Fucoxanthin induces human melanoma cytotoxicity by thwarting the JAK2/STAT3/BCL-xL signaling axis

Melanoma is the most lethal skin malignancy. Fucoxanthin is a marine carotenoid with significant anticancer activities. Intriguingly, Fucoxanthin's impact on human melanoma remains elusive. Signal Transducer and Activator of Transcription 3 (STAT3) represents a promising target in cancer therapy due to its persistent activation in various cancers, including melanoma. Herein, we revealed that Fucoxanthin is cytotoxic to human melanoma cell lines A2758 and A375 while showing limited cytotoxicity to normal human melanocytes. Apoptosis is a primary reason for Fucoxanthin's melanoma cytotoxicity, as the pan-caspase inhibitor z-VAD-fmk drastically abrogated Fucoxanthin-elicited clonogenicity blockage. Besides, Fucoxanthin downregulated tyrosine 705-phosphorylated STAT3 (p-STAT3 (Y705)), either inherently present in melanoma cells or inducible by interleukin 6 (IL-6) stimulation. Notably, ectopic expression of STAT3-C, a dominant-active STAT3 mutant, abolished Fucoxanthin-elicited melanoma cell apoptosis and clonogenicity inhibition, supporting the pivotal role of STAT3 blockage in Fucoxanthin's melanoma cytotoxicity. Moreover, Fucoxanthin lowered BCL-xL levels by blocking STAT3 activation, while ectopic BCL-xL expression rescued melanoma cells from Fucoxanthin-induced killing. Lastly, Fucoxanthin was found to diminish the levels of JAK2 with dual phosphorylation at tyrosine residues 1007 and 1008 in melanoma cells, suggesting that Fucoxanthin impairs STAT3 signaling by blocking JAK2 activation. Collectively, we present the first evidence that Fucoxanthin is cytotoxic selectively against human melanoma cells while sparing normal melanocytes. Mechanistically, Fucoxanthin targets the JAK2/STAT3/BCL-xL antiapoptotic axis to provoke melanoma cell death. This discovery implicates the potential application of Fucoxanthin as a chemopreventive or therapeutic strategy for melanoma management. © 2024 Wiley Periodicals LLC.

We present a numerical study of the charge dynamical structure factor N(k, ω) of a one-dimensional (1D) ionic Hubbard model in the Mott insulator (MI) phase. We show that the low-energy spectrum of N(k, ω) is expressed in terms of the spin operators for the spin degrees of freedom. Numerical results for the spin degrees of freedom, obtained by the Lanczos diagonalization method, well reproduce the low-energy spectrum of N(k, ω) of the 1D ionic Hubbard model. In addition, we show that these spectral peaks probe the dispersion of the spin-singlet excitations of the system and are observed in the wide parameter region of the MI phase. His Primary Responsibilities are in the development of underwater nondestructive testing equipment for use by fleet divers and inspectors. He also performs research in the characterization of metal microstructure through ultrasonic scattering measurements. Mr. Mittleman received his BS from Cornell University in 1969 and science master's in ocean engineering from the Massachusetts Institute of Technology in 1970. His research currently supports doctoral studies with Iowa State University. Mr. Mittleman received the American Society of Naval Engineer's Solberg Award in 1981 for his contributions to underwater ship hull inspection. Mr. Mittleman is a member of ASNE ASNT ASTM IoD Sigma Xi Tau Beta Pi and Phi Kappa Phi. Lisa Swan:is a mechanical engineer at the Naval Surface Warfare Center in Panama City Florida. She is involved in nondestructive testing engineering primarily in the underwater arena. Ms. Swan holds a bachelor of science in materials engineering from North Carolina State University. She is a graduate of the Federal Women's Executive Leadership Program. Ms. Swan is a member of ASNT.

