Multiplexed Nanoscopy via Buffer Exchange

Understanding cellular functions, particularly in their intricate complexity, can greatly benefit from the spatial mapping of diverse molecules through multitarget single-molecule localization microscopy (SMLM). Existing methodologies, primarily restricting the encoding dimensions to color and lifetime or requiring cyclic staining, often involve broad chromatic detection, specialized optical configurations, or sophisticated labeling techniques. Here, we propose a simple approach called buffer-exchange stochastic optical reconstruction microscopy (beSTORM), which introduces an additional dimension to differentiate between single molecules irrespective of their spectral properties. This method leverages the distinguishable photoblinking responses to distinct buffer conditions, offering a straightforward yet effective means of fluorophore discrimination. Through buffer exchanges, beSTORM achieves multitarget SMLM imaging with minimal crosstalk. Direct integration with expansion microscopy (ExM) demonstrates its capability to resolve up to six proteins at the molecular level within a single emission color without chromatic aberration. Overall, beSTORM presents a highly compatible imaging platform, promising significant advancements in highly multiplexed nanoscopy for exploring multiple targets in biological systems with nanoscale precision. © 2024 The Authors. The Vision Transformer (ViT) model serves as a powerful model to capture and comprehend global information, particularly when trained on extensive datasets. Conversely, the Convolutional Neural Network (CNN) model is beneficial to training with small datasets for retaining essential local information. Inspired by these properties of ViT and CNN models, we introduce a hybrid framework that smoothly increases the cross-domain generalization in Semi-supervised Domain Adaptation (SSDA). To achieve this goal, we first train the ViT model on abundant labeled source data, while the CNN model is trained on a few labeled target samples. Then, these models dynamically exchange their knowledge for potential generalization to unlabeled target data via the proposed method, named Dual Dynamic Consistency Regularization (D2CR). Specifically, the ViT model provides its pseudo labels to update the global perspective for the CNN model. Similarly, the CNN model offers pseudo labels to complement the local perspective for the ViT model. The previous methods use a fixed threshold algorithm for the pseudo-labeling process. However, we utilize the dynamic threshold strategy to create pseudo labels for the bi-directional consistency regularization learning between the ViT and CNN models. We verify our approach across several SSDA benchmark datasets. The outstanding experimental results provide strong evidence of the effectiveness and superiority of our approach over previous state-of-the-art SSDA methods. © 2013 IEEE.

Indonesia will benefit from a demographic boom in 2030 with a higher labor supply than in earlier decades. Then in industrial revolution 4.0 robotics and artificial intelligence will take the place of low-skilled or menial employment that don't require specialized expertise (AI). To aim research is Telematics Work Field Review Text Classification Using the Naïve Bayes Method. The method using Multinomial Naïve Bayes model which is trained to learn from patterns in training data set without being programmed explicitly. Then, based on the Term Frequency - Inverse Document Frequency, consider the weighting of the word used (TF-IDF). The text classification stage is then carried out using the multinominal nave bayes classification method with evaluation using the confusion matrix, following the acquisition of the TF-IDF value. In the study it took data with web crawling techniques on social media sites twitter. The data collected was 936 data consisting of 7,8% negative sentiments, 26,4% positive sentiments, and 65,8% neutrals. The results of accuracy testing using the Confusion Matrix. And from the results of such tests resulted in an accuracy of 66%, precision 73%, and recall 85%. © 2022 IEEE.

Implantable electrodes have raised great interest over the last years with the increasing incidence of neurodegenerative disorders. For brain implant devices, some key factors resulting in the formation of glial scars, such as mechanical mismatch and acute injury-induced inflammation, should be considered for material design. Therefore, in this study, a new biocompatible flexible electrode (e-SgG) with arbitrary shapes on a positive electrode was developed via electrogelation by applying a direct electrical voltage on a silk fibroin/gelatin/reduced graphene oxide composite hydrogel. The implantable flexible e-SgG-2 film with 1.23% rGO content showed high Young's modulus (11-150 MPa), which was sufficient for penetration under dried conditions but subsequently became a biomimetic brain tissue with low Young's modulus (50-3200 kPa) after insertion in the brain. At the same time, an anti-inflammatory drug (DEX) incorporated into the e-SgG-2 film can be electrically stimulated to exhibit two-stage release to overcome tissue inflammation during cyclic voltammetry via degradation by applying an AC field. The results of cell response to the SF/gelatin/rGO/DEX composite film showed that the released DEX could interrupt astrocyte growth to reduce the inflammatory response but showed non-toxicity toward neurons, which demonstrated a great potential for the application of the biocompatible and degradable e-SgG-D electrodes in the improvement of nerve tissue repair. © 2024 The Royal Society of Chemistry

This research article presents a modified isolated SEPIC converter suitable for high-power applications. It uses a transformer instead of coupled inductor for isolation between the input power source and the output load. The steady-state analysis is explained in detail. Thermal loss analysis is performed by using PLECS software. Further, a hardware prototype is developed to verify the performance aspects of the proposed converter. © 2023 IEEE.

