Natural waterways near urban areas are heavily impacted by anthropogenic activities, including their microbial communities. A contaminant of growing public health concern in rivers is antibiotic resistant genes (ARGs), which can spread between neighboring bacteria and increase the potential for transmission of AR bacteria to animals and humans. In order to identify the matrices of most concern for AR, we compared ARG burdens and microbial community structures between sample types from the Scioto River Watershed, Ohio, United States from 2017-2018. Five environmental matrices (water, sediment, periphyton, detritus, and fish gut) were collected from 26 river sites. Due to our focus on clinically relevant ARGs, three carbapenem resistance genes (blaKPC, blaNDM, and blaOXA-48) were quantified via DropletDigitalTM PCR. At a subset of nine urbanized sites, we conducted16S rRNA gene sequencing and functional gene predictions. Carbapenem resistance genes were quantified from all matrices, with blaKPC being the most detected (88% of samples), followed by blaNDM (64%), and blaOXA-48 (23%). Fish gut samples showed higher concentrations of blaKPC and blaNDM than any other matrix, indicating potential bioaccumulation of ARGs, and risk of broader dissemination through aquatic and nearshore food webs. Periphyton had higher concentrations of blaNDM than water, sediment, or detritus. Microbial community analysis identified differences by sample type in community diversity and structure. Sediment samples had the most diverse microbial communities, and detritus, the least. Spearman correlations did not reveal significant relationships between the concentrations of the monitored ARGs and microbial community diversity. However, several differentially abundant taxa and microbial functions were identified by sample type that are definitive of these matrices’ roles in the river ecosystem and habitat type. Cordyceps militaris (C. militaris) is a unique medicinal fungus of the genus Cordyceps. It has high economic value due to various biological functions from the extracellular polysaccharides (EPSs) it produces. The aim of this study was to establish a mycelium-immobilization system for EPS production in a bubble column system. First, EPS production was optimized by 90.2% (from 0.72 to 1.37 g/L) using response surface methodology under an alginate-immobilized mycelium system with 6 days of cultivation. The highest EPS production was obtained (5.3 g/L) in a flask system through elongation to 15 days of cultivation. EPS production subsequently further increased to 10.42 g/L utilizing a bubble column reactor. Furthermore, the system can be reused for at least three rounds. In the EPS characteristics analysis, the produced EPSs were mainly composed of glucose and galactose. They presented strong antibacterial activity against Staphylococcus aureus. In terms of antioxidant activity, respective the EPS concentration that caused 50% scavenging of DPPH and ABTS free radicals values (IC50) were 4.65 and 7.92 mg/mL. In summary, these findings should be helpful for establishing a suitable industrial fermentation system for C. militaris and can be studied in the food and medical industries in future work. © 2024 The Authors

In Brazil, the use of Eucalyptus is focused on the production of wood or pulp for the paper industry but without any general recovery of waste, with leaves and branches being left on the ground. One possibility is to use these residues as raw materials in the production of industrially relevant and value-added compounds such as essential oil. The aim of the present study was to investigate the chemical composition, yield, anti-inflammatory/antinociceptive activities, and acute toxicity in mice, as well as the antimicrobial effects of essential oils from the leaves of 7 varieties of Eucalyptus and hybrids against Escherichia coli , Staphylococcus aureus, and Candida albicans . The extraction of oils was carried out using hydrodistillation, and they were analyzed by gas chromatography coupled to mass spectrometry. Urocam and Grancam were the plants that obtained the highest oil yield, with yields of 3.32 and 2.30%, respectively. The main chemical components identified in these plants were 1.8 cineole and α -pinene. The antinociceptive effect of the 7 oils (50 mg/kg, p.o.) was initially assessed in the acetic acid-induced writhing test. In this assay, a significant ( p < 0.05) antinociceptive/anti-inflammatory effect was observed from 4 tested essential oils ( E. benthamii, E. saligna, and the hybrids Urocam and Grancam) when compared to the vehicle-treated group. This effect was then confirmed in the formalin-induced paw licking test. No toxicological effects or alterations were observed in motor coordination after the administration of the studied oils to the animals. In the antimicrobial evaluation, the seven essential oils inhibited the growth of S. aureus, E. coli , and C. albicans at different concentrations. Collectively, these results demonstrate that the essential oil from the leaves and branches of Eucalyptus species and varieties present potential biomedical applications and represent a source of antimicrobial and/or anti-inflammatory compounds.

Nowadays, worldwide packaging demand evaluates materials containing active compounds to increase the food shelf-life. Thereby, extracted buriti oil by pressurized propane was incorporated as an antioxidant and antimicrobial additive to prepare active gelatin films to preserve the Brazilian traditional and artisanal fresh coalho cheese. The oil GC/qMS analysis revealed unsaturated fatty acids, polyphenols, and carotenoids high content. Interactions oil-gelatin polymeric structure in films were analyzed by mechanical, physical-chemical and functional properties. Despite no significant differences in physical properties between oil-based and oil-free gelatin films, oil incorporation decreased the water vapor permeability. Concerning antioxidant and antibacterial effects, oil-incorporated gelatin-based films displayed bacteriostatic properties and pH responsivity during cheese storage compared to control groups. This trial revealed a significant decrease of Salmonella sp., P. aeruginosa, and E. coli, evaluated over 14-day storage at 5°C. Therefore, these films minimized microbial spoilage during cheese storage, indicating promising application for food-active packaging. © 2023, The Authors. All rights reserved.

