cell receptor. One promising epitope, which is 171-LLFLPLIKI, showed the potency in binding to both MHC class I and II alleles. Two newly designed peptides containing epitopes which can bind to over 3 of MHC alleles, LL17:171-LLFLPLIKILYVDRNKL-187 and SL19:209-SLNSGIKALPFNYEKLVNL-227, significantly increased interferon-gamma (IFNg)-producing specific T-cell responses in the rhKU_Sej_LRR_2271 immunized rabbits compared to nonimmunized rabbits. The LL17 peptide can also induce interferon-gamma-producing specific CD4⁺ T-cell responses in the immunized rabbits. For the evaluation of humoral immune responses, the immunized rabbits have a significantly greater amount of specific IgG in plasma than the nonimmunized rabbits. Ex-vivo study of T-cell responses in animal model using flow cytometry confirmed an accomplishment of the theoretically in-silico analysis for discovering potential T-cell epitopes of the protein. The results show that the rhKU_Sej_LRR_2271 protein containing promiscuous T-cell epitopes, which can induce both humoral and cell-mediated immune responses, is a prospective protein candidate for leptospiral vaccine development. © 2021 The Authors

Traditional thrombolytic therapeutics for vascular blockage are affected by their limited penetration into thrombi, associated off-target side effects, and low bioavailability, leading to insufficient thrombolytic efficacy. It is hypothesized that these limitations can be overcome by the precisely controlled and targeted delivery of thrombolytic therapeutics. A theranostic platform is developed that is biocompatible, fluorescent, magnetic, and well-characterized, with multiple targeting modes. This multimodal theranostic system can be remotely visualized and magnetically guided toward thrombi, noninvasively irradiated by near-infrared (NIR) phototherapies, and remotely activated by actuated magnets for additional mechanical therapy. Magnetic guidance can also improve the penetration of nanomedicines into thrombi. In a mouse model of thrombosis, the thrombosis residues are reduced by ≈80% and with no risk of side effects or of secondary embolization. This strategy not only enables the progression of thrombolysis but also accelerates the lysis rate, thereby facilitating its prospective use in time-critical thrombolytic treatment. © 2023 Wiley-VCH GmbH.

K2-2 b/HIP 116454 b, the first exoplanet discovery by K2 during its Two-Wheeled Concept Engineering Test, is a sub-Neptune (formula presented) orbiting a relatively bright (KS = 8.03) K-dwarf on a 9.1 day period. Unfortunately, due to a spurious follow-up transit detection and ephemeris degradation, the transit ephemeris for this planet was lost. In this work, we recover and refine the transit ephemeris for K2-2 b, showing a ∼40σ discrepancy from the discovery results. To accurately measure the transit ephemeris and update the parameters of the system, we jointly fit space-based photometric observations from NASA’s K2, TESS, and Spitzer missions with new photometric observations from the ground, as well as radial velocities from HARPS-N that are corrected for stellar activity using a new modeling technique. Ephemerides becoming lost or significantly degraded, as is the case for most transiting planets, highlights the importance of systematically updating transit ephemerides with upcoming large efforts expected to characterize hundreds of exoplanet atmospheres. K2-2 b sits at the high-mass peak of the known radius valley for sub-Neptunes, and is now well-suited for transmission spectroscopy with current and future facilities. The initial conditions found in infrared dark clouds (IRDCs) provide insights on how high-mass stars and stellar clusters form. We have conducted high-angular resolution and high-sensitivity observations toward thirty-nine massive IRDC clumps, which have been mosaicked using the 12 and 7 m arrays from the Atacama Large Millimeter/submillimeter Array. The targets are 70 μm dark massive (220-4900 M _⊙), dense (>10⁴ cm⁻³), and cold (∼10-20 K) clumps located at distances between 2 and 6 kpc. We identify an unprecedented number of 839 cores, with masses between 0.05 and 81 M _⊙ using 1.3 mm dust continuum emission. About 55% of the cores are low-mass (<1 M _⊙), whereas ≲1% (7/839) are high-mass (≳27 M _⊙). We detect no high-mass prestellar cores. The most massive cores (MMC) identified within individual clumps lack sufficient mass to form high-mass stars without additional mass feeding. We find that the mass of the MMCs is correlated with the clump surface density, implying denser clumps produce more massive cores. There is no significant mass segregation except for a few tentative detections. In contrast, most clumps show segregation once the clump density is considered instead of mass. Although the dust continuum emission resolves clumps in a network of filaments, some of which consist of hub-filament systems, the majority of the MMCs are not found in the hubs. Our analysis shows that high-mass cores and MMCs have no preferred location with respect to low-mass cores at the earliest stages of high-mass star formation. © 2023. The Author(s). The purpose of this study is to design a concept of energy management control system (EMCS) at the UNSADA campus, Jakarta, Indonesia by utilizing the results of the previous walkthrough audit. EMCS is an important first step by involving all parties on campus to be concerned the sustainable energy savings. The energy efficiency program utilizes the campus as an energy laboratory to identify, recommend, implement and evaluate the energy consumption, so that the expected energy efficiency targeted by the top managements can be achieved. This program is motivated by several reasons, such as;to support government policies in reducing dependence on fossil energy by implementing energy conservation programs;as a reflection of compliance with the implementation of educational service buildings (including the campus in Jakarta);a vision as the efficient campus in energy consumption that has been determined by the campus management. To achieve these goals, there are four stages to be done;the preparation, the conducting audit, the audit analysis, and the recommendation for implementation. The results provide an overview of the opportunities to save the energy consumption and to develop EMCS by involving all stack-holders on campus. © 2020 Published under licence by IOP Publishing Ltd.
