We developed an optical tweezers directed approach that allows synthesis of different oligonucleotide on an array of controlled pore glass (CPG) beads in parallel in a two-stream laminar flow microreactor and obtained one oligo sequence per bead. This reactor has been shown to effectively facilitate CPG bead loading and confinement, maintain liquid/ liquid (inert liquid/reagent) interface and withstand the reagents and solvents present in the modified phosphoramidite chemistry. The optical tweezers allows us to externally manipulate the CPG beads inside the channel without having to modify the microfluidic device itself. The parallel synthesis of several different oligonucleotides in a single synthetic run was demonstrated.

Micro Aerial Vehicles (MAVs) and larger Unmanned Aerial Vehicles (UAVs) constitute a powerful tool for the modern warfighter and first responder. However, every developing technology must first be tested before being exported to the battlefield or hot spot to assure predictable behaviors. In today's digital age, computers give designers and testers unprecedented ability to quickly and easily test virtual models before physical prototypes are created and field tested. We have utilized emerging Free Open Source Software (FOSS) simulators and workbenches (e.g. FlightGear and AUV Workbench) to improve mission-driven MAV/UAV video and imagery capture and analysis capabilities. In this project, use of FlightGear to analyze flight trajectories and video capture capabilities of different UAVs is presented. FlightGear allows the free implementation of changeable flight dynamic models, weather, autopilot, GPS, and multiplayer to be included in the analysis of simulated flights. Another attractive aspect of FlightGear is its extensible and easy-to-modify implementation. Modifications are accomplished by changing or adding human readable XML files in the FlightGear subdirectories. In this effort, existing aerial vehicle models are incorporated into FlightGear to create flight trajectories and analyze video capture capabilities to determine the necessary toolset for analyzing imagery data to meet the mission needs. The limitations of the existing camera systems as well as effects of various flight parameters on mosaicking and georegistration are presented here. Sample analysis of the effect of aerial vehicle speed on the overlap of frames captured at the typical 30 frames/second frame rate, which provides a base for comparison of flight attributes, are demonstrated together with other parameters that affect mission performance. © 2010 ASCE.

This research proposes a method to eliminate friction effect and reduced tracking error on CNC milling machines. A CNC milling machine has many connected mechanical components which have friction effects such as ball screws and rails. To control servo motors which are used to drive X-Y table in CNC milling machine, the conventional PID controller is widely used. In this research, Panasonic motor driver is set to the torque control mode. To compensate the friction effects, the friction feedforward compensation method is proposed. Moreover, to reduce the position tracking error, position feedforward controller is used. The simulation and implementation of position control in one axis of PID with Friction Feedforward Compensation and Position Feedforward Controller (FFC-PFC) are presented.

Devices for continuous glucose monitoring (CGM) are currently a major focus of research in the area of diabetes management. It is envisioned that such devices will have the ability to alert a diabetes patient (or the parent or medical care giver of a diabetes patient) of impending hypoglycemic/ hyperglycemic events and thereby enable the patient to avoid extreme hypoglycemic/hyperglycemic excursions as well as minimize deviations outside the normal glucose range, thus preventing both life-threatening events and the debilitating complications associated with diabetes. It is anticipated that CGM devices will utilize constant feedback of analytical information from a glucose sensor to activate an insulin delivery pump, thereby ultimately realizing the concept of an artificial pancreas. Depending on whether the CGM device penetrates/breaks the skin and/or the sample is measured extracorporeally, these devices can be categorized as totally invasive, minimally invasive, and noninvasive. In addition, CGM devices are further classified according to the transduction mechanisms used for glucose sensing (i.e., electrochemical, optical, and piezoelectric). However, at present, most of these technologies are plagued by a variety of issues that affect their accuracy and long-term performance. This article presents a critical comparison of existing CGM technologies, highlighting critical issues of device accuracy, foreign body response, calibration, and miniaturization. An outlook on future developments with an emphasis on long-term reliability and performance is also presented. © Diabetes technology Society.

A multi-source micro power generator employing thermal and vibration energy harvesting

This paper introduces a new cantilever type multi-source energy harvester generating electric power from both ambient heat and vibration. Harvesting energy from vibration was realized by electromagnetic conversion, whereas the energy generation from heat was supplied by making use of Seebeck effect of Cr–Al thermocouples implemented on the microcantilevers. The measured average Seebeck coefficient is 12 μV/K per thermocouple. A total voltage of 3.3 mV was generated from the thermoelectric part and 13.4 mV from the electromagnetic part of the device. Measured total power from the fabricated chip is 1.91 nW (1.12 nW from vibration, 0.79 nW from thermoelectric).