Significant progress has been made in making underwater ultrasonic thickness gauging and magnetic particle inspections available to the fleet. Under sponsorship from the Naval Sea Systems Command, Director of Ocean engineering, Underwater Ship Husbandry Division (NavSea 00C5), the Coastal Systems Station has developed complete hardware packages supporting these two nondestructive test methods, and has introduced them to military inspectors at a shore intermediate maintenance activity, a destroyer tender, and a naval shipyard. Performance trials conducted prior to taking the systems to the field have been accepted by NavSea, Ships' Concepts Group, materials Subgroup, Metals Division (NavSea 5142) as evidence that these inspections can reliably be performed underwater. Avenues for certifying specially trained divers and inspectors are being developed;for the first time the Navy will have all of the elements in place for underwater inspections satisfying the requirements of Mil-Std-271 (Requirements for Nondestructive Testing Methods) and the Naval Ships' Technical Manual Chapter 074. Underwater ultrasonic thickness gauging has also been slated for use in the fleet, as data from laboratory and field trials have consistently shown that reliable results can be obtained by a team comprising a certified topside inspector and a diver. In tests performed at the Ship Repair Facility (SRF), Yokosuka, underwater readings were compared to those taken in dry dock by SRF inspectors, and independently by contract inspectors. On the basis of approximately 800 locations, differences between the data sets were found to be randomly distributed, with a standard deviation on the order of 0.02''. This level of accuracy is largely sufficient to distinguish plate which will need replacement during overhaul, or plate which is thick enough to weld on.

Efforts to enhance the electrochemical properties of materials have become the focus of numerous studies because these properties are essential in various fields of application. Zeolitic imidazole framework-67 (ZIF-67) is a type of metal-organic framework (MOFs) expected to demonstrate excellent performance in electrochemical applications due to its numerous distinct properties. Consequently, various strategies and techniques have been developed to improve the electrochemical performance of ZIF-67. In this study, we employed bimetallic ZIF-67 constructed with cobalt (Co) and copper (Cu) metal ions within the imidazole frameworks. The use of bimetal is expected to increase conductivity and fine-tune the physicochemical properties of ZIF-67. Using coprecipitation methods, we synthesized both single-metal and bimetallic ZIF-67 and compared their characterizations. The addition of Cu metal ions does not alter the materials phase, ensuring compatibility with the single-metal ZIF-67 structure. However, the rhombic dodecahedron morphology of ZIF-67 shifts from a smooth to a concave and rough surface in Co/Cu ZIF-67. Furthermore, Co/Cu ZIF-67 exhibits higher peak current on their cyclic voltammetry (CV) curve by 46.15 μA. The results effectively illustrate the advantages of bimetal on ZIF-67 properties and performance. Finally, this study succesfully briefly demonstrate the potential development of Co/Cu-based ZIF-67 for various electrochemical applications. © Published under licence by IOP Publishing Ltd.

Composite materials are a combination of several materials to form a new material with specific properties. Chemical compatibility of the constituent materials is a factor that affects the characteristics of composite materials. Currently, composite phase change materials (PCM) are commonly applied as thermal energy storage systems. This study investigated the characteristics of beef tallow/rice husk charcoal/plaster PCM composite. The weight fraction of rice husk charcoal used was 10%. To prevent leakage of beef tallow from the composite structure, the researcher used polyvinyl acetate (PVAc) to cover the tallow/RHC composite before mixing it with plaster material. As much as 30% of the tallow/RHC/PVAc composite weight fraction is integrated into the plaster. PCM composite was made by using a direct incorporation method. The FTIR test results on the PCM composites produced showed good chemical compatibility among different components. The scanning electron microscope results showed the formation of microcapsules where the rice husk charcoal and polyvinyl acetate functions as a wrapper/capsule for beef tallow. This composite is suitable for application in building walls as thermal energy storage.

This study aimed to assesses the effect of various commercial Na2SiO3 on the compressive strength (CS) of alkaline activated fly ash mortar (AAFM). The three mixture of alkaline activated mortar (AAM) C1, C2 and C3 were prepared from the source material of fly ash and alkaline activator solution (AAS). The initial AAS was comprised of NaOH (10M) and various grade of Na2SiO3. The various grades of Na2SiO3 were characterized by their SiO2/Na2O molar ratio of 2.0, 2.2, and 3.3, respectively. The sample from each mixture was characterized based on the CS and microstructure changes using useful tools of XRD and FTIR analysis. The results obtained indicated that the highest CS achieved among the three mixtures were 48.23MPa of mixture C2 prepared with SiO2/Na2O molar ratio of 2.2. This was mainly due to higher binder formation (N-A-S-H gel type) and a higher rate of reaction of the main source material. This result is in line with XRD and FTIR analysis results finding.

