Effect of thigh muscle strength training through telerehabilitation in volunteers with low back pain. A controlled and randomized clinical trial

Background: Low back pain (LBP) is a clinical condition with a high global incidence that significantly impacts the quality of life, including psychological, physical, and social aspects. It commonly leads to an impairment in muscle strength and activation. Research question: The aim of the study was to assess the short-term effects of thigh muscle strength training conducted via telerehabilitation and in-person in patients with self-reported LBP. Methods: The telerehabilitation and control groups completed 10 strength training sessions, five times a week. Both groups were evaluated for electromyographic parameters, maximal isometric voluntary contraction (MIVC), and the number of maximum repetitions of knee flexion-extension movements. Results: Telerehabilitation increased the recruitment of the right rectus femoris (p < 0.05). It improved the maximum repetition capacity for the left knee extension (p < 0.01) movement and for the right and left knee flexion(p < 0.05);however, it was able to reduce the MIVC right and left knee flexion (p < 0.05). Conclusion: Telerehabilitation was able to increase the recruitment of the right biceps femoris muscles and maintain the recruitment of the right and left rectus femoris;improve the maximum repetition capacity for left knee extension and of both the right and left knees flexion movement. However, it could not increase the MVIC after the training period. © 2025 Elsevier Ltd

Transcriptomic implications of low herbicide concentrations in hepatic cells: Insights into the individual and combined effects of 2,4-D, glyphosate, and AMPA

Glyphosate and 2,4-D are among the most widely used herbicides globally, leading to environmental presence, food contamination, and human contact. Investigations based on current toxicological limits or populational-based herbicide exposures are warranted, and in vitro bioassays provide useful tools for toxicological screening. Thus, this study evaluated the transcriptomic implications of non-cytotoxic exposures to glyphosate, its metabolite aminomethylphosphonic acid (AMPA), or 2,4-D - or to their mixes - on hepatic cells. The half maximal effective concentration (IC50) of each herbicide was calculated (cell viability) in human hepatic C3A cells and 1000-fold lower concentrations were used for transcriptomic analysis (RNA-Seq) after 48 h exposure, resembling current toxicological limits and considering herbicide water levels (glyphosate: 0.95 µg/mL;AMPA: 3.7 µg/mL;2,4-D: 0.56 µg/mL). Glyphosate exposure enriched MAPK-related biological processes (upregulated TNF, FOS, IGF1, and PDGFB), and downregulated genes associated with lipid metabolism (CD36 and PPARA). Many AMPA exposure-related differentially expressed genes (DEGs, such as PFKFB3, HK2, and ALDOA) were associated with glucose metabolic pathways. Glyphosate and its metabolite yielded a common molecular signature, as illustrated by principal component analysis and the function of 212 shared DEGs. The exposure to 2,4-D was associated with the JNK cascade and the solute carrier family annotations. The herbicide mixtures had a discrete effect on enhancing the impact of individual herbicides, although important epithelial-mesenchymal transition genes were exclusively modified by the mixes (COL11A2, LOXL3, SNAI1). Altogether, our data reveals new perspectives on the short-term molecular effects of herbicide exposure in liver cells, emphasizing potential avenues for further exploration. © 2025 Elsevier B.V.

Penile cancer, although rare, has an increasing mortality rate in Brazil, highlighting the need for effective diagnostic methods. Artificial Intelligence (AI) in histopathological analysis can speed up and objectify diagnosis, but designing an ideal architecture is challenging. In this study, we propose a neural architecture search (NAS) methodology for detecting penile cancer in digital histopathology images. We explored different configurations of stem blocks and the inclusion of attention mechanisms, highlighting specific preferences depending on the magnification of the images. The results showed that the NAS methodology enabled the discovery of more accurate and optimized architectures for this task, surpassing conventional models. The proposed models achieve 89.5% and 88.5% F1-Score for 40X and 100X magnification, respectively. © 2025 The Author(s).

