The role of genetic diversity in crop germplasm is an important concept within genetic conservation. In this research, 43 accessions were analyzed at the agro-morphological and genetic levels. Clustering based on the agro-morphological resulted in four sub-groups. Analysis at the genetic level was conducted using 22 microsatellites, which revealed a total number of alleles to be 203, with a range per allele between 2 and 17 and an average of 9.2 alleles per locus. The highest and lowest Polymorphic Information Content (PIC) values were found in RM431 and RM11, which were 0.95 and 0.67, respectively. The genetic diversity value ranged from 0.71 to 0.95. The genetic similarity among accessions ranged from 0.00 to 0.90. Clustering based on the genetic relatedness divided the Java rice samples into two major groups. The classification created through this research many inform future breeding programs aimed at improving the quality and quantity of yield production. © 2020, Korean Society of Crop Science (KSCS).

The wide distribution of mosquitoes in the territory of Indonesia with differences in geographical location causes mosquitoes to have the ability to adapt to the environment. The ability to adapt to the environment results in high variations phenotypically and genotypically so that Mutation, recombination or migration of genes is the cause of genetic diversity in a population. The purpose of this research is to study the characteristics and inventory of distribution areas, as well as to analyze genetic variations using morphological and molecular approaches. Genetic typology data through the RAPD approach provides information about the genetic variation of mosquitoes in an effort to control them as disease vectors. Genetic characterization was carried out by PCR-RAPD using three primers, namely OPA 11, OPA 12, OPA 15 and OPL12. Data were analyzed using UPGMA with a similarity coefficient using the Jaccard Coefficient of Similarity presented as a dendrogram. The results showed that the variation of the mosquito polymorphy band from the Kelurahan Kemelak Bindung Langit, Komering Ulu Regency was high. With the DNA band profile of RAPD results as many as 37 bands with a polymorphy DNA band presentation of (86.65%). © 2021 IEEE.

A Systematic Review of the Potential Effects of Propolis Extracts on Experimentally-induced Diabetes

Oxidative stress (OS) is involved in the development of diabetes mellitus (DM) and its complications. Thus, OS reduction may be an important strategy for DM therapy. Propolis is bee resins with high antioxidant activity and is used in the treatment of different diseases, including DM. Therefore, in this systematic review, we evaluated the impact of propolis administration in diabetic animals. We used the PRISMA strategy to collect preclinical studies published in English up to November 2021 in three databases (PubMed/Medline, Scopus, and Web of Science). We used the SYRCLE tool to analyze the risk of methodological bias. Our primary search returned 198 studies, of which 14 were considered eligible to be included in this review. The administration of propolis induced a hypoglycemic effect in the treated animals, which is probably due to the reduction of OS. The animals showed restoration of endogenous antioxidant defenses and reduced levels of markers for OS. The administration of propolis resulted in improvement in the lipid profile of treated animals. Our risk of bias assessment showed a methodological quality score of less than 30% due to a lack of randomization, blinding, and proper allocation of animals. Heterogeneity in treatments, lack of results, and use of non-standard extracts are limitations in our data analysis. Despite these limitations, propolis induced a significant hypoglycemic effect in diabetic animals when compared to untreated controls. This effect was associated with a reduction in OS, a process mediated by ROS neutralization and restoration of endogenous antioxidant defenses.

We introduce two thermodynamics models of solid-liquid phase equilibrium of a binary system in the presence of (a) a chemical reaction in the liquid involving only one of the binary components and, (b) strain-inducing epitaxial conditions on the solid in addition to the chemical reaction in the liquid. These models show the existence of the counter intuitive behavior that epitaxy may promote recrystallization of the solid in spite of the reaction driving dissolution. The conditions for this recrystallization behavior are determined by the value of the epitaxial lattice matching composition, the temperature and the overall composition of the binary solid-liquid system. © 2023, The Authors. Exergames can become an alternative tool for enhancing neuromotor capacity from routines and systematization. Thirty-two students (age group: 8 - 10 years) were evaluated and divided into two random groups, the control group - CG (n = 10) and the exergame group - EG (n = 22). Flexibility capacity and abdominal and upper limbs resistance were evaluated before and after 16 weeks. A 2x2 factorial ANOVA, Post-Hoc Bonferroni, and the effect size was calculated using SPSS for Windows v.23. Significant changes in flexibility were observed, which F (1.30) = 7.683; p < 0.01; r = 0.45; EG presented significant alteration F (1.30) = 15.1; p < 0.01; r = 0.57, however, CG showed no significant changes F (1.30) = 0.52; p = 0.47; r = 0.13. For the upper limbs strength analysis, only EG presented significant adaptation F (1.30) = 5.01; p < 0.05; r = 0.37. Finally, there were no significant changes in the variable abdominal strength.

