The estrous cycle modulates hippocampal spine dynamics, dendritic processing, and spatial coding

Histological evidence suggests that the estrous cycle exerts a powerful influence on CA1 neurons in the mammalian hippocampus. Decades have passed since this landmark observation, yet how the estrous cycle shapes dendritic spine dynamics and hippocampal spatial coding in vivo remains a mystery. Here, we used a custom hippocampal microperiscope and two-photon calcium imaging to track CA1 pyramidal neurons in female mice across multiple cycles. Estrous cycle stage had a potent effect on spine dynamics, with spine density peaking during proestrus when estradiol levels are highest. These morphological changes coincided with greater somatodendritic coupling and increased infiltration of back-propagating action potentials into the apical dendrite. Finally, tracking CA1 response properties during navigation revealed greater place field stability during proestrus, evident at both the single-cell and population levels. These findings demonstrate that the estrous cycle drives large-scale structural and functional plasticity in hippocampal neurons essential for learning and memory. © 2025 The Author(s)

Background: Decades of steady improvements in life expectancy in Europe slowed down from around 2011, well before the COVID-19 pandemic, for reasons which remain disputed. We aimed to assess how changes in risk factors and cause-specific death rates in different European countries related to changes in life expectancy in those countries before and during the COVID-19 pandemic. Methods: We used data and methods from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to compare changes in life expectancy at birth, causes of death, and population exposure to risk factors in 16 European Economic Area countries (Austria, Belgium, Denmark, Finland, France, Germany, Greece, Iceland, Ireland, Italy, Luxembourg, the Netherlands, Norway, Portugal, Spain, and Sweden) and the four UK nations (England, Northern Ireland, Scotland, and Wales) for three time periods: 1990–2011, 2011–19, and 2019–21. Changes in life expectancy and causes of death were estimated with an established life expectancy cause-specific decomposition method, and compared with summary exposure values of risk factors for the major causes of death influencing life expectancy. Findings: All countries showed mean annual improvements in life expectancy in both 1990–2011 (overall mean 0·23 years [95% uncertainty interval [UI] 0·23 to 0·24]) and 2011–19 (overall mean 0·15 years [0·13 to 0·16]). The rate of improvement was lower in 2011–19 than in 1990–2011 in all countries except for Norway, where the mean annual increase in life expectancy rose from 0·21 years (95% UI 0·20 to 0·22) in 1990–2011 to 0·23 years (0·21 to 0·26) in 2011–19 (difference of 0·03 years). In other countries, the difference in mean annual improvement between these periods ranged from –0·01 years in Iceland (0·19 years [95% UI 0·16 to 0·21] vs 0·18 years [0·09 to 0·26]), to –0·18 years in England (0·25 years [0·24 to 0·25] vs 0·07 years [0·06 to 0·08]). Baccharis trimera aqueous extract modulates inflammation and nociception in mice

The aerial parts of Baccharis trimera (Less.) are frequently used as a tea to treat several diseases. Therefore, the aim of this study was to identify the constituents of an aqueous extract of B. trimera, focusing on their antioxidant, anti-inflammation, and antinociception activities and properties. For that, the researchers performed in vivo assays using the formalin test and Freund’s Complete Adjuvant (FCA) to measure the acute and chronic inflammatory pain in mice. Moreover, the myeloperoxidase enzyme (MPO) was analyzed in the subcutaneous tissue after the FCA injection, together with the counting of lymphocytes in the peripheral blood of the mice. The qualitative phytochemical analysis indicated the presence of flavonoids and saponins in the B. trimera aqueous extract. The high-performance liquid chromatography (HPLC) analyses showed the presence of phenolic compounds, such as chlorogenic acid, ellagic acid, rosmarinic acid, as well as flavonoids, such as rutin, quercetin, and luteolin. The DPPH assay was used in order to measure the antioxidant activity of the aqueous extract of B. trimera and this showed an IC50 of 118.18 ± 1.02 μg/mg. The data from the formalin test demonstrated that a single dose of the aqueous extract of B. trimera was not able to decrease the nociceptive behavior during the neurogenic phase, at any of the tested doses (20, 40, or 80 mg/kg p.o.). However, during the inflammatory phase of this test, the aqueous extract of B. trimera at 80 mg/kg (p.o.) significantly decreased the nociceptive behavior, showing more effectiveness when compared to the other tested doses (p < 0.05). Importantly, in the chronic inflammatory model on the 5th day of treatment, the aqueous extract of B. trimera (80 mg/kg p.o.) significantly reduced mechanical allodynia (p < 0.01), heat thermal hyperalgesia (p < 0.001), and paw edema (p < 0.05). This study aimed to analyze the factors associated with catheter-related bloodstream infection (CRBSI) in newborns undergoing infusion therapy using a peripherally inserted central catheter (PICC). A prospective cohort of 281 newborns who underwent installation of 483 PICCs in a neonatal intensive care unit (NICU) was followed from catheter insertion until 72 h after removal. Risk factors were assessed by analyzing the association between variables using the chi-square test and relative risk (RR) using the Poisson regression model. The significance level was 5%. Most of the patients were preterm infants with inadequate weight, respiratory disorders, and heart disease, but these variables were not associated with CRBSI. There was a significant association between CRBSI and PICC insertion in the upper limbs of newborns who received infusion therapy in the NICU (RR = 2.84;95% confidence interval 1.02–6.85). © 2022