The dietary inflammatory index (DII) has been associated with obesity and cardiovascular risk factors (CVRF) in the general population. We hypothesized that in kidney transplant recipients (KTR), a positive relationship between DII, body adiposity and CVRF would also be observed. To test this hypothesis, we conducted a cross-sectional study with adult KTR. Body mass index (BMI), body adiposity index (BAI) and waist circumference (WC) were assessed. Total fat mass (FM), trunk FM, and load-capacity index (LCI) were evaluated using dual-energy X-ray absorptiometry. Energy-adjusted DII (E-DII) was estimated based on three 24-h recalls and stratified as anti-inflammatory (E-DII<0) and pro-inflammatory (E-DII>0). CVRF included hypertension, diabetes, dyslipidemia, and metabolic syndrome. A total of 170 KTR, 59% male, with 49.5 (42-57) years and E-DII from -2.89 to 4.78 were evaluated. KTR with E-DII>0, compared to those with E-DII<0, exhibited significantly higher values of BAI, total FM (kg), and LCI. In multiple adjusted linear regression, E-DII was significantly associated with WC, total FM (kg), and trunk FM (kg). Logistic regression analysis indicated that E-DII>0 was significantly associated with obesity, as assessed by BAI. E-DII was not associated with CVRF. The present study suggests that a pro-inflammatory diet is associated with higher total and central body adiposity in KTR. Interventions targeting an anti-inflammatory diet may contribute to reducing excessive body adiposity in this population. © 2025 Elsevier Inc.

Background & aims Recent evidence suggests that gut microbiota has a potential role in the pathophysiology of obesity and other cardiovascular disease (CVD) risk factors, including hypertension, dyslipidemia, and type 2 diabetes. However, clinical trials evaluating the effects of probiotics supplementation on these outcomes have found inconsistent results, probably due to the wide heterogeneity in trial designs. In addition, there is a lack of studies investigating whether probiotics can enhance the beneficial effects of caloric restriction in individuals with increased risk of CVD as individuals with hypertension and excess body adiposity. Thus, the aim of this study was to evaluate the effects of multi-strain probiotics supplementation on body adiposity, glycemic homeostasis, lipid profile, and serum adipokine levels in individuals with hypertension and excess body weight following an energy restricted diet. Methods A randomized, double-blind, placebo controlled clinical trial was conducted for 12 weeks. Were included 66 individuals aged between 40 and 65 years; both sexes; body mass index (BMI) ≥ 25 and < 40 kg/m 2 and diagnosis of hypertension. Were excluded smokers; individuals using probiotics, prebiotics, symbiotics and antibiotics in the last 3 months; presenting diabetes, chronic kidney disease or liver failure; and pregnant and lactating women. Participants were allocated into 2 groups: group with supplementation of 8 probiotic strains in capsules (3 × 10 10 CFU/day) or control group (placebo capsules). Both groups followed a low-calorie diet. Participants underwent anthropometric, body composition (dual-energy radiological absorptiometry) and biochemical (glucose metabolism, lipid profile, adiponectin, and leptin) evaluation at baseline and at the end of the study. The rat posterodorsal medial amygdala (MePD) is sexually dimorphic, has a high concentration of receptors for gonadal hormones and prolactin (PRL), and modulates reproduction. To unravel genetic and functional data for this relevant node of the social behavior network, we studied the expression of ERα, ERβ, GPER1, Kiss1, Kiss1R, PRGR, PRL, PRLR, EGR1, JAK2, STAT5A, and STAT5B in the MePD of males and females along the estrous cycle using the RT-qPCR technique. We also investigated whether PRL in the MePD would affect the sexual behavior display of proestrus females by microinjecting saline, the PRL receptor antagonist Del1-9-G129R-hPRL (1 µM and 10 µM), or PRL (1 nM) and Del1-9-G129R-hPRL (10 µM) 3 h before the onset of the dark-cycle period. The estrogen-dependent lordosis behavior, indicative of sexual receptivity of proestrus females, was recorded and compared before (control) and after (test) microinjections in these groups. Sex differences were found in the right and left MePD gene expression. ERα and Kiss1R, as well as PRL, Short PRLR, and STAT5B expression is higher in cycling females than males. Kiss1 expression is higher in males than females, and GPER1 is higher during diestrus than proestrus. Furthermore, Del1-9-G129R-hPRL in the MePD significantly reduced the full display and quotient of lordosis in proestrus females, an effect restored by the co-microinjection of PRL. In conjunction, the expression of studied genes showed specific sex and estrous cycle phase features, and PRL action in the MePD plays an essential role in the display of lordosis during the ovulatory period. © 2025 Elsevier B.V.