The area of the rice harvest in Banten Province in 2021 was 319,558 ha with a production of 1,63 million tons of unhusked rice or with a productivity of 5.1 tons/ha (CBS,2022). The objectives of this study were: 1) to optimize the target of rice and corn production in Banten Province, and 2) to optimize the target farmers' income in Banten Province. The study method used was a survey, with purposive sampling at the farmer level. Data were analyzed qualitatively (uses descriptive tabulation) and quantitatively (uses goal programming). The results of this study were:1) The target of rice production in the Banten Province of 2.1 million tons of unhusked rice can be achieved with the optimal area of irrigated paddy of 299,755.7 ha and the optimal area of field/dry land of 87,750.9 ha. The target for maize production of 1.75 million tonnes in Banten Province can be achieved with an optimal planting area of 342.211.7 ha of dry land. 2) The optimal area of food crops to achieve the farmer's income target in Banten Province in 2018 is IDR. 8.79 trillion comes from the planting area of lowland rice covering an area of 391,222.1 ha, the corn planting area of 169,059.9 ha, soybean planting area of 23,074.82 ha and cassava planting area of 2,832.77 ha. © 2023, The Authors. The food packaging market has been seeking to meet consumer health and personal standards. In this sense, plant-based ingredients and food such as vegetable extracts are the future. Among the trends, active packaging stands out since it can extend the shelf-life of packaged food or improve its sensory properties and food safety. In this work, a watchful literature review was conducted to identify studies on food packaging coatings and vegetable extracts coating and their benefits. The innovations and technological advances in the food area have collaborated in several aspects, allowing the improvement of food quality, the protection of bioactive compounds from adverse effects of harsh environment, and the replacement of conventional plastics. The use of vegetable extracts and nanocomposites in edible coatings and films has shown positive results in different types of food and is a promising material for being considered a safe compound and a sustainable alternative. © 2022

Cassava is a staple crop that plays a significant role in the food security of many countries. However, its processing produces a liquid by-product known as cassava wastewater (CW), which can have adverse environmental consequences if discarded without treatment. Despite its cyanide content, CW has a high organic content and may be profitable when used to produce biogas. In this study, the influence of calcium particles from eggshell residues was investigated on the anaerobic digestion of CW. Moreover, the performance of the bioreactor was remotely monitored. Calcium particles from milled-calcined chicken eggshells were added to the bioreactor, and biogas production was investigated for 21 days. Adding 1 g/L and 3 g/L of calcium particles increased biogas (Bio H 2 + Bio CH 4 ) production by 195% and 338%, respectively. Finally, the requirement for digestate post-treatment before use in agriculture was observed after assessing its phytotoxicity through the germination and root growth of L. sativa seeds. © 2022, The Authors. Industrial adoption of microalgae biofuel technology has always been hindered by its economic viability. To increase the feasibility of bioethanol production from microalgae, fermentation was applied to Chlorella vulgaris FSP-E biomass at high-solid-loading conditions. First, Chlorella vulgaris FSP-E was cultivated to produce microalgae biomass with high carbohydrate content. Next, different ethanol-producing microorganisms were screened. Saccharomyces cerevisiae FAY-1 showed no inhibition when fermenting high initial glucose concentrations and was selected for the fermentation experiments at high solids loading. Optimization of acid hydrolysis at high biomass loading was also performed. The fermentation of microalgal biomass hydrolysate produced a final ethanol concentration and yield higher than most reported literature using microalgae feedstock. In addition, the kinetics of bioethanol fermentation of microalgae hydrolysate under high solid loading were evaluated. These results show the potential of fermenting microalgae biomass at high solids loading in improving the viability of microalgae bioethanol production. © 2022, The Authors. All rights reserved.

Cattle farming is considered a major contributor to antibiotic resistant (AR) genes and bacteria in nearby environments, predominantly due to the large amount of manure released by cattle to soils. This study sought to compare soil within and outside of cattle pens from seven farms in the US. Soil microbial community differences are described, as well as differences in AR gene and enteric pathogen concentrations in the soils. Sources of fecal loading are quantified via microbial source tracking (MST) methods. The family Peptostreptococcaceae was more abundant in pen soils, and Rhodanobacteraceae was more abundant in non-pen soils. There were no differences in microbial community diversity between soils within and outside of cattle pens. The MST result showed that ruminant-specific fecal contamination (Rum2Bac) was abundant both in pen and non-pen soils, likely indicating heavy cattle fecal loading. However, the soil within cattle pens had higher levels of Campylobacter, Salmonella, and AR genes for resistance to sulfonamides (sul1) and carbapenems (KPC) than the soil outside of pens (p<0.05). This is the first study identifying KPC in livestock-associated soils in the US. This study supports increased hazard associated with livestock soils for animal and human health, as well as environmental quality. © 2021
