Optimization of process parameters for manufacturing of individual surfaces and the product as a whole is one of the key tasks of technological preparation of production. Processing of individual design elements of the product is a structurally complex process, which is a clear sequence of actions characterized by a set of parameters. The effectiveness of the optimization process directly depends on the level of detail of the process under study and the optimal choice of targets and control parameters. Thus, for a single design element of the product, the control task is reduced to the structural and parametric optimization of its processing. For this purpose, it is necessary to develop a hierarchical model of structural and parametric optimization of the processing of individual design elements of the product. The paper presents a structural hierarchical model of the design element forming process, describing the sequence of actions and intermediate states of the object at different control levels. The model is based on the structural decomposition of the process and includes three levels: technological processing route, technological transition, and working stroke. For each level, theoretical-multiple models, vector optimization criteria, control parameters, and boundary conditions are defined. The article also demonstrates the application of the model on the example of optimization for the process of shaping a group of threaded holes, where the Pareto-optimization method was used to obtain optimal machining parameters that reduced the labor intensity with an acceptable increase in error and surface roughness. The paper presents the results of controlling the machining process of a group of threaded holes by means of structural optimization of the machining process route and parametric optimization of cutting parameters for each working stroke. The hydrolysis of DNA serves as the foundational principle for gene engineering that enable precise gene cleavage at the molecular level. This process typically occurs in biological nucleases, which exhibit nucleobase-selective and catalytic hydrolysis capabilities rarely replicated in abiotic nanomaterials. Here, we demonstrate that molybdenum-terminated (Mo-terminated) edges of molybdenum disulfide (MoS2) possess the unique ability to abstract a proton (H+) from water molecules, thereby facilitating catalytic hydrolysis reactions that cleave the phosphodiester bonds in DNA through the action of hydroxide ions. The enhanced proton absorption at Mo-terminated edges of MoS2 significantly reduces the activation energy required for the DNA hydrolysis reaction. Furthermore, the favorable interaction between the Mo-terminated edges and thymine nucleobases promotes both charge transfer and P-O bond cleavage, enabling targeted DNA hydrolysis at 'TTTTTTT' sequences under dark conditions. This discovery underscores the potential of MoS2 as a stable, efficient nanosystem for precise genetic editing, heralding advanced applications in the field of gene engineering. © 2024, The Authors. Impact of flood depth on traffic volume in two different zones of the Bangkok road network was investigated using traffic data obtained from probe vehicle trajectories.A Macroscopic Fundamental Diagram(MFD)was utilized to compare traffic flow rates across two road network zones in the city for a variety of flood depths,namely 0-5 cm,5-10 cm,10-15 cm,15-30 cm,and more than 30 *** of empirical analysis over an observation period of one year as 2019 showed that flood depths had a strong correlation with the MFD parameters of free-flow speed,maximum flow,and traffic jam *** particular,road floods greatly reduced average maximum flow across the inner city road network in *** floods had a significant impact on traffic characteristics of urban road networks.

Reactive oxygen species (ROS), released as byproducts of mitochondrial metabolism or as products of NADPH oxidases and other processes, can directly oxidize the active-site cysteine (Cys) residue of protein tyrosine phosphatases (PTPs) in a mammalian cell. Robust degradation of irreversibly oxidized PTPs is essential for preventing accumulation of these permanently inactive enzymes. However, the mechanism underlying the degradation of these proteins was unknown. In this study, we found that the active-site Cys215 of endogenous PTP1B is sulfonated in H9c2 cardiomyocytes under physiological conditions. The sulfonation of Cys215 led PTP1B to exhibit a conformational change, and drive the subsequent ubiquitination and degradation of this protein. We then discovered that Cullin1, an E3 ligase, interacts with the Cys215-sulfonated PTP1B. The functional impairment of Cullin1 prevented PTP1B from oxidation-dependent ubiquitination and degradation in H9c2 cells. Moreover, delivery of the terminally oxidized PTP1B resulted in proteotoxicity-caused injury in the affected cells. In conclusion, we elucidate how sulfonation of the active-site Cys215 can direct turnover of endogenous PTP1B through the engagement of ubiquitin-proteasome system. These data highlight a novel mechanism that maintains PTP homeostasis in cardiomyocytes with constitutive ROS production.

