Overexpression of tyrosine phosphorylated proteins in reproductive tissues of polycystic ovary syndrome rats induced by letrozole

Objective:To identify the alteration of tyrosine phosphorylated protein expression in rats with polycystic ovary syndrome(PCOS).Methods:Sixteen female Sprague-Dawley rats were divided into the control and letrozole-induced PCOS *** oestrus cycle of rats was performed by vaginal *** hormones and morphology of the ovary,oviduct,and uterus were *** and intensity of androgen receptor and tyrosine phosphorylated proteins of reproductive organs were investigated by Western ***:Various polycysts and increased androgen receptor expression were present in the ovary of the PCOS *** levels of follicle-stimulating hormone and testosteone were significantly higher in the PCOS group while progesterone and estradiol levels were significantly decreased as compared with the control group(P<0.05).Only the size of uterus in the PCOS group was significantly smaller than the control ***,the density of collagen fibers observed in PCOS uterus was greater than the control ***,tyrosine phosphorylated proteins were significantly overexpressed in ovary(52,42,and 28 kDa),oviduct(72,56,42,and 28 kDa),and uterus(53 and 42 kDa)of the PCOS group compared to the control ***:Presence of tyrosine phosphorylated proteins in the ovary,oviduct and uterus suggests that overexpression of tyrosine phosphorylated proteins may be involved in potential mechanism of female infertility especially in PCOS.

Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Our findings provide insights into colorectal oncogenesis and highlight potential targets across tissues for new CRC treatment and chemoprevention strategies.

Dengue Virus (DENV) infection now has become a major public health threat, with nearly majority of population around the globe at high risk of infection. Effective treatments and drugs against DENV infection have not been established yet. With the emergence of the first licensed vaccine, Dengvaxia® (CYD-TDV), became the start to the development of several vaccine candidates. Hence, with the advent of recombinant DNA technology, DNA vaccines are such a promising approach compared to conventional vaccine methods. The recombinant multivalent protein envelope of DENV, known as rE, was constructed based on protein envelope domain III of four serotypes DENV and was reported structurally stable as well as potentially immunogenic. This research is conducted as an early stage in the development of DNA vaccine candidates against DENV, by transforming optimized rE gene mediated with plasmid vector pEGFP-C1 using a brief heat-shock treatment in E. coli DH5α as cloning host. Competent cells were produced using the combination of Magnesium Chloride and Calcium Chloride solution. The results showed growth signs of transformant colonies on a selective media containing kanamycin 100µg/ml, relatively. Colonies was confirmed carrying the target gene by colony polymerase chain reaction (PCR) screening. Observation on agarose gel under UV transIlluminator indicate bands that appeared in the product size of 157 base pairs.