N-ACT: An Interpretable Deep Learning Model for Automatic Cell Type and Salient Gene Identification

Single-cell RNA sequencing (scRNAseq) is rapidly advancing our understanding of cellular composition within complex tissues and organisms. A major limitation in most scRNAseq analysis pipelines is the reliance on manual annotations to determine cell identities, which are time consuming, subjective, and require expertise. Given the surge in cell sequencing, supervised methods–especially deep learning models–have been developed for automatic cell type identification (ACTI), which achieve high accuracy and scalability. However, all existing deep learning frameworks for ACTI lack interpretability and are used as "black-box" models. We present N-ACT (Neural-Attention for Cell Type identification): the first-of-its-kind interpretable deep neural network for ACTI utilizing neural attention to detect salient genes for use in cell-types identification. We compare N-ACT to conventional annotation methods on two previously manually annotated data sets, demonstrating that N-ACT accurately identifies marker genes and cell types in an unsupervised manner, while performing comparably on multiple data sets to current state-of-the-art model in traditional supervised ACTI. © 2022, CC BY.

Baccharis trimera aqueous extract modulates inflammation and nociception in mice

The aerial parts of Baccharis trimera (Less.) are frequently used as a tea to treat several diseases. Therefore, the aim of this study was to identify the constituents of an aqueous extract of B. trimera, focusing on their antioxidant, anti-inflammation, and antinociception activities and properties. For that, the researchers performed in vivo assays using the formalin test and Freund’s Complete Adjuvant (FCA) to measure the acute and chronic inflammatory pain in mice. Moreover, the myeloperoxidase enzyme (MPO) was analyzed in the subcutaneous tissue after the FCA injection, together with the counting of lymphocytes in the peripheral blood of the mice. The qualitative phytochemical analysis indicated the presence of flavonoids and saponins in the B. trimera aqueous extract. The high-performance liquid chromatography (HPLC) analyses showed the presence of phenolic compounds, such as chlorogenic acid, ellagic acid, rosmarinic acid, as well as flavonoids, such as rutin, quercetin, and luteolin. The DPPH assay was used in order to measure the antioxidant activity of the aqueous extract of B. trimera and this showed an IC50 of 118.18 ± 1.02 μg/mg. The data from the formalin test demonstrated that a single dose of the aqueous extract of B. trimera was not able to decrease the nociceptive behavior during the neurogenic phase, at any of the tested doses (20, 40, or 80 mg/kg p.o.). However, during the inflammatory phase of this test, the aqueous extract of B. trimera at 80 mg/kg (p.o.) significantly decreased the nociceptive behavior, showing more effectiveness when compared to the other tested doses (p < 0.05). Importantly, in the chronic inflammatory model on the 5th day of treatment, the aqueous extract of B. trimera (80 mg/kg p.o.) significantly reduced mechanical allodynia (p < 0.01), heat thermal hyperalgesia (p < 0.001), and paw edema (p < 0.05). Despite efforts to elucidate Zika virus (ZIKV) teratogenesis, still several issues remain unresolved, particularly on the molecular mechanisms behind the pathogenesis of Congenital Zika Syndrome (CZS). To answer this question, we used bioinformatics tools, animal experiments and human gene expression analysis to investigate genes related to brain development potentially involved in CZS. Searches in databases for genes related to brain development and CZS were performed, and a protein interaction network was created. The expression of these genes was analyzed in a CZS animal model and secondary gene expression analysis (DGE) was performed in human cells exposed to ZIKV. A total of 2610 genes were identified in the databases, of which 1013 were connected. By applying centrality statistics of the global network, 36 candidate genes were identified, which, after selection resulted in nine genes. Gene expression analysis revealed distinctive expression patterns for PRKDC, PCNA, ATM, SMC3 as well as for FGF8 and SHH in the CZS model. Furthermore, DGE analysis altered expression of ATM, PRKDC, PCNA. In conclusion, systems biology are helpful tools to identify candidate genes to be validated in vitro and in vivo. PRKDC, PCNA, ATM, SMC3, FGF8 and SHH have altered expression in ZIKV-induced brain malformations. © Elsevier Ltd