3D stratigraphic modelling of the Bangkok basin using Kriging on borehole data

The Lower Central Plain of Thailand has a deep and highly irregular basement filled with complex layers of alluvial sandy soil and deltaic clay or silt. The Bangkok Metropolitan region with its high level of infrastructure development is located in this plain. With high population density, the problem of land subsidence is critical. This study uses borehole data and the Kriging method to interpolate the data. A detailed 3D stratigraphic model of the basin is presented and several cross sections along two directions. Seventeen layers and the points used for modelling each layer are presented. The model shows eight aquifers in the Bangkok basin, lying between eight layers of clay, and a bottom layer above the basement. The bottom of the lowest aquifer of the Bangkok aquifer system is 610 m deep. The basement extends down to a depth of more than 2000 m in some places. Our 3D model, which extends to the basement, is of interest for mining and site-specific seismic risk analysis. Moreover, our results can be very useful for groundwater and land subsidence studies.

Data combination for problem-solving: A case of an open data exchange platform

In recent years, rather than enclosing data within a single organization, exchanging and combining data from different domains has become an emerging practice. Many studies have discussed the economic and utility value of data and data exchange, but the characteristics of data that contribute to problem solving through data combination have not been fully understood. In big data and interdisciplinary data combinations, large-scale data with many variables are expected to be used, and value is expected to be created by combining data as much as possible. In this study, we conduct three experiments to investigate the characteristics of data, focusing on the relationships between data combinations and variables in each dataset, using empirical data shared by the local government. The results indicate that even datasets that have a few variables are frequently used to propose solutions for problem solving. Moreover, we found that even if the datasets in the solution do not have common variables, there are some well-established solutions to the problems. The findings of this study shed light on mechanisms behind data combination for problem-solving involving multiple datasets and variables. © 2020, CC BY-SA.

Social infrastructure such as roads and bridges have to be maintained under both strict budgets of local governments and scarcity of skillful engineers. Maintenance policies which consider various kind of factors such as maintenance periods, cost, degradation level, and natural disaster distribution, have to be established. In this paper, considering natural disasters, we form stochastically cumulative damage models and discuss their optimal policies theoretically and numerically. The total expected cost rates are obtained and optimal preventive maintenance levels which minimize them, are derived. Based this model, we consider extended models with a natural disaster and its recovery. © 2019 International Society of Science and Applied Technologies. All rights reserved.

Among a set of tools to emerge in recent years in environmental management, one appears to meet the criteria necessary to bridge this gap is the environmental footprint analysis based on biophysical reality of sustainable living, people must use natural renewable products and services not sooner than they are replacedand produces no waste faster than nature can absorb. The purpose of this study is to develop a model of noise environment footprint toward sustainability airport with the environmental assessment at Soekarno-Hatta International Airport. The results showed that the airport ecological footprint of the model can be built through the factors analysis that can give an idea of the influence of ecoairport. The conclusion of this study is to obtain the noise ecological footprint provides a method for the total impact of air transport to be assessed, the ecological footprint can be used as a communication and awareness tool to inform communities about the impact they make. It can lead to shifts to public transport for airport access, ecological footprint can be used by airports and airlines to identify operations where energy efficiency measures can be made and where resource consumption can be reduced

Soekarno-Hatta International Airport is prone to various environmental problems. The problem of population growth has an impact on increasing economic activity which directly impacts the environment. In every production activity from the environmental side, it will bring up the carrying capacity of the environment and the carrying capacity of the environment or ecological footprint. The problem will be difficult if it turns out that the ecological footprint that occurs exceeds the capacity of the existing environmental carrying capacity, also for airport problems. This will result in the airport being unsustainable. This article will discuss the model of the airport environmental footprint that is currently occurring at Soekarno-Hatta International Airport by utilizing the opinions of respondents who live and have activities around the airport. The factor analysis method is used to process the respondent's data to obtain the factors that influence the environmental footprint and then through these findings a model can be developed which is expected to be able to abstract the current conditions. The research results show that the dominant environmental footprint at Soekarno-Hatta airport is overload airport activities impact, operational slot time optimization, and renewable of airport energy, which can then be built as an instrument to evaluate, build and develop airport control policies that are environmentally friendly and sustainable.

In an oil drilling area, there are two oil drilling companies that have obtained permits to drill. As time goes by, the availability of existing petroleum decreases. To anticipate natural resources the government only prioritizes one company. Competition occurs between the two of them to explore petroleum from the ground so that they can drill. The purpose of this study is to provide an overview, how to make a system dynamics model related to the problem of oil drilling as an increasingly limited natural resource, by taking the case of the relationship between drilling companies in facing business competition. The method used is system dynamics with a standard model of mutual difficulties (drifting goals). The result obtained is a system dynamics model that can be used to demonstrate the behavior of a simulated petroleum drilling model to get an overview or condition of the limitations of petroleum and future availability.