Our surroundings' auditory landscapes are a wealth of knowledge, providing insights into a range of outdoor pursuits. The automatic classification of these actions using audio event detection and classification (AEDC) systems has a great deal of potential applications in environmental monitoring, security, surveillance, and driverless cars. Even though AEDC has advanced significantly in indoor settings, difficulties still exist outside because of the variable and changing acoustic conditions brought on by elements like the weather, different sound sources, and ambient noise from traffic and industry. This study suggests an outdoor audio event categorization model based on convolutional neural networks (CNNs). The suggested model shows passable accuracy by utilizing adaptation to the downstream task using the ESC-50 dataset and transfer learning from a pre-trained model. The effectiveness of multi-class audio classification models in downstream tasks is analyzed in this paper, with an emphasis on the effect of an increasing number of output classes on accuracy. Three models - three, four, and five classes - with different output class configurations are used in the study, and their training and validation accuracies are assessed. Although the accuracy scores above 80% are commendable, the data show a discernible reduction in performance as the number of classes rises. Notably, the three-class model attains a validation accuracy exceeding 90%, whereas the four-class and five-class models exhibit reduced accuracies, falling below 90% and 85%, respectively. © 2023 IEEE.

This paper proposes a reconfiguration for a single-phase, double conversion uninterruptible power supply (UPS). With the addition of two static switches to the original circuit, the UPS mitigates the ripple in the current, through battery discharge performed by two legs. Other than these switches, the circuit topology is preserved. The preliminary results show a satisfactory level of ripples for the inductor current and suitable power quality for the output voltage. © 2023 IEEE.

Learning to assemble geometric shapes into a larger target structure is a pivotal task in various practical applications. In this work, we tackle this problem by establishing local correspondences between point clouds of part shapes in both coarse- and fine-levels. To this end, we introduce Proxy Match Transform (PMT), an approximate high-order feature transform layer that enables reliable matching between mating surfaces of parts while incurring low costs in memory and compute. Building upon PMT, we introduce a new framework, dubbed Proxy Match TransformeR (PMTR), for the geometric assembly task. We evaluate the proposed PMTR on the large-scale 3D geometric shape assembly benchmark dataset of Breaking Bad and demonstrate its superior performance and efficiency compared to state-of-the-art methods. Due to the disparity in the levels of difficulty presented by the several tasks, doing domain adaptation in an adversarial way may result in an imbalanced learning process. In the MNIST dataset, this phenomenon also manifests itself in the form of domain adaptation for color-shifted distribution. In this particular situation, the domain classifier has a higher tendency to fit more quickly, but the category classifier fits quite poorly in the learning process. In order to address this problem, a new hyper-parameter has been added to the loss function in order to strike a compromise between the learning speed of the domain and the categorical classifier. By using this technique, the categorical classifier may better match the data while still maintaining the same level of performance as the domain classifier. In order to determine whether or not making use of this hyper-parameter is useful, the phenomena in question is examined using three distinct color-shifted settings. Following the evaluations, it was discovered that the newly introduced hyper-parameter is capable of coping with imbalanced learning while simultaneously engaging in domain adaptation. © 2023 IEEE.

Effect of electrolyte anions on the cycle life of a polymer electrode in aqueous batteries

Redox polymers are a class of high-capacity, low-cost electrode materials for electrochemical energy storage, butthe mechanisms governing their cycling stability are not well understood. Here we investigate the effect of anionson the longevity of a p-dopable polymer through comparing two aqueous zinc-based electrolytes. Galvanostaticcycling studies reveal the polymer has better capacity retention in the presence of triflate anions than that withsulfate anions. Based on electrode microstructural analysis and evolution profiles of the cell stacking pressure, theorigin of capacity decay is ascribed to mechanical fractures induced by volume change of the polymer activematerials during repeated cycling. The volume change of the polymer with the triflate anion is 61% less than thatwith the sulfate anion, resulting in fewer cracks in the electrodes. The difference is related to the different anionsolvation structures—the triflate anion has fewer solvated water molecules compared with the sulfate anion,leading to smaller volume expansion. This work highlights that anions with low solvation degree are preferablefor long-term cycling.