Lead (Pb) is one of the most harmful soil contaminants affecting plant growth. Cymbopogon citratus (lemongrass) stands out due to its essential oil rich in citral. Studies indicate that Azospirillum brasilense stimulates the development of lemongrass, optimizing nutrient uptake. The methodology involved a factorial greenhouse experiment using lemongrass grown in Pb-contaminated soil, with or without inoculation by A. brasilense (strains Ab-V5 and Ab-V6), and included evaluations of plant growth, Pb and nitrogen content, nitrate reductase activity, essential oil composition, and antibacterial activity. This study tested the effects of A. brasilense in soils with different Pb concentrations, analyzing growth parameters and essential oil composition. The results showed that lemongrass exhibited favorable root development at Pb concentrations up to 300 mg kg⁻¹, and A. brasilense improved nitrogen fixation and stress tolerance. The essential oil of lemongrass, free of Pb contamination, contains high levels of neral and geranial and demonstrated bactericidal activity against Xanthomonas citri and inhibitory effects on X. vesicatoria. These findings suggest that lemongrass oil can be used in biological pest control, highlighting its potential for sustainable applications in the cosmetic and natural disinfectant industries. © 2025 Elsevier B.V.

Are glyphosate or glyphosate-based herbicides linked to metabolic dysfunction-associated steatotic liver disease (MASLD)? The weight of current evidence

Metabolic dysfunction-associated steatotic liver disease (MASLD) affects around 30 % of the world's population, increasing its prevalence by 50 % in the last three decades. MASLD pathogenesis is considered multiaxial, involving disturbances in the liver, adipose tissue (AT), and gut microbiome. In parallel with MASLD increasing trends, the total herbicide use has nearly tripled over the last three decades. Glyphosate (GLY) is the most used herbicide worldwide (825 mi kg/year). The intensive use of GLY-based herbicides (GBH) – largely driven by the adoption of glyphosate-tolerant genetically modified crops over the past two decades - has led to environmental (soil and water) and food contamination, resulting in continuous human exposure. Emerging (pre)clinical data highlights the significant implications of this herbicide on MASLD, marking a critical research area. Thus, this narrative review paper aimed at gathering and evaluating all epidemiological and (pre)clinical data on the implications of GLY or GBH on MASLD outcomes. Our work encompassed literature published between 2008–2025. Human urinary GLY levels are associated with different MASLD outcomes (steatosis risk, advanced fibrosis, increased transaminases) and comorbidities (higher risk for metabolic syndrome, diabetes, obesity and cardiovascular diseases) (6 studies). In vitro data indicate that GBH/GLY cause oxidative stress, genomic instability, apoptosis, and membrane disruption in hepatocytes, while promoting apoptosis and lipid peroxidation in (pre)adipocytes and cytokine production in monocytes (15 studies). In rodent studies (21 studies), GLY/GBH - in doses based on human exposure/toxicological limits - induces inflammatory and oxidative responses in the liver and AT, while causing dysbiosis and metabolic alterations in the gut microbiome axis. Background: The first isoxazoline-based acaricide (fluralaner) for the control of Rhipicephalus microplus was introduced onto the market in 2022, initially in Brazil, followed by other Latin American countries. Therefore, it is important to establish laboratory methods to monitor the susceptibility of populations of R. microplus to this molecule and to determine the relationship between the results of laboratory tests and those from field trials. Methods: A larval immersion test (LIT) was performed on 18 populations of R. microplus. The lethal concentration 50 (concentration causing 50% mortality [LC50]) values were calculated to determine the resistance ratios (RRs) of the populations. The lethal concentration 99 (concentration causing 99% mortality [LC99]) values were calculated to determine the discriminating doses (DDs = 2 × LC99). The DDs were applied in tests with the POA (susceptible) and GYN (resistant) strains, as well as in tests with the population that presented the lowest LC50 value (population 14) and with the two populations that presented the highest LC50 value (populations 10 and 16). Finally, we performed field trials with the population that presented the lowest and two highest LC50 values. Results: In the LIT with fluralaner, the LC50 values ranged from 0.144 to 0.481 µg/mL for the 18 R. microplus populations. The mortality rate was 100% in the tests of the DDs in the five populations tested. In the field trials, the efficacy of fluralaner was similar for the three populations of R. microplus tested (populations 14, 10 and 16), with therapeutic efficacy (until day 21) of 100% and persistent efficacy (between days 28 and 42) > 95%. Conclusion: We observed natural variability in the susceptibility of larvae from the different populations of R. microplus that had never been treated with this compound. Despite the observed variability in the in vitro results (LC50), a comparable efficacy of > 90% lasting until day 42 was observed in the field trials.
