Basophils are the rarest granulocytes circulating in the peripheral blood. They play critical roles in anti-parasite Th2-type immune responses and chronic allergic disorders. The developmental pathway for basophils has been recently demonstrated; myeloid progenitors pass through common myeloid progenitors, granulocyte-monocyte progenitors, granulocyte-committed progenitors (GPs), and basophil-committed progenitors (BaPs) in the bone marrow. BaPs then give rise to mature basophils. However, our understanding of how this pathway is regulated remains still elusive. Interferon Regulatory Factor-8 (IRF8), a hematopoietic cell-specific IRF transcription factor, is essential for the development of monocytes, dendritic cells, and eosinophils, while it inhibits neutrophil differentiation. Its role in the development of basophils has yet to be *** this study, we investigated whether IRF8 has any role in the development of the basophil lineage. We found that Irf8–/– mice displayed a severe reduction of basophil counts in the bone marrow, peripheral blood and spleen compared to wild-type (WT) mice. Irf8–/– mice retained GPs but lacked BaPs. Cell transfer experiments revealed that the defect of basophil development in Irf8–/– mice resides in bone marrow cells. We utilized IRF8-GFP chimera knock-in mice to examine IRF8 protein expression in the basophil lineage at a single cell level. We found that GPs, but not BaPs and mature basophils, expressed IRF8. Furthermore, purified Irf8–/– GPs failed to efficiently give rise to basophils in vitro. These results indicate that IRF8 acts at the stage of GPs in a cell-intrinsic manner. To understand the mechanism by which IRF8 promotes basophil development, we performed transcriptome analysis of purified GPs from WT and Irf8–/– mice by microarray. Interferon Regulatory Factor-8 (IRF8) is a transcription factor known to promote the development of monocytes/macrophages and dendritic cells while inhibiting that of neutrophils . In this report, we show that this transcription factor is required for the development of the basophil lineage . Irf8 −/− mice display a severe reduction of basophil counts in the bone marrow, peripheral blood and spleen. Bone marrow transfer experiments showed that IRF8 functions in a bone marrow cell-intrinsic manner. Analysis of bone marrow progenitors revealed that the number of granulocyte-committed progenitors (GPs) in Irf8 −/− mice is comparable to that of wild-type mice, while that of basophil-committed progenitors (BaPs) is diminished. Using IRF8-GFP knock-in mice, we found that GPs but not BaPs and mature basophils express IRF8, suggesting that IRF8 may act in the stage of GPs. Indeed, GPs purified from Irf8 −/− mice failed to efficiently give rise to basophils in vitro in the presence of IL-3. Taken together, these results uncover a previously unrecognized role of IRF8 in granulocyte development.

The effectiveness of exercise interventions for the management of frailty: A systematic review

This systematic review examines the effectiveness of current exercise interventions for the management of frailty. Eight electronic databases were searched for randomized controlled trials that identified their participants as frail either in the title, abstract, and/or text and included exercise as an independent component of the intervention. Three of the 47 included studies utilized a validated definition of frailty to categorize participants. Emerging evidence suggests that exercise has a positive impact on some physical determinants and on all functional ability outcomes reported in this systematic review. Exercise programs that optimize the health of frail older adults seem to be different from those recommended for healthy older adults. There was a paucity of evidence to characterize the most beneficial exercise program for this population. However, multicomponent training interventions, of long duration (≥5 months), performed three times per week, for 30-45 minutes per session, generally had superior outcomes than other exercise programs. In conclusion, structured exercise training seems to have a positive impact on frail older adults and may be used for the management of frailty. Copyright © 2011 Olga Theou et al.

He has published several articles on cognitive factors that mediate harvest behavior in common dilemmas risk perceptions related to the disposal of low-level and high-level nuclear waste and the relationship between the physical environment and substance misuse. CRAIG SUMMERS is an Associate Professor in Psychology at Laurentian University. He has published recent articles on risk perception and decision traps inPeace and Conflict Current Research on Peace and Violence Perception and Psycho- physics Cortex Behavioral Brain Science Ethics and Behavior andBehavior Research Methods Instruments and Computers. KATE TILLECZEK currently teaches “The Adolescent and Society” in the Department of Sociology at Laurentian University. Her research interests include healthy transitions of adolescents into adulthood including: school to work transitions adolescent career choice designing developmentally appropriate secondary schools student participation in educational change innovations such as school councils and understanding risk behaviors in social context. She has co-authored a policy document for the Premier's Council on Health Well-Being and Social Justice on “Transition Aged Youth” and a chapter in Learning and Sociological Profiles of Canadian High School Students on high school dropouts. JOHN LEWKO is the director of the Centre for Research in Human Development at Laurentian University and a Full Professor in the Child and Development Studies graduate program. He has expertise in adolescent risk behavior social policy and health promotion.

Cigarette smoking has been identified as the single most preventable source of mortality and morbidity in the United States. Experimentation with tobacco typically begins in adolescence, and smoking during this period is one of the strongest and most consistent predictors of adult smoking status. In the present paper, we review several prominent decision models that have been applied to adolescent smoking, and point out important limitations of these models. We then propose a new process-oriented framework of decision-making based on the concept of mental models, and discuss the implications of this framework for developing effective smoking prevention and cessation interventions for adolescents.

Physical activity offers one of the greatest opportunities for people to extend years of active independent life and reduce functional limitations. The purpose of this paper is to identify key practices for promoting physical activity in older adults, with a focus on older adults with chronic disease or low fitness and those with low levels of physical activity. Key practices identified in promotion activity in older adults include: (1) a multidimensional activity program that includes endurance, strength, balance, and flexibility training is optimal for health and functional benefits;(2) principles of behavior change, including social support, self-efficacy, active choices, health contracts, assurances of safety, and positive reinforcement, enhance adherence;(3) management of risk by beginning at low intensity but gradually increasing to moderate physical activity which has a better risk/benefit ratio should be the goal for older adults;(4) an emergency procedure plan is prudent for community-based programs;and (5) monitoring aerobic intensity is important for progression and for motivation. Selected content reviews of physical activity programming from major organizations and institutions are provided. Regular participation in physical activity is one of the most effective ways for older adults, including those with disabilities, to help prevent chronic disease, promote independence, and increase quality of life in old age. © EGREPA 2006.
