Blastomere instance segmentation is important for analyzing embryos’ abnormality. To measure the accurate shapes and sizes of blastomeres, their amodal segmentation is necessary. Amodal instance segmentation aims to recover an object’s complete silhouette even when the object is not fully visible. For each detected object, previous methods directly regress the target mask from input features. However, images of an object under different amounts of occlusion should have the same amodal mask output, making it harder to train the regression model. To alleviate the problem, we propose to classify input features into intermediate shape codes and recover complete object shapes. First, we pre-train the Vector Quantized Variational Autoencoder (VQ-VAE) model to learn these discrete shape codes from ground truth amodal masks. Then, we incorporate the VQ-VAE model into the amodal instance segmentation pipeline with an additional refinement module. We also detect an occlusion map to integrate occlusion information with a backbone feature. As such, our network faithfully detects bounding boxes of amodal objects. On an internal embryo cell image benchmark, the proposed method outperforms previous state-of-the-art methods. To show generalizability, we show segmentation results on the public KINS natural image benchmark. Our method would enable accurate measurement of blastomeres in In Vitro Fertilization (IVF) clinics, potentially increasing the IVF success rate.

Dynamic axial focusing functionality has recently experienced widespread incorporation in microscopy,augmented/virtual reality(AR/VR),adaptive optics and material ***,the limitations of existing varifocal tools continue to beset the performance capabilities and operating overhead of the optical systems that mobilize such *** varifocal tools that are the least burdensome to operate(*** crystal,elastomeric or optofluidic lenses)suffer from low(≈100 Hz)refresh ***,the fastest devices sacrifice either critical capabilities such as their dwelling capacity(*** gradient lenses or monolithic micromechanical mirrors)or low operating overhead(*** mirrors).Here,we present a general-purpose random-access axial focusing device that bridges these previously conflicting features of high speed,dwelling capacity and lightweight drive by employing lowrigidity micromirrors that exploit the robustness of defocusing phase ***,the device consists of an 8.2mm diameter array of piston-motion and 48-μm-pitch micromirror pixels that provide 2πphase shifting for wavelengths shorter than 1100 nm with 10-90% settling in 64.8μs(i.e.,15.44 kHz refresh rate).The pixels are electrically partitioned into 32 rings for a driving scheme that enables phase-wrapped operation with circular symmetry and requires<30 V per *** experiments demonstrated the array’s wide focusing range with a measured ability to target 29 distinct resolvable depth ***,the features of the proposed array offer the potential for compact,straightforward methods of tackling bottlenecked applications,including high-throughput single-cell targeting in neurobiology and the delivery of dense 3D visual information in AR/VR.

Understanding how neural dynamics shape cognitive experiences remains a central challenge in neuroscience and psychiatry. Here, we present a novel framework leveraging state-to-output controllability from dynamical systems theory to model the interplay between cognitive perturbations, neural activity, and subjective experience. We demonstrate that large-scale fMRI signals are constrained to low-dimensional manifolds, where affective and cognitive states are naturally organized. Furthermore, we provide a theoretically robust method to estimate the controllability Gramian from steady-state neural responses, offering a direct measure of the energy required to steer cognitive outcomes. In five healthy participants viewing 2,185 emotionally evocative short videos, our analyses reveal a strong alignment between neural activations and affective ratings, with an average correlation of r ≈ 0.7. In a clinical cohort of 255 patients with major depressive disorder, biweekly Hamilton Rating Scale trajectories over 11 weeks significantly mapped onto these manifolds, explaining approximately 20% more variance than chance (p −10, numerically better than chance in 93% reaching statistical significance in one-third of subjects). Our work bridges dynamical systems theory and clinical neuroscience, providing a principled approach to optimize mental health treatments by targeting the most efficient neural pathways for cognitive change. © 2025, CC BY-NC-SA.

In the Philippines road accidents were prevalent among motorcycle riders. The motorcyclist was required to wear their helmet when on the road. Some motorcyclists didn't follow the rules on wearing a helmet as safety precautions on the road. To address this, policymakers are focusing on enforcing safe and law-abiding behavior in traffic. There is, however, a lack of comprehensive data on the safety-critical behavioral metric of the use of motorcycle helmets, especially in developing countries where the main mode of transport is the motorcycle. Targeted enforcement and safety campaigns that are critical for accident reduction are prohibited by this shortage of knowledge. Hence, The researchers developed an algorithm for detecting the helmet that was worn by the motorcyclist, by using a deep learning approach the object detection was done successfully. Based on the annotated images, frames of video, and live feed data that was collected, To detect active bikes, the researchers taught the algorithm to use their helmets. An overview of the algorithm's success on an annotated research data set and a study of available data on the use of human-registered helmets indicate that our approach is extremely reliable. © 2020 IEEE.

Isobaric labeling relative quantitation is one of the dominating proteomic quantitation technologies. Traditional quantitation pipelines for isobaric-labeled mass spectrometry data are based on sequence database searching. In this study, we present a novel quantitation pipeline that integrates sequence database searching, spectral library searching, and a feature-based peptide-spectrum-match (PSM) filter using various spectral features for filtering. The combined database and spectral library searching results in larger quantitation coverage, and the filter removes PSMs with larger quantitation errors, retaining those with higher quantitation accuracy. Quantitation results show that the proposed pipeline can improve the overall quantitation accuracy at the PSM and protein levels. To our knowledge, this is the first study that utilizes spectral library searching to improve isobaric labeling-based quantitation. For users to conveniently perform the proposed pipeline, we have implemented the feature-based filter being executable on both Windows and Linux platforms;its executable files, user manual, and sample data sets are freely available at https://***/comics/Software_*** . Furthermore, with the developed filter, the proposed pipeline is fully compatible with the Trans-Proteomic Pipeline. © 2023, Springer Nature Limited.

