Recent advancements in the discovery of natural products have made significant progress in rapidly identifying known compounds from complex extracts using advanced spectroscopic technologies. Among these methods, one powerful approach called MS/MS molecular networking (MN) organizes chemical similarity based on MS/MS fragmentation data, serving as a valuable complement to traditional dereplication strategies. In our ongoing research focused on lead discovery from a sustainable medicinal source, an aquaculture soft coral Sarcophyton trocheliophorum, the MN profiling approach revealed the chemical diversity of biscembranoid analogs. By analyzing the MN pattern, biscembranoid derivatives with unique MS/MS fragments were prioritized for further investigation. Two novel biscembranoids, sarcotroxide A (1) and B (2), were successfully obtained through targeted isolation. These novel biscembranes contained an unprecedented 3-hydroperoxy-3-methylcyclohex-1-ene moiety compared with all biscembranoid analogs. The chemical structures of the two new biscembranoids were confirmed through spectroscopic analyses and in silico calculations. Their anti-inflammatory properties were assessed against the generation of superoxide anion (O₂^•−) and elastase release in activated human neutrophils. Sarcotroxides A (1) and B (2) exhibited anti-inflammatory activity by inhibiting O₂^•− accumulation (with 43.67% and 61.62% inhibition) and elastase release (with 38.21% and 61.62% inhibition) at 10 µM. These findings validated the potential of these biscembranoids from the cultured S. trocheliophorum as promising candidates for the future development of anti-inflammatory agents specifically designed to target neutrophils. © 2023 The Authors

Comparison of Neuromuscular Fatigue in Powerlifting Paralympics in Different Training Methods

Mean propulsive velocity (MPV) has been associated with neuromuscular fatigue; however, its suitability for strength training in Paralympic powerlifting (PP) remains uncertain. The objective of this work was to evaluate the MPV in two training methods (traditional-TRAD and eccentric-ECC). Eleven PP athletes were evaluated pre, during the intervention and post intervention at a load of 80% of the 1RM for TRAD and 110%-80% of 1 RM for ECC. The results demonstrated that there was no significant neuromuscular fatigue for the TRAD (~5% performance loss), as well as no significant decline in MPV during the intervention. For the ECC, there is a significant reduction in MPV before and after training (~12% loss of performance). A difference between TRAD and ECC after the intervention was also identified (0.87 m/s±0.22, 95% CI 0.72-1.02 vs. 0.72±0.20, 95% CI 0.59-0.86 p=0.042, F(3.30)=10.190, η2p=0.505 - very high effect). During the intervention for ECC, no significant decline in MPV was observed. The results of this study suggest that the mechanical indices of MPV do not seem to be effective indicators of neuromuscular fatigue in the sample studied or in the context of this specific training regime, being more indicated as a control of training volume.

Software is vital for the advancement of biology and medicine. Through analysis of usage and impact metrics of software, developers can help determine user and community engagement. These metrics can be used to justify additional funding, encourage additional use, and identify unanticipated use cases. Such analyses can help define improvement areas and assist with managing project resources. However, there are challenges associated with assessing usage and impact, many of which vary widely depending on the type of software being evaluated. These challenges involve issues of distorted, exaggerated, understated, or misleading metrics, as well as ethical and security concerns. More attention to the nuances, challenges, and considerations involved in capturing impact across the diverse spectrum of biological software is needed. Furthermore, some tools may be especially beneficial to a small audience, yet may not have comparatively compelling metrics of high usage. Although some principles are generally applicable, there is not a single perfect metric or approach to effectively evaluate a software tool's impact, as this depends on aspects unique to each tool, how it is used, and how one wishes to evaluate engagement. We propose more broadly applicable guidelines (such as infrastructure that supports the usage of software and the collection of metrics about usage), as well as strategies for various types of software and resources. We also highlight outstanding issues in the field regarding how communities measure or evaluate software impact. To gain a deeper understanding of the issues hindering software evaluations, as well as to determine what appears to be helpful, we performed a survey of participants involved with scientific software projects for the Informatics Technology for Cancer Research (ITCR) program funded by the National Cancer Institute (NCI). Type 2 diabetes (T2D) is a heterogeneous disease that develops through diverse pathophysiological processes and molecular mechanisms that are often specific to cell type. Here, to characterize the genetic contribution to these processes across ancestry groups, we aggregate genome-wide association study data from 2,535,601 individuals (39.7% not of European ancestry), including 428,452 cases of T2D. We identify 1,289 independent association signals at genome-wide significance (P < 5 × 10) that map to 611 loci, of which 145 loci are, to our knowledge, previously unreported. We define eight non-overlapping clusters of T2D signals that are characterized by distinct profiles of cardiometabolic trait associations. These clusters are differentially enriched for cell-type-specific regions of open chromatin, including pancreatic islets, adipocytes, endothelial cells and enteroendocrine cells. We build cluster-specific partitioned polygenic scores in a further 279,552 individuals of diverse ancestry, including 30,288 cases of T2D, and test their association with T2D-related vascular outcomes. Cluster-specific partitioned polygenic scores are associated with coronary artery disease, peripheral artery disease and end-stage diabetic nephropathy across ancestry groups, highlighting the importance of obesity-related processes in the development of vascular outcomes. Our findings show the value of integrating multi-ancestry genome-wide association study data with single-cell epigenomics to disentangle the aetiological heterogeneity that drives the development and progression of T2D. This might offer a route to optimize global access to genetically informed diabetes care.