There exists a need for additional transmission capacity to connect distributed resources such as wind energy from remote locations to the power grid. The high cost of running transmission lines necessitates having a way to minimize the length of transmission required to connect the distributed resources to load centers. In addition, the substations that connect to the transmission lines need to be fault tolerant by ensuring that the loss of any one transmission line does not result in the loss of power to any substation. In this paper, the relative cost of connecting a range of sources to a range of loads with and without fault tolerance is presented. Fault tolerance is achieved through a 2-path redundant graph approach. Studies show that the additional cost of ensuring fault tolerance for selected schemes ranges between 8% and 33%. This percentage value can support decision making in practice.

In this paper, we investigate a dual-hop opportunistic decode-and-forward relaying scheme where the source may or not be able to communicate directly with the destination. We first derive statistics based on exact probability density function (PDF) of each hop. Then, the PDFs are used to determine closed-form outage probability expression for a transmission rate R. Furthermore, we evaluate the asymptotic outage performance and the diversity order is deduced. Unlike existing works where the analysis focused on high signal-to-noise ratio (SNR) regime, such results are important to enable the designers to take decisions regarding practical systems that operate at low SNR regime. We show that performance simulation results coincide with our analytical results under practical assumption of unbalanced hops.

We demonstrate GaInN multiple quantum well (MQW) light-emitting diodes (LEDs) having ternary GaInN quantum barriers (QBs) instead of conventional binary GaN QBs for a reduced polarization mismatch between QWs and QBs and an additional separate confinement of carriers to the MQW active region. In comparison with GaInN LEDs with conventional GaN QBs, the GaInN/GaInN LEDs show a reduced blueshift of the peak wavelength with increasing injection current and a reduced forward voltage. In addition, we investigate the density of pits emerging on top of the MQW layer that are correlated with V-defects and act as a path for the reverse leakage current. The GaInN/GaInN MQW structure has a lower pit density than the GaInN/GaN MQW structure as well as a lower reverse leakage current. Finally, the GaInN/GaInN MQW LEDs show higher light output power and external quantum efficiency at high injection currents compared to the conventional GaInN/GaN MQW LEDs. We attribute these results to the reduced polarization mismatch and the reduced lattice mismatch in the GaInN/GaInN MQW active region. © 2010 American Institute of Physics.

Short-term load forecasting has received a lot of attention from both researchers and practitioners. Many techniques, such as neural networks, fuzzy logic, and time series models, are developed to improve the modeling and forecasting process with varying success. Some research has also been devoted to improving the weather forecast or relieving the impact of uncertainties in the weather forecast. Nowadays, the quality of load and weather data history becomes a bottleneck of enhancing the forecast accuracy. With the emerging Smart Grid initiatives, the utilities have the opportunities to upgrade their infrastructure, which includes the advanced metering systems. These upgrades will potentially lead to a high quality load history in the near future. On the other hand, quality of weather history has been a concern of the small and medium sized utilities. This paper presents a recent study for a medium sized utility in eastern US. Multiple linear regression, which is currently deployed in this utility, is used to generate the base forecast in this paper. A Monte Carlo based methodology is proposed to quantify the cost of the data uncertainties in the temperature history. Finally, a cost-benefit analysis is performed to help the utilities decide how many weather stations should be installed. ©2010 IEEE.

The ability to think critically is vital to success in engineering and technology practice. Employers in these fields, however, consistently identify critical thinking as one of the skills that is not sufficiently developed in new college graduates, and call upon engineering and technology educators to address this obvious need. Unfortunately, critical thinking is a developmental skill that cannot be taught simply by the usual methods - step-by-step instruction followed by repetitive drills - used for other technical skills. Critical thinking must instead be nurtured through practical experience solving problems with appropriate guidance and reinforcement. One very effective context for developing such skills is in open-ended assignments with no single "right" answer, to which students must apply not only their technical knowledge, but also an element of critical judgment, to determine which approach among many possible will yield the most reasonable and applicable results. For educators, a key component of nurturing critical thinking is learning to recognize and reinforce it when it happens, or nudge students toward such behaviors when it is not happening but should be. Toward that end, we have developed a rubric to assess critical thinking during several phases (initial design or set-up;testing of method;evaluation of results) of open-ended assignments in engineering and technology. The rubric is designed to be generally applicable to open-ended assignments at every level (freshman through senior) in engineering and technology, allowing users to track the development of critical thinking skills as students progress through the curriculum. We present the rubric and preliminary results from applying it to two different open-ended assignments. © American Society for engineering Education, 2009.
