ABSTRACTIntroductionMore than 70% of sporadic colorectal carcinomas (CRC) harbour loss-of-function mutations in the APC gene, which cause aberrant activation of Wnt signalling and ultimately lead to the onset of colon adenomas. The Apc min/+ mouse model is an appropriate tool for the study of the formation of early adenomas, which progress into carcinomas when mice are crossed with other oncogenic models, establishing a direct link between Wnt signalling and colorectal cancer. The EMT-TF ZEB1 is induced by Wnt pathway and, at the same time, acts as a mediator of the pathway in the regulation of key Wnt target genes. However, the role of ZEB1 during the formation of early intestinal adenomas remains unknown. Material and methodsThe study has made use of a wide range of in vitro, in vivo and high throughput approaches, including a mouse model of intestinal adenomas, a mouse deficient for ZEB1 and RNA sequencing. Results and discussionsTo explore the role of ZEB1 in adenoma formation, we downregulated its expression in the Apc min/+ mouse by crossing it with Zeb1 +/- mice (Apc min/+/Zeb1 +/-) and compared the formation of adenomas with respect Apc min/+/Zeb1 +/+. Compared to Apc min/+/Zeb1 +/+ counterparts, Apc min/+/Zeb1 +/- mice presented half the number of tumours and a more differentiated histological pattern. Likewise, tumours in Apc min/+/Zeb1 +/- showed no significant difference in proliferation—as assessed by PCNA staining—but increased number of senescent and apoptotic cells—determined by β-galactosidase and TUNEL assays—than the tumours developed in Apc min/+/Zeb1 +/+ mice. In order to ascertain the mechanisms by which ZEB1 regulates adenoma formation, the gene signature of Apc min/+/Zeb1 +/+ and Apc min/+/Zeb1 +/- adenomas was determined by RNA sequencing. A modern day light microscope has evolved from a tool devoted to making primarily empirical observations to what is now a sophisticated, quantitative device that is an integral part of both physical and life science research. Nowadays, microscopes are found in nearly every experimental laboratory. However, despite their prevalent use in capturing and quantifying scientific phenomena, neither a thorough understanding of the principles underlying quantitative imaging techniques nor appropriate knowledge of how to calibrate, operate and maintain microscopes can be taken for granted. This is clearly demonstrated by the well-documented and widespread difficulties that are routinely encountered in evaluating acquired data and reproducing scientific experiments. Indeed, studies have shown that more than 70% of researchers have tried and failed to repeat another scientist’s experiments, while more than half have even failed to reproduce their own experiments1. One factor behind the reproducibility crisis of experiments published in scientific journals is the frequent underreporting of imaging methods caused by a lack of awareness and/or a lack of knowledge of the applied technique2. Whereas quality control procedures for some methods used in biomedical research, such as genomics (e.g., DNA sequencing, RNA-seq) or cytometry, have been introduced (e.g. ENCODE3), this issue has not been tackled for optical microscopy instrumentation and images. Although many calibration standards and protocols have been published, there is a lack of awareness and agreement on common standards and guidelines for quality assessment and reproducibility4. In April 2020, the QUality Assessment and REProducibility for Instruments and Images in Light Microscopy (QUAREP-LiMi) initiative5 was formed. Selective recruitment of host factors by HSV-1 replication centers

Herpes simplex virus type 1（HSV-1） enters productive infection after infecting epithelial cells, where it controls the host nucleus to make viral proteins, starts viral DNA synthesis and assembles infectious virions. In this process, replicating viral genomes are organized into replication centers to facilitate viral growth. HSV-1 is known to use host factors, including host chromatin and host transcription regulators, to transcribe its genes; however, the invading virus also encounters host defense and stress responses to inhibit viral growth. Recently, we found that HSV-1 replication centers recruit host factor CTCF but exclude γH2A.X. Thus, HSV-1 replication centers may selectively recruit cellular factors needed for viral growth, while excluding host factors that are deleterious for viral transcription or replication. Here we report that the viral replication centers selectively excluded modified histone H3, including heterochromatin mark H3K9me3, H3S10 P and active chromatin mark H3K4me3, but not unmodified H3. We found a dynamic association between the viral replication centers and host RNA polymerase II. The centers also recruited components of the DNA damage response pathway, including 53BP1, BRCA1 and host antiviral protein SP100. Importantly, we found that ATM kinase was needed for the recruitment of CTCF to the viral centers. These results suggest that the HSV-1 replication centers took advantage of host signaling pathways to actively recruit or exclude host factors to benefit viral growth.

The definition of an oncogene is a gene that actively promotes tumorigenesis. For example, activation of RAS oncogene promotes cell transformation and cancer. Paradoxically, in primary mammalian cells, oncogenic RAS typically triggers cellular senescence, a state of irreversible cell growth arrest. Oncogene-induced senescence is an important tumor suppression mechanism in vivo. Here, we discuss our recent evidence that RAS-induced suppression of DNA repair response via dissociation of BRCA1 from chromatin promotes senescence while predisposing cells to senescence bypass and transformation by allowing for secondary hits. The molecular mechanism we uncovered helps reconcile the tumor-promoting nature of oncogenic RAS with the tumor-suppressing role of oncogene-induced senescence. © 2012 Landes Bioscience.

Histone acetyltransferases (HATs) and histone deacetylases (HDACs) conduct many critical functions through nonhistone substrates in metazoans, but only chromatin-associated nonhistone substrates are known in Saccharomyces cerevisiae. Using yeast proteome microarrays, we identified and validated many nonchromatin substrates of the essential nucleosome acetyltransferase of H4 (NuA4) complex. Among these, acetylation sites (Lys19 and 514) of phosphoenolpyruvate carboxykinase (Pck1p) were determined by tandem mass spectrometry. Acetylation at Lys514 was crucial for enzymatic activity and the ability of yeast cells to grow on nonfermentable carbon sources. Furthermore, Sir2p deacetylated Pck1p both in vitro and in vivo. Loss of Pck1p activity blocked the extension of yeast chronological life span caused by water starvation. In human hepatocellular carcinoma (HepG2) cells, human Pck1 acetylation and glucose production were dependent on TIP60, the human homolog of ESA1. Our findings demonstrate a regulatory function for the NuA4 complex in glucose metabolism and life span by acetylating a critical metabolic enzyme. © 2009 Elsevier Inc. All rights reserved.
