Questionnaire design for analyzing important risk factors of cervical cancer

Cervical cancer is Indonesia's second leading cause of death, following only breast cancer. According to the World Health Organization (WHO), non-communicable disorders will be blamed for 70% of all deaths and illnesses in 2020, while other disease types will account for 60%. Cervical cancer can be detected earlier, lowering the chance of death. This study aims to validate and check the pap smear test results obtained from the previously conducted pap smear screening program. The pap smear questionnaire developed would help in the compilation of a patient's habit history that has undergone a pap smear test during his life based on the factors associated with the occurrence of cervical cancer. Specific details, social-demographic features, diet, genital hygiene activities, reproductive background and behaviors, and health care and cancer prevention behavior are also included in the questionnaire template. The consequence of this design is the completeness of evidence on the theory of anomalies dependent on the findings of pap smear sampling.

Loss of heterochromatin has been implicated as a cause of pre-mature aging and age-associated decline in organ functions in mammals;however,the specific cell types and gene loci affected by this type of epigenetic change have remained *** address this knowledge gap,we probed chromatin accessibility at single-cell resolution in the brains,hearts,skeletal muscles,and bone marrows from young,middle-aged,and old mice,and assessed age-associated changes at 353,126 candidate cis-regulatory elements(cCREs)across 32 major cell ***,we detected increased chromatin accessibility within specific heterochromatin domains in old mouse excitatory *** gain of chromatin accessibility at these genomic loci was accompanied by the cell-type-specific loss of heterochromatin and activation of LINE1 *** further confirmed the loss of the heterochromatin mark H3K9me3 in the excitatory neurons but not in inhibitory neurons or glial *** results reveal the cell-type-specific changes in chromatin landscapes in old mice and shed light on the scope of heterochromatin loss in mammalian aging.

Worldwide outbreaks of hand, foot, and mouth disease (HFMD) are caused by enterovirus A71 (EV-A71) and coxsackievirus A16 (CV-A16). Since no anti-HFMD drugs are currently available, it is interesting to study potential viral inhibitors. Rupintrivir is a rhinovirus 3C protease (3Cpro) inhibitor with apparent inhibitory activity against HFMD. This study constructs pharmacophore models of the EV-A71 and CV-A16 3Cpro complexed with rupintrivir using molecular dynamics (MD) simulations. Both models had similar pharmacophore features, including hydrogen bond donors and acceptors and hydrophobic interactions. These pharmacophore models were then used as a template to screen 39 flavonoid compounds as potential novel inhibitors. Diosmin, epigallocatechin gallate (EGCG), and RTH-011 showed high binding affinities for EV-A71 and CV-A16 3Cpro. They formed hydrogen bonds with important surrounding residues in both proteins, including H40, L127, T142, A144, T145, H161, I162, G163, and G164. In addition, their effective concentrations against rhabdomyosarcoma (RD) cell infection by EV-A71 and CV-A16 were determined. EGCG had the highest half maximal effective concentration (EC50) of 12.86 ± 1.30 µM for EV-A71 and 15.54 ± 1.50 µM for CV-A16, while diosmin had EC50 of 21.01±1.57 µM for EV-A71 and 30.68 ± 3.25 µM for CV-A16. Both compounds were non-toxic in RD cells, with 50% cytotoxic concentrations of >100 µM for EGCG and >500 µM for diosmin. In addition, MD simulation analysis showed that EGCG had a higher binding affinity than diosmin, supported by its significantly lower solvated binding free energies and greater numbers of contact atoms and key binding residues. Moreover, previous studies reported EGCG’s inhibitory effect on other viruses, such as severe acute respiratory syndrome coronavirus 2. Therefore, our findings suggest that EGCG can effectively inhibit the EV-A71 and CV-A16 3Cpro. © 2023, The Authors. All rights reserved.

This study aims to develop a valid and reliable video analysis instrument to determine teachers’ character in the opening lesson in terms of its utterances and produce video analysis on micro-teaching that will be used in training related to Artificial Intelligence (AI). The type of research used is research and development. The subject of this research is the micro-teaching video of class E students’ batch 2019, Mathematics Education Study program at Sanata Dharma University. The product of this research is a video analysis instrument to find out the character of the prospective teachers, such as confidence, enthusiasm, and happiness in terms of voice utterances. This study uses the ADDIE research model. The results show that the video analysis instrument has a validity value of 4.80 which means it's very valid. Video analysis instrument to find out the enthusiastic character has a value of validity of 4.70, which means it's very valid. Then, the video analysis instrument to find out the character of happiness has a validity value of 4.60 which means it's very valid. The practicality of the developed video analysis instrument meets the required criteria with a practicality value of 4.31. Overall, the video analysis instrument developed belongs to an effective category.

Mathematical modeling in the epidemiology study can be applied to describe the current transmission of viruses, one of which is compartments. However, this simulation model is rigorous to understand, especially in interpreting the parameter values in influencing the solution. Therefore, it is necessary to present a coherent mathematical model solution. This study aims to determine the SEIRD model solution in the COVID-19 transmission using Microsoft Excel with three conditions (normal, new normal, and lockdown) to facilitate the interpretation of data. The SEIRD model used in this study considers natural population growth, namely natural births and deaths. Three stages to evaluate the model solution in this study are constructing a mathematical model, deciding the parameter intervals, and creating an applet in Microsoft Excel. The system of differential equations is converted into a system of difference equations to obtain numerical model solutions. The results showed that the differences in the infection rates for old normal, new normal, and lockdown conditions were 24%, 4%, and 3%, respectively.

