The outbreak and transmission of disease-causing pathogens are contributing to the unprecedented rate of biodiversity decline. Recent advances in genomics have coalesced into powerful tools to monitor, detect, and reconstruct the role of pathogens impacting wildlife populations. Wildlife researchers are thus uniquely positioned to merge ecological and evolutionary studies with genomic technologies to exploit unprecedented “Big Data” tools in disease research; however, many researchers lack the training and expertise required to use these computationally intensive methodologies. To address this disparity, the inaugural “Genomics of Disease in Wildlife” workshop assembled early to mid-career professionals with expertise across scientific disciplines (e.g., genomics, wildlife biology, veterinary sciences, and conservation management) for training in the application of genomic tools to wildlife disease research. A horizon scanning-like exercise, an activity to identify forthcoming trends and challenges, performed by the workshop participants identified and discussed 5 themes considered to be the most pressing to the application of genomics in wildlife disease research: 1) “Improving communication,” 2) “Methodological and analytical advancements,” 3) “Translation into practice,” 4) “Integrating landscape ecology and genomics,” and 5) “Emerging new questions.” Wide-ranging solutions from the horizon scan were international in scope, itemized both deficiencies and strengths in wildlife genomic initiatives, promoted the use of genomic technologies to unite wildlife and human disease research, and advocated best practices for optimal use of genomic tools in wildlife disease projects. The results offer a glimpse of the potential revolution in human and wildlife disease research possible through multi-disciplinary collaborations at local, regional, and global scales.

Several oxylipins are potent lipid mediators that regulate diverse aspects of health and disease and whose quantitative analysis by liquid chromatography-mass spectrometry (LC-MS) presents substantial technical challenges. As members of the lipidomics community, we developed technical recommendations to ensure best practices when quantifying oxylipins by LC-MS.

BACKGROUND:In patients resuscitated after cardiac arrest, a higher mean arterial pressure (MAP) may increase cerebral perfusion and attenuate hypoxic brain injury. Here we present the protocol of the mean arterial pressure after cardiac arrest and resuscitation (MAP-CARE) trial aiming to investigate the influence of MAP targets on patient outcomes. METHODS:MAP-CARE is one component of the Sedation, Temperature and Pressure after Cardiac Arrest and Resuscitation (STEPCARE) 2 x 2 x 2 factorial randomized trial. The MAP-CARE trial is an international, multicenter, parallel-group, investigator-initiated, superiority trial designed to test the hypothesis that targeting a higher (>85 mmHg) (intervention) versus a lower (>65 mmHg) (comparator) MAP after resuscitation from cardiac arrest reduces 6-month mortality (primary outcome). Trial participants are adults with sustained return of spontaneous circulation who are comatose following resuscitation from out-of-hospital cardiac arrest. The two other components of the STEPCARE trial evaluate sedation and temperature control strategies. Apart from the STEPCARE trial interventions, all other aspects of general intensive care will be according to the local practices of the participating site. Neurological prognostication will be performed according to European Resuscitation Council and European Society of Intensive Care Medicine guidelines by a physician blinded to allocation group. The sample size of 3500 participants provides 90% power with an alpha of 0.05 to detect a 5.6 absolute risk reduction in 6-month mortality, assuming a mortality of 60% in the control group. Secondary outcomes will be poor functional outcome 6 months after randomization, patient-reported overall health 6 months after randomization, and the proportion of participants with predefined severe adverse events. CONCLUSION:The MAP-CARE trial will investigate if targeting a higher MAP compared to a lower MAP during intensive care of adults who are comatose fo