Background: Wound healing is a complex and dynamic process involving the replacement of devitalized and missing cellular structures and tissue layers. Methods: This study involved developing a drug-loaded biomimetic micropillar wound dressing based upon inspiration from witnessing the adhesive ability in a gecko's footpad. The conformal adhesion was set up by applying a soft elastomer thin film on the developed wound dressing surfaces. The results showed that the developed surfaces prepared based on polydimethylsiloxane (PDMS) had low surface free energy, high chemical stability, and good plasticity. Astaxanthin (AST) and curcumin were applied as the patch's bioactive compounds to help accelerate the wound healing process. Results: The results showed that the developed surfaces prepared based on polydimethylsiloxane (PDMS) had low surface free energy, high chemical stability, and good plasticity. The results indicate that the developed biomimetic micropillar patches could release the loaded drugs and significantly increase cell viability and collagen content to 20 % ∼ 25 % in vitro and in vivo. Conclusion: In summation, the primary purpose of this study has been to design a novel encapsulated carrier (micropillar patch) for use in adjustable drug release, while combining with the biomimetic microstructure to reach wound healing through percutaneous absorption. © 2024 Taiwan Institute of Chemical Engineers

Objectives: The 12-item Self-Compassion Scale–Short Form (SCS–SF) is a widely used instrument for the assessment of self-compassion. To date, there have been few examinations of this instrument’s psychometric properties, particularly across nations and languages. Therefore, we used data from the Body Image in Nature Survey (BINS) to assess measurement invariance of the SCS–SF across nations, languages, gender identities, and age groups. Methods: Participants (N = 56,968) from 65 nations completed the SCS–SF in 40 languages. Using these data, we tested various hypothesised models of the SCS–SF in the total sample and, using multi-group confirmatory factor analysis, tested for invariance of the optimal model across national groups, languages, gender identities, and age groups. Results: In the total dataset, we found that an 11-item, 2-factor model (i.e., SCS-11) provided best fit to the data, with the two factors tapping distinct constructs of compassionate and uncompassionate self-responding. The SCS-11 was found to be partially scalar invariant across national groups and languages, and fully scalar invariant across gender identities and age groups. There was wide variation in latent means for the two factors, particularly across national groups and languages. Further analyses showed negligible associations between the two factors and sociodemographic variables, including marital status, financial security, and urbanicity. Conclusions: Our results suggest that it may be possible to derive a stable 2-factor model of the SCS–SF for use in cross-cultural research, but also highlight the likelihood of cross-national and cross-linguistic variations in the way that self-compassion is understood. © The Author(s) 2025.

Recently, there has been a growing interest in artificial skin on hydrogels as a potential alternative for animal testing to evaluate the efficacy and safety of new drugs or cosmetics. Synthetic polymers have been widely used due to their superior physicochemical properties. However, synthetic polymers such as polyvinyl alcohol (PVA) are limited by their poor biocompatibility and cell adhesion. Combining synthetic and natural polymers can improve these properties. Therefore, in this study, we aimed to investigate the effect of alginate and chitosan addition to PVA-based hydrogels on protein adsorption, cell viability and cell attachment. The addition of chitosan decreased protein adsorption, which was caused by the changes in electrostatic charge by PVA-chitosan interaction and changes in surface roughness. Primary human fibroblast cells cultured on PVA, PVA-alginate and PVA-chitosan had >80% viability which indicates the cytocompatibility of the hydrogels. More importantly, alginate and chitosan also improved the cell attachment on the PVA-based hydrogels. Taken together, the addition of both alginate and chitosan has the potential to improve PVA-based hydrogels for artificial skin application.
