This paper studies the application of the adaptive neuro-fuzzy inference system (ANFIS). We propose the ANFIS used for region of interest (ROI) detection in brain fMRI images. The ROI is used for estimating the tissue activity by Blood-Oxygen Level Dependent technique (BOLD). The result shows the ANFIS model can be used to fit the image time-intensity data. A 3 Gaussian membership functions were proposed to estimate the time-intensity relation. The result shows the rate of time-intensity relation to determine the tissue activity. The tissue activity should be determined the normal pattern of reading process. © 2011 IEEE.

This paper studies the application of the non-linear model (ANN, ANFIS). ANN and ANFIS were used to fit a Blood-Oxygen Level Dependent (BOLD) signal with design architecture. A model proposes to define region of interest (ROI) in brain fMRI images. The result shows the ANFIS model can be represent to fit the BOLD data better than ANN. Three paradigms were stimulated at difference issue activation in specific parameter (activation time, duration and signal strength). In this case a higher frequency and amplitude paradigm has stimulated on the brain area activation faster and more frequently.

Background aims: Mesenchymal stromal cells (MSCs) are recognized as a potential cell-based therapy for regenerative medicine. Short-term inflammatory cytokine pre-stimulation (cytokine priming) is a promising approach to enhance regenerative efficacy of MSCs. However, it is unclear whether their intrinsic heterogenic nature causes an unequal response to cytokine priming, which might blunt the accessibility of clinical applications. Methods: In this study, by analyzing the single-cell transcriptomic landscape of human bone marrow MSCs from a naïve to cytokine-primed state, we elucidated the potential mechanism of superior therapeutic potential in cytokine-primed MSCs. Results: We found that cytokine-primed MSCs had a distinct transcriptome landscape. Although substantial heterogeneity was identified within the population in both naïve and primed states, cytokine priming enhanced the several characteristics of MSCs associated with therapeutic efficacy irrespective of heterogeneity. After cytokine-priming, all sub-clusters of MSCs possessed high levels of immunoregulatory molecules, trophic factors, stemness-related genes, anti-apoptosis markers and low levels of multi-lineage and senescence signatures, which are critical for their therapeutic potency. Conclusions: In conclusion, our results provide new insights into MSC heterogeneity under cytokine stimulation and suggest that cytokine priming reprogrammed MSCs independent of heterogeneity. © 2023 International Society for Cell & Gene Therapy

Urinary tract stones are a common and frequently recurring medical issue. Accurately predicting the success rate after surgery can help avoid ineffective medical procedures and reduce unnecessary healthcare costs. This study collected data from patients with upper ureter stones who underwent extracorporeal shock wave lithotripsy, including cases of successful as well as unsuccessful stone removal after the first and second lithotripsy procedures, and constructed prediction systems for the outcomes of the first and second lithotripsy procedures. Features were extracted from three categories of information: patient characteristics, stone characteristics, and extracorporeal shock wave lithotripsy machine data, and additional features were created using Feature Creation. Finally, the impact of features on the models was analyzed using six methods to calculate feature importance. Our prediction model for the first lithotripsy, selected from among 43 methods and seven ensemble learning techniques, achieves an AUC of 0.91. For the second lithotripsy, the AUC reaches 0.76. The results indicate that the detailed and binary information provided by patients regarding their history of stone experiences contributes differently to the predictive accuracy of the first and second lithotripsy procedures. Macular pigments produced from microalga Chlorella sp. and applied to alleviate the pathogenic process in diabetic mice