In this paper, an approach to aid in introductory programming courses is presented, focused on students that had never had any contact with computer programming. The approach, that was introduced in a previous paper [3], presents programming concepts using icons and “flowcharts”, and a 3D robotics simulator (CoppeliaSim), that has free educational use. This time, a real version of the simulated robot, built using Arduino and cheap, easily obtainable electronic components, is also presented. The programs to control the robot are developed by graphically constructing flowcharts on the visual editor, and then following the execution of programs using the simulated (or real) robot, step by step. This graphic program editor is named “FluxProg”, and its 2nd version is now available online, with new features comprising variables, arithmetic and logic expressions, thus providing a complete programming environment.

Neurotrophic receptor tyrosine kinase 3 (NTRK3) has pleiotropic functions: it acts not only as an oncogene in breast and gastric cancers but also as a dependence receptor in tumor suppressor genes in colon cancer and neuroblastomas. However, the role of NTRK3 in upper tract urothelial carcinoma (UTUC) is not well documented. This study investigated the association between NTRK3 expression and outcomes in UTUC patients and validated the results in tests on UTUC cell lines. A total of 118 UTUC cancer tissue samples were examined to evaluate the expression of NTRK3. Survival curves were generated using Kaplan–Meier estimates, and Cox regression models were used for investigating survival outcomes. Higher NTRK3 expression was correlated with worse progression-free survival, cancer-specific survival, and overall survival. Moreover, the results of an Ingenuity Pathway Analysis suggested that NTRK3 may interact with the PI3K-AKT-mTOR signaling pathway to promote cancer. NTRK3 downregulation in BFTC909 cells through shRNA reduced cellular migration, invasion, and activity in the AKT-mTOR pathway. Furthermore, the overexpression of NTRK3 in UM-UC-14 cells promoted AKT-mTOR pathway activity, cellular migration, and cell invasion. From these observations, we concluded that NTRK3 may contribute to aggressive behaviors in UTUC by facilitating cell migration and invasion through its interaction with the AKT-mTOR pathway and the expression of NTRK3 is a potential predictor of clinical outcomes in cases of UTUC. © 2024 The Authors. Phased arrays are crucial in various technologies, such as radar and wireless communications, due to their ability to precisely control and steer electromagnetic waves. This precise control improves signal processing and enhances imaging performance. However, extending phased arrays to the terahertz (THz) frequency range has proven challenging, especially for high-frequency operation, broadband performance, two-dimensional (2D) phase control with large antenna arrays, and strong phase modulation. Here, we introduce a photonic platform to realize a THz phased array that bypasses the above challenges. Our method employs 2D phase coding with 2-bit across a broad THz frequency range from 0.8 to 1.4 THz. The core of our design is a pixelated nonlinear Pancharatnam-Berry metasurface driven by a spatially modulated femtosecond laser, allowing precise phase control of THz signals. We showcase the effectiveness of our method through four proof-of-concept applications: single beamforming, dual beamforming, imaging and vortex beam generation. The realized photonic platform provides a promising pathway for developing broadband phased arrays in the THz regime. Copyright © 2024, The Authors. All rights reserved.

Coherent wave control exploits the interference among multiple waves impinging on a system to suppress or enhance outgoing signals based on their relative phase and amplitude. This process inherently requires the combination of non-Hermiticity, in order to enable energy exchanges among the waves, and spatial interfaces in order to tailor their scattering. Here we explore the temporal analogue of this phenomenon, based on time-interfaces that support instantaneous non-Hermitian scattering events for photons. Based on this mechanism, we demonstrate ultra-broadband temporal coherent wave control and the photonic analogue of mechanical collisions with phase-tunable elastic features. We apply them to erase, enhance and reshape arbitrary pulses by suitably tailoring the amplitude and phase of counterpropagating signals. Our findings provide a pathway to effectively sculpt broadband light with light without requiring spatial boundaries, within an ultrafast and low-energy platform. © 2022, CC BY.

Self-supervised approach for facial movement based optical flow

Computing optical flow is a fundamental problem in computer vision. However, deep learning-based optical flow techniques do not perform well for non-rigid movements such as those found in faces, primarily due to lack of the training data representing the fine facial motion. We hypothesize that learning optical flow on face motion data will improve the quality of predicted flow on faces. The aim of this work is threefold: (1) exploring self-supervised techniques to generate optical flow ground truth for face images;(2) computing baseline results on the effects of using face data to train Convolutional Neural Networks (CNN) for predicting optical flow;and (3) using the learned optical flow in micro-expression recognition to demonstrate its effectiveness. We generate optical flow ground truth using facial key-points in the BP4D-Spontaneous dataset. The generated optical flow is used to train the FlowNetS architecture to test its performance on the generated dataset. The performance of FlowNetS trained on face images surpassed that of other optical flow CNN architectures, demonstrating its usefulness. Our optical flow features are further compared with other methods using the STSTNet micro-expression classifier, and the results indicate that the optical flow obtained using this work has promising applications in facial expression analysis. © 2021, CC BY.
