Functional role of Annexin A1 and S100A11 in psoriasis pathogenesis: Insights from IMQ-induced models and keratinocyte modulation

Psoriasis (Pso) is a chronic inflammatory skin disease involving immune dysregulation and epidermal hyperplasia. Annexin A1 (ANXA1), a glucocorticoid-regulated protein, and S100A11 are implicated in keratinocyte function, but their roles in Pso remain unclear. This study translationally elucidates the functional role of ANXA1 and S100A11 in Pso by integrating analyses in human and murine skin biopsies and in vitro assays using human keratinocytes. The expression patterns of the ANXA1 and S100A11 in Pso were investigated using an imiquimod (IMQ)- induced murine model and human skin biopsies. Furthermore, the in vitro effect of the ANXA1-derived peptide Ac₂_–₂₆ (5, 25, and 50 ng/mL) on IL-17-stimulated human keratinocytes (HaCaT cell line) was evaluated. IMQ-treated mice exhibited hallmark psoriatic features, including epidermal thickening, mast cell degranulation, and elevated IL-23 levels. Immunohistochemical analyses revealed significant upregulation of ANXA1 and S100A11 in psoriatic skin, with coexpression observed in keratinocytes, particularly in superficial epidermal layers. Transcriptomic analysis corroborated these findings, showing elevated ANXA1 and S100A11 expression in psoriatic lesions compared to controls. In HaCaT cells, IL-17 stimulation reduced ANXA1 and S100A11 levels. Treatment with Ac₂_–₂₆ at the highest concentrations (25 and 50 ng/mL) significantly reduced viability/metabolism of IL-17-stimulated keratinocytes, while the lowest concentration of Ac₂_–₂₆ (5 ng/mL) effectively increased ROS production. In conclusion, ANXA1 and S100A11 are key players in Pso, with their expression and coexpression closely linked to disease pathogenesis. Further studies are warranted to explore the clinical implications of targeting the ANXA1/S100A11 axis in inflammatory skin diseases. © 2025

Objectives: To evaluate changes in the age at menarche in Asian populations. Study design: Retrospective cohort study. Methods: We included 548,830 women from six countries in Asia. The data were sourced from 20 cohorts participating in the Asia Cohort Consortium (ACC) and two additional cohort studies: Japan Multi-institutional Collaborative Cohorts (J-MICC), and Japan Nurse Health Study (JNHS) with data on age at menarche. Joinpoint regression was used to evaluate changes in age at menarche by birth year and by country. Results: The study includes data from cohorts in six Asian countries namely, China, Iran, Japan, Korea, Malaysia and Singapore. Birth cohorts ranged from 1873 to 1995. The mean age of menarche was 14.0 years with a standard deviation (SD) of 1.4 years, ranged from 12.6 to 15.5 years. Over 100 years age at menarche showed an overall decrease in all six countries. China showed a mixed pattern of decrease, increase, and subsequent decrease from 1926 to 1960. Iran and Malaysia experienced a sharp decline between about 1985 and 1990, with APC values of −4.48 and −1.24, respectively, while Japan, South Korea, and Singapore exhibited a nearly linear decline since the 1980s, notably with an APC of −3.41 in Singapore from 1993 to 1995. Conclusions: Overall, we observed a declining age at menarche, while the pace of the change differed by country. Additional long-term observation is needed to examine the contributing factors of differences in trend across Asian countries. The study could serve as a tool to strengthen global health campaigns. © 2024 The Royal Society for Public Health