Improper feeding methods can be linked to canine behaviour problems, as the conventional bowl-feeding approach fail to mimic natural behaviour and is associated with obesity. Feeding toys can potentially contribute to the enhancement of dog behaviour by mimicking natural hunting behaviour and promoting increased canine activity. Para rubber is a natural rubber with high flexibility and is tolerant of animal teeth, making it safer for animals compared to plastic and silicone. This study had two main objectives: firstly, to examine dogs' preferences for the scent and colour added to a para rubber product;and secondly, to explore the impact of using para rubber feeding toys on dog behaviours. The feeding toys were produced from natural rubber, with added scents and colours incorporated during their production. Subsequently, these toys were distributed to dog owners for evaluation. Owners completed the Thai version of the C-BARQ questionnaire, which evaluate canine behaviour both prior to and following a month of interaction with the toys. The findings indicated that dogs exhibited a preference for yellow toys over blue ones, as well as a preference for vanilla-scented toys compared to those with a beef scent. Following one month of consistent use of the para rubber toy, dogs tended to reduce various undesirable behaviours. Notably, overexcitement behaviour showed a nearly threefold decrease, and this reduction was statistically significant (p = 0.003). These outcomes emphasise the potential of natural rubber feeding toys in mitigating excitation-related behaviours, while also highlighting dogs' preference towards specific colours and scents. Consequently, there is a suggestion that enhancing toys with appealing scents or colours might foster greater interaction between dogs and their toys, potentially leading to a positive influence on their overall behavioural welfare. © 2023, The Authors. In 2020 SARS-CoV-2 reached pandemic status, reaching Brazil in mid-February. As of now, no specific drugs for treating the disease are available. In this work, the possibility of interaction between SARS-CoV-2 viral proteins (open and closed spike protein, isolate spike protein RBD, NSP 10, NSP 16, main protease, and RdRp polymerase) and multiple molecules is addressed through the repositioning of drugs available for the treatment of other diseases that are approved by the FDA and covered by SUS, the Brazilian Public health System. Three different docking software were used, followed by a unification of the results by independent evaluation. Afterwards, the chemical interactions of the compounds with the targets were inspected via molecular dynamics and analyzed. The results point to a potential effectiveness of Penciclovir, Ribavirin, and Zanamivir, from a set of 48 potential candidates. They may also be multi-target drugs, showing high affinity with more than one viral protein. Further in vitro and in vivo validation is required to assess the suitability of repositioning the proposed drugs for COVID-19. © 2021 The Authors

Recent advancements in the discovery of natural products have made significant progress in rapidly identifying known compounds from complex extracts using advanced spectroscopic technologies. Among these methods, one powerful approach called MS/MS molecular networking (MN) organizes chemical similarity based on MS/MS fragmentation data, serving as a valuable complement to traditional dereplication strategies. In our ongoing research focused on lead discovery from a sustainable medicinal source, an aquaculture soft coral Sarcophyton trocheliophorum, the MN profiling approach revealed the chemical diversity of biscembranoid analogs. By analyzing the MN pattern, biscembranoid derivatives with unique MS/MS fragments were prioritized for further investigation. Two novel biscembranoids, sarcotroxide A (1) and B (2), were successfully obtained through targeted isolation. These novel biscembranes contained an unprecedented 3-hydroperoxy-3-methylcyclohex-1-ene moiety compared with all biscembranoid analogs. The chemical structures of the two new biscembranoids were confirmed through spectroscopic analyses and in silico calculations. Their anti-inflammatory properties were assessed against the generation of superoxide anion (O₂^•−) and elastase release in activated human neutrophils. Sarcotroxides A (1) and B (2) exhibited anti-inflammatory activity by inhibiting O₂^•− accumulation (with 43.67% and 61.62% inhibition) and elastase release (with 38.21% and 61.62% inhibition) at 10 µM. These findings validated the potential of these biscembranoids from the cultured S. trocheliophorum as promising candidates for the future development of anti-inflammatory agents specifically designed to target neutrophils. © 2023 The Authors

The evaluation of an on-site monitoring program for activity meter quality assurance with exemption-level sources

This paper discusses the feasibility of a monitoring program for the quality assurance status of activity meters. We sent a questionnaire to clinical nuclear medicine departments of medical institutions, requesting information on their activity meters and quality assurance practices. On-site visits were conducted with exemption-level standard sources (Co-57, Cs-137 and Ba-133) for dose calibrators in nuclear medicine departments including physical inspection, accuracy and reproducibility. A method offering a quick check on the detection efficiency of the space dimension inside the activity meters was also introduced. For dose calibrator quality assurance, the daily checks had the highest implementation. However, annual checks and upon acceptance/after a repair check were reduced to 50% and 44%, respectively. The accuracy results of dose calibrators showed that all models exceeded the ±10% criteria with Co-57 and Cs-137 sources. The reproducibility results showed that some models exceeded the ±5% criteria with Co-57 and Cs-137 sources. The appropriate application of exemption-level standard sources considering the uncertainty that affects the measurement is discussed. © 2023 Society for Radiological Protection.