Purpose: Demanding lifestyle characterized by extended working hours, shift work schedules as well as excessive use of mobile gadgets leads to the disruption of the circadian and homeostatic factors affecting the sleep quality of individuals. As consequence, subjects complain of suffering several sleep disorders some of them characterized by inducing excessive daytime sleepiness (EDS). Currently, the therapeutic approaches for managing EDS include medication, promotion of sleep hygiene, cognitive and behavioral therapy or using of continuous positive airway pressure machine. In this review, we propose the posology of the personalized sleep medicine by the prescription of naps for treating EDS. Methods: This review included the online search in PubMed and manual review of articles (basic and clinical trials) of a range of personalized medicine potentially associated to factors of dosage in areas such as nutrition, sports and sleep. Articles in English were identified and subsequently analyzed for consideration for this review. Results: Current evidence has demonstrated that naps exert positive outcomes for individuals complaining with EDS. The dosage of naps might follow similar procedures as reported for personalized interventions in diets or exercise programs (by taking the right dose, at the proper time, with a recommended frequency) which have demonstrated successfully results. Conclusions: The management of EDS may include the personalized sleep medicine considering the prescription of dosage of naps. © 2020, Springer Nature Singapore Pte Ltd.

In an elaborate form of inter-species exploitation, many insects hijack plant development to induce novel plant organs called galls that provide the insect with a source of nutrition and a temporary home. Galls result from dramatic reprogramming of plant cell biology driven by insect molecules, but the roles of specific insect molecules in gall development have not yet been determined. Here, we study the aphid Hormaphis cornu, which makes distinctive “cone” galls on leaves of witch hazel Hamamelis virginiana. We found that derived genetic variants in the aphid gene determinant of gall color (dgc) are associated with strong downregulation of dgc transcription in aphid salivary glands, upregulation in galls of seven genes involved in anthocyanin synthesis, and deposition of two red anthocyanins in galls. We hypothesize that aphids inject DGC protein into galls and that this results in differential expression of a small number of plant genes. dgc is a member of a large, diverse family of novel predicted secreted proteins characterized by a pair of widely spaced cysteine-tyrosine-cysteine (CYC) residues, which we named BICYCLE proteins. bicycle genes are most strongly expressed in the salivary glands specifically of galling aphid generations, suggesting that they may regulate many aspects of gall development. bicycle genes have experienced unusually frequent diversifying selection, consistent with their potential role controlling gall development in a molecular arms race between aphids and their host plants.

Background: The COVID-19 pandemic highlighted gaps in health surveillance systems, disease prevention, and treatment globally. Among the many factors that might have led to these gaps is the issue of the financing of national health systems, especially in low-income and middle-income countries (LMICs), as well as a robust global system for pandemic preparedness. We aimed to provide a comparative assessment of global health spending at the onset of the pandemic;characterise the amount of development assistance for pandemic preparedness and response disbursed in the first 2 years of the COVID-19 pandemic;and examine expectations for future health spending and put into context the expected need for investment in pandemic preparedness. Methods: In this analysis of global health spending between 1990 and 2021, and prediction from 2021 to 2026, we estimated four sources of health spending: development assistance for health (DAH), government spending, out-of-pocket spending, and prepaid private spending across 204 countries and territories. We used the Organisation for Economic Co-operation and development (OECD)'s Creditor Reporting System (CRS) and the WHO Global health Expenditure Database (GHED) to estimate spending. We estimated development assistance for general health, COVID-19 response, and pandemic preparedness and response using a keyword search. health spending estimates were combined with estimates of resources needed for pandemic prevention and preparedness to analyse future health spending patterns, relative to need. Findings: In 2019, at the onset of the COVID-19 pandemic, US$9·2 trillion (95% uncertainty interval [UI] 9·1–9·3) was spent on health worldwide. We found great disparities in the amount of resources devoted to health, with high-income countries spending $7·3 trillion (95% UI 7·2–7·4) in 2019;293·7 times the $24·8 billion (95% UI 24·3–25·3) spent by low-income countries in 2019.