Detection of spatiotemporal variation in ranavirus distribution using eDNA

Amphibian population declines have been associated with emerging diseases including ranaviruses, which can cause mass die-offs across entire amphibian communities. Understanding and mitigating disease spread requires knowledge of spatial and temporal patterns of pathogen distribution, but also how environmental factors influence pathogen occurrence. We applied environmental DNA (eDNA) detection tools to survey spatial and temporal distributions of ranaviruses by sampling 103 waterbodies in southeastern Ontario, Canada and assessed the role of abiotic factors as predictors of pathogen occurrence. Ten waterbodies sampled during June–August (>30 km between sites) revealed that ranavirus was marginally more prevalent (p =.055) during the latter part of the summer. Ninety-three sites sampled at a finer scale (<10 km between sites) exhibited seasonal variability in ranavirus detection (site prevalence: 56% May;66% July). Occupancy modeling revealed that wetland size and elevation influenced ranavirus occurrence while sampling date and water temperature influenced probability of detection. These findings indicate that biotic factors, such as host density and alternative hosts, should be investigated further as likely determinants of ranavirus prevalence across the landscape. Further, these results highlight the sensitivity of eDNA for detecting widespread presence of ranavirus and that abiotic factors may have a limited role in determining its prevalence and infectivity. © 2019 The Authors. This article explores the impact of a critical pedagogy of forensic science approach combined with an immersive field experience to train student members of a volunteer humanitarian forensic science team in preparation for immersion in the politically charged and emotionally challenging conditions surrounding migrant death at the United States-Mexico Border. Utilizing self-reflections from student team members before and after their fieldwork, the impact of a holistic and multi-faceted training approach can be evaluated. The goal of this training approach is to produce capable forensic scientists, as well as anthropologists who recognize the power structures inherent in such situations and subsequently work to kindle social change. Comparing expectations before fieldwork to experiences obtained during fieldwork can be a powerful way to gauge their progress in a transformative learning process.

This report describes the first documented case of intracerebral hemorrhage (ICH) in a horse following Bothrops snakebite envenomation. A 3-year-old mare developed acute kidney injury, systemic hemorrhage, and neurological signs before euthanasia. Postmortem findings confirmed massive ICH with intralesional venom identified by immunohistochemistry. Delayed treatment and insufficient antivenom likely contributed to the outcome. This case highlights the potential for cerebrovascular complications following Bothrops snakebite envenomation in horses, which may present as subtle neurological signs.

Fingerprints are unique to each individual who contributes to human identification in forensic cases. In this sense, the present work aimed to synthesize 1H-pyrazol-1-yl-4-phenylthiazoles and apply them as powder agents to reveal latent finger marks as an alternative and low cost related to powders found on the market. The compounds were characterized by infrared; fluorescence; 1H NMR; 13C NMR; laser diffractometer and SEM. In addition, cytotoxicity was also evaluated in oral keratinocytes showing high cell viability. The result of the hemolytic assay revealed that compound 2a does not present red cell cytotoxicity. The prepared powders were applied to fingermarks deposited on different surfaces providing quality visualization at level three so that human identification is possible.

Carbon acquisition and metabolism changes during fungal biotrophic plant pathogenesis: insights from Ustilago maydis (vol 39, pg 247, 2017)

Throughout the plant disease cycle, biotrophic fungal pathogens must obtain host-derived carbon molecules to act as building blocks and sources of energy. Gaining access to these resources requires biotrophic fungi to breach plant cell walls without eliciting substantial plant defences. The plant cell wall is composed mostly of glucose- and xylose-based polysaccharides, which can support fungal growth. Thus, fungi can acquire carbon compounds through the targeted depolymerization of specific wall components. When the plant cell wall is breached, biotrophs redirect photoassimilates, increase sink strengths, and express invertases and transporters to acquire carbon compounds. Transitions in enzyme and transporter expression during pathogenesis must be tightly controlled to ensure a continued supply of carbon compounds. This review describes carbon acquisition and metabolism, including regulation of available carbon source utilization mechanisms such as carbon catabolite repression. While carbon acquisition has been extensively studied in the ascomycetes, the mechanisms used by biotrophic fungi to acquire carbon during pathogenesis are poorly understood. Furthermore, the relationship between plant cell wall-degrading enzymes and carbon acquisition in biotrophic fungal pathogens is not well characterized. As such, this review summarizes the current knowledge of carbon source utilization by fungal pathogens, with an emphasis on research involving the corn smut pathogen Ustilago maydis, and provides a basis from which to extend our knowledge in this key area of fungal plant pathogenesis.
