Assessment of Decontamination and Reuse of Disposable Filter Funnels Used in Microbiological Water Quality Tests

To monitor safely managed drinking water services, an increasing number of countries have integrated water quality testing for Escherichia coli into nationally-representative household surveys such as the Multiple Indicator Cluster Surveys (MICS). However, plastic waste generated during such water quality testing programs, mostly through the use of pre-sterilized disposable materials, is non-negligible. The objective of this study was to evaluate several re-use protocols for disposable filter funnels used by the MICS water quality test kits. Decontamination and re-use protocols were assessed in centralized laboratory and decentralized field settings and neither yielded positive results. Re-use of 100 mL sterile funnels decontaminated with an alcohol wipe resulted in a higher incidence of false positive results (i.e., positive contamination when processing sterile water), both in the laboratory and field;therefore, a higher proportion of positives tests can be expected if these components are re-used. Further improvements to the decontamination technique and training are needed before material re-use can be reliably adopted. Autoclaving the funnels for re-use is feasible, provided that there is capacity to re-package and distribute funnels in a sterile manner. © The Author(s) 2021.

The integration of methanotrophs and microalgae in coculture systems presents a promising approach for sustainable biogas valorization and single-cell protein (SCP) production, offering dual benefits of greenhouse gas mitigation and nutrient recovery from waste streams. However, the resilience and metabolic interplay of these consortia under ammonium stress, common in industrial wastewater, remain poorly understood, limiting their scalability. This study systematically investigated the performance of a microalgae-methanotroph consortium under stepwise ammonium concentrations (130, 200 and 260 mg NH 4 + -N/L). The system demonstrated remarkable acclimation, achieving stable biogas conversion with 95.8 ± 5.3 mg CO 2 -C/(L·day) and 109.0 ± 11.4 mg CH 4 -C/(L·day) even at highest ammonium concentration. The SCP content increased from 32 % to over 52 % of cell dry weight with yield peaking at 90 mg/(L·day) under 200 mg N/L. A microbial community shift from Methylosinus to ammonia-tolerant Methylococcus dominance underpinned functional stability. Metagenomic analyses revealed ammonium-driven metabolic adaptations: extracellular substance secretion reprograms under stress, nitrogen assimilation was enhanced via glutamine synthetase, and antioxidant defenses were activated. Network analysis highlighted intensified competition (31 % negative correlations) under stress, yet key synergies within coculture system sustained carbon and nitrogen metabolism. These findings resolve knowledge gaps in ammonium-stressed consortia dynamics and provide insights for engineering systems to advancing the circular bioeconomy.

Traditional building design education in the disciplines of building services, civil engineering, mechanical engineering, etc. focusses on quantifiable factors, i.e. factors which can be subject of calculation, simulation and dimensioning. Since non-quantifiable factors are not part of calculation or simulation models, there is a tendency that they are not adequately considered in education and, as a result, in building planning and operation. In the case of human thermal comfort, building professionals are required to understand the mechanisms and impact of non-quantifiable factors related to human thermal comfort and health, most of which are part of the adaptive thermal comfort concept. This paper identifies the challenges in teaching the concept of adaptive thermal comfort through a collection of lived experiences of the authors. Upcycling waste polyethylene terephthalate (PET) bottles has attracted intensive research interests. This simultaneously alleviates plastic pollution and achieves a waste-to-resource strategy. Waste PET water bottles were used to fabricate value-added activated carbon (AC) electrodes for capacitive deionization (CDI). The KOH activation temperature (greater than 700 °C) prominently affected the physi-chemical properties and desalination performance of PET-derived activated carbons (PET-AC). Profiting from a large Brunauer-Emmet-Teller specific surface area (1448 m g) with a good mesoporous structure (the ratio of the mesopore volume to the total pore volume was 41.3%), PET-AC-1000 (activated at 1000 °C) possessed a huge specific capacitance of 108 F g for capacitive ion storage. Moreover, when utilized as the electrode material in single-pass CDI, PET-AC-1000 exhibited a maximum electrosorption capacity of 10.82 ± 0.11 mg g and a low level of energy consumption (0.07 kWh mol), associated with good electrochemical charging-discharging cyclic stability. The results provide a promising facile approach to tackle the challenge of plastic pollution and promote the advancement of electrode materials for economic affordable and energy-efficient electrochemical desalination process, which meets the United Nations (UN) sustainable development goals (SDGs).