Alzheimer's disease (AD) is a severely debilitating neurodegenerative disease with a rapidly increasing global prevalence, poorly understood causes, and no efficient treatments. Experimental models are valuable for studying AD pathogenesis, including amyloid beta and tau accumulation, synaptic dysfunction, and neuroinflammation. While no model fully reproduces the disease, we take an evolutionary biology approach to discuss available models across taxa, from mammals (rodents, primates) to zebrafish, Drosophila melanogaster , and Caenorhabditis elegans . Evaluating their strengths and limitations provides insight into disease mechanisms and may refine research strategies for improved diagnostics and therapeutic screening. Traditional models have significantly contributed to AD research, yet their translational limitations highlight the need for physiologically relevant alternatives. Integrating humanized rodent models, zebrafish, organoids, and induced pluripotent stem cell–based systems—along with advances in bioengineering and genetic editing—may offer a more comprehensive framework to bridge the gap between preclinical research and clinical application. Highlights Experimental models across rodents, primates, zebrafish, fruit flies, and worms provide key insights into Alzheimer's disease (AD). Cross-taxon comparisons assess strengths and weaknesses in AD models. Evolutionary biology approaches refine experimental strategies for AD research. Diverse animal models improve understanding of AD pathogenesis. Cross-species models enhance diagnostics and therapeutic strategy development.

Africa, the world's second-largest continent is home to 1.5 billion people, accounting for nearly 20% of the global population, (60% under age 25). By 2050, Africa's population will be 2.5 billion, and by 2035, more young Africans will be entering the workforce each year than in the rest of the world combined. Africa also hosts a rich social, cultural, and geopolitical diversity across its 5 geopolitical zones covering 54 countries. It is the most genetically, culturally, and linguistically diverse region on the planet. However, Africa's contribution to the global economy could be more significant if it urgently embraces the brain economy and leads in the development of new methodologies and approaches which can be exported around the world. In this paper, we explain our strategy to advance the Yaoundé Declaration for the Brain Economy, Brain health, and Brain Capital. The Declaration has been endorsed by Cameroon's President, His Excellency Paul Biya, and demonstrates African leadership in global brain and society innovations, laying out a roadmap for how Africa can outcompete other economies by deftly deploying brain science-inspired policies and investments. We outline a new economic approach for African jobs, economic growth, sustainability, resilience, health, and well-being. The brain economy offers a broader framework than the current sustainable development goals (SDG) agenda. The Yaoundé Declaration is trans-disciplinary and cross-cutting across sectors: 32 sitting members of government from different sectors having co-authored this paper. It aligns with many aspects of the United Nations Pact for the Future and can accelerate the SDG. © 2025

Despite being present in many drugs, guanylhydrazones and semicarbazones are two functional groups that have been little investigated as potential therapeutic strategies for the treatment of Alzheimer's disease (AD). For this reason, we initiated the synthesis and evaluation of these compounds as potential anticholinesterase agents, aiming to offer new alternatives for drug development against AD. In the severe phase of AD butyrylcholinesterase (BChE) becomes the main enzyme responsible for the hydrolysis of acetylcholine (ACh). Therefore, in this project, we present the results of BChE inhibitory activity, enzyme kinetics, cytotoxicity, and molecular modeling studies for three guanylhydrazone and two semicarbazone derivatives that were previously synthesized and evaluated as acetylcholinesterase (AChE) inhibitors. Among the compounds tested, guanylhydrazones (1, 2, and 3) showed inhibitory activity against BChE, exhibiting a mixed non-competitive inhibition profile. Specifically, compound 2 (phenanthrenequinone) demonstrated superior inhibitory potency with an IC₅₀ of 0.68 μM, compared to compound 1 (acridinone) with an IC₅₀ of 3.87 μM, and compound 3 (benzodioxole) with an IC₅₀ of 101.7 μM. In contrast, semicarbazones (4 and 5) showed no BChE inhibition up to the highest concentration tested (300 μM). Importantly, all five compounds were found to be non-cytotoxic. Our results suggest that these compounds have potential as drug prototypes targeting different phases of AD. Compounds 3, 4, and 5 may be more effective in the early phase, when AChE activity remains high;compound 1 could be useful in the intermediate phase;and compound 2 appears particularly promising for the severe phase, when BChE plays a more dominant role. © 2025 Elsevier B.V.

This study aims to develop an edible coating based on virgin coconut oil (VCO) nanoemulsion, iota-carrageenan and alginate to prolong the shelf life of pindang (mildly salted cooked) fish products. The research involved three stages: material preparation, characterization and determination of factors and responses for experimental design;parameter optimization to achieve optimal conditions;and application and analysis of pre- and post-coating characteristics of pindang fish. The parameter ranges were 0.5–1.5% for alginate concentration, 0.5–1.5% for iota-carrageenan concentration and 0.5–1.0% for VCO nanoemulsion concentration. The result of this study was an optimized food coating solution that was clear and bright white in color. Pindang fish coated with this layer showed improved quality in terms of water content (48.46%), weight loss (0.39%), pH (5.84), total volatile base (47.01%), salt content (2.9%), appearance (7), texture (7) and slime (7). At the same time, the total bacterial count remained unchanged (8 log colony-forming units (CFU)/g), the mold count remained unchanged (5 log CFU/g) and the aroma and taste received a score of 6. Thus, this study successfully produced an optimized edible coating that can enhance the quality and extend the shelf life of pindang fish, making it suitable for broader market distribution. © 2023 ICE Publishing.
