Finding or discovering data is a necessary precursor to being able to reuse data, although relatively little large-scale empirical evidence exists about how researchers discover, make sense of and (re)use data for research. This study presents evidence from the largest known survey investigating how researchers discover and use data that they do not create themselves. We examine the data needs and discovery strategies of respondents, propose a typology for data (re)use and probe the role of social interactions and other research practices in data discovery, with the aim of informing the design of community-centric solutions and policies. Clinical prediction models (CPMs) have become fundamental for risk stratification across healthcare. The CPM pipeline (development, validation, deployment, and impact assessment) is commonly viewed as a one-time activity, with model updating rarely considered and done in a somewhat ad hoc manner. This fails to address the fact that the performance of a CPM worsens over time as natural changes in populations and care pathways occur. CPMs need constant surveillance to maintain adequate predictive performance. Rather than reactively updating a developed CPM once evidence of deteriorated performance accumulates, it is possible to proactively adapt CPMs whenever new data becomes available. Approaches for validation then need to be changed accordingly, making validation a continuous rather than a discrete effort. As such, "living" (dynamic) CPMs represent a paradigm shift, where the analytical methods dynamically generate updated versions of a model through time; one then needs to validate the system rather than each subsequent model revision.

The Rubin Observatory’s 10-year Legacy Survey of Space and Time will observe near to 20 billion galaxies. For each galaxy the properties can be inferred. Approximately 105 galaxies observed per year will contain Type Ia supernovae (SNe), allowing SN host-galaxy properties to be calculated on a large scale. Measuring the properties of SN host-galaxies serves two main purposes. The first is that there are known correlations between host-galaxy type and supernova type, which can be used to aid in the classification of SNe. Secondly, Type Ia SNe exhibit correlations between host-galaxy properties and the peak luminosities of the SNe, which has implications for their use as standardisable candles in cosmology. We have used simulations to quantify the improvement in host-galaxy stellar mass (M∗) measurements when supplementing photometry from Rubin with spectroscopy from the 4-metre Multi-Object Spectroscopic Telescope (4MOST) instrument. We provide results in the form of expected uncertainties in M∗ for galaxies with 0.1 AB ∗ by more than a factor of 2 compared with Rubin data alone. This applies for elliptical and Sc type hosts. We demonstrate that the reduced uncertainties in M∗ lead to an improvement of 7% in the precision of the "mass step" correction. We expect our improved measurements of host-galaxy properties to aid in the photometric classification of SNe observed by Rubin. © 2023, CC BY.

Back pain is a common and debilitating disorder with largely unknown underlying biology. Here we report a genome-wide association study of back pain using diagnoses assigned in clinical practice; dorsalgia (119,100 cases, 909,847 controls) and intervertebral disc disorder (IDD) (58,854 cases, 922,958 controls). We identify 41 variants at 33 loci. The most significant association (ORIDD = 0.92, P = 1.6 × 10−39; ORdorsalgia = 0.92, P = 7.2 × 10−15) is with a 3’UTR variant (rs1871452-T) in CHST3, encoding a sulfotransferase enzyme expressed in intervertebral discs. The largest effects on IDD are conferred by rare (MAF = 0.07 − 0.32%) loss-of-function (LoF) variants in SLC13A1, encoding a sodium-sulfate co-transporter (LoF burden OR = 1.44, P = 3.1 × 10−11); variants that also associate with reduced serum sulfate. Genes implicated by this study are involved in cartilage and bone biology, as well as neurological and inflammatory *** is known about the biology of back pain, a leading cause of disability. Here the authors report 30 new back pain loci, implicating genes involved in cartilage/bone biology, as well as neurological and inflammatory processes.

Neuroprotective effect of Spilanthes acmella Murr. on pesticide-induced neuronal cells death

Objective: To investigate protective effects of Spilanthes acmella(S,acmella) Murr,extracts against pesticide-induced neuronal cells death and to elucidate the underlying molecular mechanism in dopaminergic(SH-SY5Y) cells lines,Methods: Cell viability of SH-SY5 Y cells was studied by treating the cells with various concentration of pirimicarb for 24 hr,Neuroprotective effect of S,acmella Murr,extracts was investigated by adding the plant extracts to the medium for 24 hr prior to the incubation with 100 μM H_2O_2 or with pirimicarb for 24 hr,Control-untreated cells were incubated with the culture medium,Cell viability was measured by MTT assay,calpain and calpastatin expressions were analyzed by Western blotting and immunocytochemistry,Results: Pretreatment of SH-SY5 Y cells with S,acmella Murr,extracts(1 μg/m L) for 24 hr significantly increased the dopaminergic neurons in pirimicarb-induced neurotoxicity,In addition,pretreatment with the S,acmella Murr,extracts led to decreased calpain but increased calpastatin protein levels,Conclusion: S,acmella Murr,extracts exerted neuroprotective effect,via an alteration of calcium homeostasis,against pirimicarb induced neurotoxicity,The S,acmella Murr,might be a potential natural candidate with neuroprotective activity.