Assembling optimal microbial communities is key for various applications in biofuel production, agriculture, and human health. Finding the optimal community is challenging because the number of possible communities grows exponentially with the number of species, and so an exhaustive search cannot be performed even for a dozen species. A heuristic search that improves community function by adding or removing one species at a time is more practical, but it is unknown whether this strategy can discover an optimal or nearly optimal community. Using consumer-resource models with and without cross-feeding, we investigate how the efficacy of search depends on the distribution of resources, niche overlap, cross-feeding, and other aspects of community ecology. We show that search efficacy is determined by the ruggedness of the appropriately-defined ecological landscape. We identify specific ruggedness measures that are both predictive of search performance and robust to noise and low sampling density. The feasibility of our approach is demonstrated using experimental data from a soil microbial community. Overall, our results establish the conditions necessary for the success of the heuristic search and provide concrete design principles for building high-performing microbial consortia. © 2021, CC BY-NC-ND.

This study investigated the effect of metal-impregnated carbon cloth (CC) supplementation on thermophilic anaerobic digestion (AD) improvement utilizing municipal sludge as a biomass input. Four types of metal-impregnated CC nanocomposites (Fe/CC, Ni/CC, Ni/Fe/CC, NiPcS/Fe/CC) were synthesized from activated CC (A-CC) using hydrothermal synthesis and compared with untreated CC (U-CC) to evaluate their functional significance for enhanced AD. The main factors inspected were electrical conductivity (EC) and Brunauer–Emmett–Teller (BET) surface area. A-CC had the highest surface area of 386.15 m2/g, followed by Fe/CC, NiPcS/Fe/CC, Ni/CC and Ni/Fe/CC with 140.96, 52.27, 43.52 and 36.07 m2/g, respectively. The A-CC and Fe/CC reactors produced 3.45 and 2.70 times more methane than Control (without CC) in the first 9 days. In contrast, the methane production for U-CC, Ni/CC, Ni/Fe/CC and NiPcS/Fe/CC was only 0.87, 1.26, 1.01 and 1.29 times, respectively, compared to Control. At a higher BET surface (>350 m2/g), the surface area becomes a more dominant factor, and the increase of EC value (>2.9*104 1/Ω.m) does not have an impact on methane enhancement. Microbial community analysis revealed that A-CC had the highest abundances of Syntrophomonas, Coprothermobacter and methanogenic populations, including Methanothermobacter and Methanosarcina, within 3 days compared to Control and metal-impregnated CC. There were minimal changes in microbial community composition in metal-impregnated CC compared to the U-CC. The result implies that the efficacy of CC supplementation for AD upgrading largely depends on the higher surface area compared to metal impregnation or conductivity. © 2023, The Authors. Having good sleep is important to maintain physical and mental health. However, many young adults suffer poor sleep due to work or academic demands. Even more, sleep is associated with stress which is many young adults experienced it. To solve this problem, many epidemiologic studies suggest that exercise may improve sleep quality. On the other hand, several studies show that daily activity has no association with sleep quality. In this study, we tried to find more evidence of whether the daily activity has an association with sleep quality or not. We used a commercially available wearable device to capture the subject's heart rate, step count and deep sleep duration for 7 months. Then we used linear regression, XGBoost, and Long Short-Term Memory to find an association between these variables. The results show that there was no association between these variables.

BACKGROUND:Splicing variants are a major class of pathogenic mutations, with their severity equivalent to nonsense mutations. However, redundant and degenerate splicing signals hinder functional assessments of sequence variations within introns, particularly at branch sites. We have established a massively parallel splicing assay to assess the impact on splicing of 11,191 disease-relevant variants. Based on the experimental results, we then applied regression-based methods to identify factors determining splicing decisions and their respective weights. RESULTS:Our statistical modeling is highly sensitive, accurately annotating the splicing defects of near-exon intronic variants, outperforming state-of-the-art predictive tools. We have incorporated the algorithm and branchpoint information into a web-based tool, SpliceAPP, to provide an interactive application. This user-friendly website allows users to upload any genetic variants with genome coordinates (e.g., chr15 74,687,208 A G), and the tool will output predictions for splicing error scores and evaluate the impact on nearby splice sites. Additionally, users can query branch site information within the region of interest. CONCLUSIONS:In summary, SpliceAPP represents a pioneering approach to screening pathogenic intronic variants, contributing to the development of precision medicine. Purpose: Obstetric brachial plexus lesions (OBPL) are caused by the lesion or rupture of the cervical and thoracic nerves during birth. Physical therapy, based on ludic activities, can be useful to improve the OBPL treatment in children. This paper proposes a rehabilitation platform with a ludic interface (lights and sounds), using the magnitude motion gain of a user to aid in the OBPL treatment. Methods: The motor development table (MDT) is an electronic platform composed of eight light segments (with green and red LEDs) that detect the touch of a child’s hand (by optic proximity sensors). Each touch generates a different sound, depending on the exercise. The physiotherapist can choose the MDT game, and the child score can be recorded. The operation of the rehabilitation platform was tested in relation to its behavior depending on the environmental lighting, its efficiency in data processing, and its maximum distance of drive. We also applied an opinion survey. Results: The tests showed that the MDT can be used in environments with up to 1015 lx of illumination and that the incidence of high-intensity light affects the segments’ ability to detect the hand. In addition, the tests demonstrated that the behavior of this device is not affected by the number of errors or triggers performed during the exercises. Conclusion: Based on these findings and on the opinion survey, it is possible to conclude that the MDT can reliably assist the physiotherapist in orthopedic management by providing feedback from the sessions and the patients’ temporal progress. © 2019, Sociedade Brasileira de Engenharia Biomedica.