Macular pigments (MP), which consist of lutein and zeaxanthin, have antioxidant capabilities and are widely used in eye healthcare. However, few studies have focused on the protective mechanisms of MP against diabetes mellitus. In addition, the advantages of MP produced by microalgae include non-competition with grains for land, year-round harvesting, and rapid growth. Therefore, this study aims to investigate the MP produced and extracted from the microalga Chlorella sp. AT1 and assess its impact on mitigating retinopathy and atherosclerotic damage in diabetic apolipoprotein E -deficient mice (ApoE −/− ) subjected to high-fat diet and streptozotocin treatment. The results showed that the major components of MP were lutein and zeaxanthin in a 5:1 ratio. MP supplementation significantly reduced glucose tolerance and insulin resistance by 25 and 41 %, respectively, in comparison to the diabetic group. MP supplementation resulted in the moderation of various inflammatory cytokines and lowered the serum levels of disease markers in comparison to the control group. In addition, the expression levels of cellular antioxidant enzymes were significantly elevated, aligning with a reduction in lipid peroxidation after MP supplementation, in comparison to the diabetic group. Pathological changes in the outer nuclear layer and aortic plaques in diabetic ApoE −/− mice were significantly alleviated by MP supplementation. In summary, microalgal MP alleviated the symptoms of type 2 diabetes mellitus in the diabetic ApoE −/− mice. Consequently, MP holds the potential to serve as a beneficial ingredient in healthy food, contributing to biological efficacy by scavenging free radicals and stabilizing blood glucose levels.

Acute lung injury (ALI) is a life-threatening condition with limited treatment options. The pathogenesis of ALI involves macrophage-mediated disruption and subsequent repair of the alveolar barriers, which ultimately results in lung damage and regeneration, highlighting the pivotal role of macrophage polarization in ALI. Although exosomes derived from mesenchymal stromal cells have been established as influential modulators of macrophage polarization, the specific role of exosomal microRNAs (miRNAs) remains underexplored. This study aimed to elucidate the role of specific exosomal miRNAs in driving macrophage polarization, thereby providing a reference for developing novel therapeutic interventions for ALI. We found that miR-7704 is the most abundant and efficacious miRNA for promoting the switch to the M2 phenotype in macrophages. Mechanistically, we determined that miR-7704 stimulates M2 polarization by inhibiting the MyD88/STAT1 signaling pathway. Notably, intra-tracheal delivery of miR-7704 alone in a lipopolysaccharide-induced murine ALI model significantly drove M2 polarization in lung macrophages and remarkably restored pulmonary function, thus increasing survival. Our findings highlight miR-7704 as a valuable tool for treating ALI by driving the beneficial M2 polarization of macrophages. Our findings pave the way for deeper exploration into the therapeutic potential of exosomal miRNAs in inflammatory lung diseases. © 2023 The Author(s)

Background: Although evidence indicates that extracorporeal shockwave therapy (ESWT) is effective in treating calcifying shoulder tendinitis, incomplete resorption and dissatisfactory results are still reported in many cases. Data mining techniques have been applied in health care in the past decade to predict outcomes of disease and treatment. Purpose: To identify the ideal data mining technique for the prediction of ESWT-induced shoulder calcification resorption and the most accurate algorithm for use in the clinical setting. Study Design: Case-control study. Methods: Patients with painful calcified shoulder tendinitis treated by ESWT were enrolled. Seven clinical factors related to shoulder calcification were adopted as the input attributes: sex, age, side affected, symptom duration, pretreatment Constant-Murley score, and calcification size and type. The 5 data mining techniques assessed were multilayer perceptron (neural network), naïve Bayes, sequential minimal optimization, logistic regression, and the J48 decision tree classifier. Results: A total of 248 patients with calcified shoulder tendinitis were enrolled in this study. Shorter symptom duration yielded the highest gain ratio (0.374), followed by smaller calcification size (0.336) and calcification type (0.253). With the J48 decision tree method, the accuracy of 3 input attributes was 89.5% by 10-fold cross-validation, indicating satisfactory accuracy. A treatment algorithm using the J48 decision tree indicated that a symptom duration of ≤10 months was the most positive indicator of calcification resorption, followed by a calcification size of ≤10.82 mm. Conclusion: The J48 decision tree method demonstrated the highest precision and accuracy in the prediction of shoulder calcification resorption by ESWT. A symptom duration of ≤10 months or calcification size of ≤10.82 mm represented the clinical scenarios most likely to show resorption after ESWT. © The Author(s) 2024.