Adaptive thermal comfort guidelines have been developed within the work of Annex 69: "Strategy and practice of adaptive thermal comfort in low energy buildings". The guidelines have been established based on a framework for adopting adaptive thermal comfort principles in building design and operation developed by the authors. The guidelines target building practitioners, addressing the critical interrelated role building planners, building operators and occupants play. A successful adaptive thermal comfort design, in which design for human thermal adaptation is foreseen, planned, and carefully embedded in the design and operation intent, is based on broad knowledge and understanding of the multiple quantifiable and non-quantifiable factors influencing human perception, as well as human building interaction. Adaptive building design follows a user-centric integrated design approach and therefore it is critical to consider the occupants' and the operators' role in buildings already in the design phase. This paper focuses on three main challenges identified earlier and how these are addressed in the guidelines, i.e. i) updating prevailing knowledge about human thermophysiology and adaptation, ii) developing a procedure for design of adaptive opportunities, and iii) providing guidance for operational planning and operation of adaptive buildings. Urban parks provide vital social, environmental and health benefits to residents. However, the spatial distribution of parks, and the amenities they provide, may not be equitably distributed within cities. We examine the accessibility of urban parks with different social, environmental and health amenities by race and ethnicity. We identified 122,988 urban parks across the USA, measured the racial/ethnic population distribution within a 10-min walkshed around each park and compared these distributions to the overall demographics of the city. We found that the spatial distribution of parks as well as park amenities differ according to the neighborhood demographics. Racial/ethnic compositions of neighborhoods surrounding parks tend to be whiter than other parts of the same cities, though there are regional differences. Parks in predominantly white neighborhoods are cooler in the summer and have more tree cover compared with parks in neighborhoods with greater proportions of Hispanic and Black residents. Differences in amenities hold across regions of the country. Our study demonstrates that inequities in access to high-quality parks are widespread across the USA.

Optimum tank size for a rainwater harvesting system: Case study for Northern Cyprus

The available freshwater is limited on earth. On the other hand, available water resources on earth have been depleting and being polluted due to climate change and population growth. In order to reduce the risk of water scarcity and water resources contamination, Integrated water resources management (IWRM) is required. IWRM is a concept to manage water resources that aims to balance economic efficiency, social equity, and environmental sustainability. When rainwater harvesting systems (RWHS), one of the techniques of IWRM, are implemented, the stress on water resources is reduced. Since the installation cost of rainwater harvesting systems significantly depends on the size of the rainwater storage tanks, in the implementation of rainwater harvesting, the selection of tank size is one of the main concerns for the feasibility of the system. This study aims to investigate the feasibility of domestic rainwater harvesting systems for a single house. In order to find the optimum storage tank size of the rainwater harvesting system, a linear programming (LP) optimization model is employed. As a case study, the LP model is applied to six regions from semi-arid Eastern Mediterranean island Northern Cyprus, where water resources are limited. The model considers thirty-seven years monthly rainfall data, the roof area of the building, the water consumption per capita, the discount rate, the cost of the rainwater storage tank, and the number of residents. The results of the selected study areas show that the implementation of the RWHS for a single house is infeasible due to the substantial installation costs and maintenance expenses. The financial losses caused by the implementation of the RWHS are found higher than the installation costs and maintenance expenses for all regions. In addition to economic analyses, environmental benefits of the RWHS should be included into the feasibility analysis.