The “QuantitatEVs: multiscale analyses, from bulk to single vesicle” workshop aimed to discuss quantitative strategies and harmonized wet and computational approaches toward the comprehensive analysis of extracellular vesicles (EVs) from bulk to single vesicle analyses with a special focus on emerging technologies. The workshop covered the key issues in the quantitative analysis of different EV-associated molecular components and EV biophysical features, which are considered the core of EV-associated biomarker discovery and validation for their clinical translation. The in-person-only workshop was held in Trento, Italy, from January 31^st to February 2^nd, 2023, and continued in Milan on February 3^rd with “Next Generation EVs,” a satellite event dedicated to early career researchers (ECR). This report summarizes the main topics and outcomes of the workshop. © 2024 The Authors. To gain insight into the active microbiota during spontaneous fermentation of Theobroma cacao L., this study assessed protein diversity during 120 h using a combined metabarconding and metaproteomics approach. During the first days of fermentation, most of the peptides were associated with T. cacao and yeast (0–72 h). Peptides associated with bacteria became more abundant after 72 h of fermentation, coinciding with a decrease in peptides associated with cocoa (96–120 h). In addition to the known microorganisms involved in fermentation, such as Saccharomyces, Lactobacillus and Acetobacter, novel genera were also metabolically active, including Microvirga, Inquilinus, Candolleomyces and Lasiodiplodia. The results showed a consistency in the main genera detected by both techniques, but the identification of unexplored genera such as Inquilinus, Microvirga, Cyphellophora and Ashbya gossypii, among others, suggests that this omics approach needs to be used together for more comprehensive results on spontaneous fermentation. In conclusion, studies combining techniques such as metabarcoding and metaproteomics should be considered in fermentation studies, as a single technique would result in omissions regarding the activity of certain microorganisms that may be important for the course of spontaneous fermentation. © 2024

Dr. Hine's research interests include: applied decision making risk perceptions resource management mining safety and adolescent risk behavior. He has published several articles on cognitive factors that mediate harvest behavior in common dilemmas risk perceptions related to the disposal of low-level and high-level nuclear waste and the relationship between the physical environment and substance misuse. CRAIG SUMMERS is an Associate Professor in Psychology at Laurentian University. He has published recent articles on risk perception and decision traps inPeace and Conflict Current Research on Peace and Violence Perception and Psycho- physics Cortex Behavioral Brain Science Ethics and Behavior andBehavior Research Methods Instruments and Computers. KATE TILLECZEK currently teaches “The Adolescent and Society” in the Department of Sociology at Laurentian University. Her research interests include healthy transitions of adolescents into adulthood including: school to work transitions adolescent career choice designing developmentally appropriate secondary schools student participation in educational change innovations such as school councils and understanding risk behaviors in social context. She has co-authored a policy document for the Premier's Council on Health Well-Being and Social Justice on “Transition Aged Youth” and a chapter in Learning and Sociological Profiles of Canadian High School Students on high school dropouts. JOHN LEWKO is the director of the Centre for Research in Human Development at Laurentian University and a Full Professor in the Child and Development Studies graduate program. He has expertise in adolescent risk behavior social policy and health promotion.

Cigarette smoking has been identified as the single most preventable source of mortality and morbidity in the United States. Experimentation with tobacco typically begins in adolescence, and smoking during this period is one of the strongest and most consistent predictors of adult smoking status. In the present paper, we review several prominent decision models that have been applied to adolescent smoking, and point out important limitations of these models. We then propose a new process-oriented framework of decision-making based on the concept of mental models, and discuss the implications of this framework for developing effective smoking prevention and cessation interventions for adolescents.

Background: Telomeres are protein complexes located at the top of each arm of chromosomes, essential for maintaining the stability of the genome. Telomere shortening is found in physiological aging and senescence-related to systemic lupus erythematosus (SLE). Objectives: To compare the relative telomere length (TL) in patients with SLE and controls and to correlate the TL with clinical characteristics, treatments, disease activity, and damage. Methods: A cross-sectional study comparing SLE patients and controls, from 18 to 60 years old, matched by sex and age. Clinical and demographic data from all participants were collected, and relative TL by qPCR. For cases, previous and current history, disease activity, damage, cognitive assessment, fatigue, presence of depression, anxiety, stress symptoms, and physical activity were analyzed. Results were obtained using frequencies and percentages for qualitative characteristics and measures of central tendency and standard deviation for quantitative ones. Comparisons of relative TL and other variables were performed using the t-Student test when assumptions of normality and homoscedasticity were satisfied and the Mann-Whitney test otherwise. A significance level of 5% was considered. Results: Sixty SLE patients and 55 controls were included. Among patients, 75% were diagnosed in adulthood; 86.7% were female (mean age 37 years); 15% had antiphospholipid syndrome (APS), 51.7% had a previous history of nephritis, and 23.3% had neuropsychiatric involvement. Among controls, 87.3% were female (mean age 38 years). There was no difference between groups regarding sex and age (p=0.924 and p=0.737, respectively). SLE patients had a median relative TL of 0.80 versus 1.07 in the control group (p=0.005). In the multiple regression model (R 2 =18.4%), patients with anxiety symptoms in a moderate grade had lower TL (p=0.015) when compared to patients without or with low-grade anxiety symptoms.
