Educational attainment (EduYears), a heritable trait often used as a proxy for cognitive ability, is associated with various health and social outcomes. Previous genome-wide association studies (GWASs) on EduYears have been focused on samples of European (EUR) genetic ancestries. Here we present the first large-scale GWAS of EduYears in people of East Asian (EAS) ancestry (n = 176,400) and conduct a cross-ancestry meta-analysis with EduYears GWAS in people of EUR ancestry (n = 766,345). EduYears showed a high genetic correlation and power-adjusted transferability ratio between EAS and EUR. We also found similar functional enrichment, gene expression enrichment and cross-trait genetic correlations between two populations. Cross-ancestry fine-mapping identified refined credible sets with a higher posterior inclusion probability than single population fine-mapping. Polygenic prediction analysis in four independent EAS and EUR cohorts demonstrated transferability between populations. Our study supports the need for further research on diverse ancestries to increase our understanding of the genetic basis of educational attainment.

Missense driver mutations in cancer are concentrated in a few hotspots1. Various mechanisms have been proposed to explain this skew, including biased mutational processes2, phenotypic differences3,4,5,6 and immunoediting of neoantigens7,8; however, to our knowledge, no existing model weighs the relative contribution of these features to tumour evolution. We propose a unified theoretical ‘free fitness’ framework that parsimoniously integrates multimodal genomic, epigenetic, transcriptomic and proteomic data into a biophysical model of the rate-limiting processes underlying the fitness advantage conferred on cancer cells by driver gene mutations. Focusing on TP53, the most mutated gene in cancer1, we present an inference of mutant p53 concentration and demonstrate that TP53 hotspot mutations optimally solve an evolutionary trade-off between oncogenic potential and neoantigen immunogenicity. Our model anticipates patient survival in The Cancer Genome Atlas and patients with lung cancer treated with immunotherapy as well as the age of tumour onset in germline carriers of TP53 variants. The predicted differential immunogenicity between hotspot mutations was validated experimentally in patients with cancer and in a unique large dataset of healthy individuals. Our data indicate that immune selective pressure on TP53 mutations has a smaller role in non-cancerous lesions than in tumours, suggesting that targeted immunotherapy may offer an early prophylactic opportunity for the former. Determining the relative contribution of immunogenicity and oncogenic function to the selective advantage of hotspot mutations thus has important implications for both precision immunotherapies and our understanding of tumour evolution.

Objective: Sarcopenia is a frequently observed comorbidity of rheumatoid arthritis (RA) due to the chronic activation of the innate immune system. Accumulating evidence has indicated that Chinese herbal medicine (CHM) safely suppresses proinflammatory pathways and controls inflammation-associated disease, but its effect in reducing the risk of developing sarcopenia among RA subjects has not been established. We conducted a population-level cohort study to compare the sarcopenia risk in patients with RA who use or do not use CHM. Methods: Using claims from a nationwide insurance database, we recruited patients with newly diagnosed RA and without sarcopenia between 2002 and 2010. Propensity score matching was applied to randomly select sets of CHM users and non-CHM users to compare the sarcopenia risk until the end of 2013. The risk of new-onset sarcopenia was assessed using the Cox proportional hazards model. Results: As compared to non-CHM users, those receiving CHM treatment had a lower incidence of sarcopenia (7.69 vs 9.83 per 1000 person-years). CHM was correlated with a decreased chance of sarcopenia after controlling for potential covariates. Notably, use of CHM for more than two years may diminish the risk of getting sarcopenia by about 47% when taken as prescribed. Prescriptions of several herbal formulae may benefit the reduction of sarcopenia risk, such as Yan-Hu-Suo, Bei-Mu, Da-Huang, Huang Qin, Ping-Wei-San (PWS), Shu-Jing-Huo-Xue-Tang (SJHXT) and Chuan-Xiong-Cha-Tiao-San (CXCTS). Conclusion: This study produced new evidence as it is the first to show that the longer duration of CHM use was correlated to reduced risk of sarcopenia in a dose-dependent manner, implying that CHM treatment could be embraced as a routine care strategy for preventing sarcopenia. © 2023 Li et al.

Streptococcus iniae (S. iniae) has become an important aquatic pathogen to cause serious streptococcosis outbreaks in cultured fish species, thus resulting in great economic losses in the worldwide aquaculture industry. Although vaccination has been so far an effective strategy to alleviate the threats of streptococcosis, antigenic variations displayed by different strains/isolates of S. iniae have brought about a severe challenge in the vaccine development. In the present study, three local S. iniae isolates, MML SI-007, MML SI-018 and MML SI-148, were first evaluated for their virulence in tilapia. Proteomics was then performed to analyze protein profiles between the highly virulent isolate (MML SI-007) and lowly virulent isolate (MML SI-018) in order to identify important virulence factors/antigens shared by different isolates. After proteomics, eleven proteins that displayed more than four times the differences in expression between MML SI-007 and MML SI-018 were identified. Two proteins, glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and superoxide dismutase (SOD), were selected to be vaccine candidates for respectively cloning their genes and expressing their recombinant proteins, rGAPDH and rSOD. After peritoneal vaccination in tilapia, mixed recombinant proteins (rGAPDH + rSOD) elicited significantly higher serum titers and lymphocyte proliferation than individual recombinant proteins (rGAPDH or rSOD). In addition, maximum production of IL-6 in the head kidney was detected in fish peritoneally vaccinated with mixed recombinant proteins. More importantly, 84% (21/25) of tilapia peritoneally vaccinated with mixed recombinant proteins survived at least four weeks after a lethal peritoneal challenge of heterologous S. iniae (MML SI-148). The association between working memory precision and the nonlinear dynamics of frontal and parieto-occipital EEG activity

Electrophysiological working memory (WM) research shows brain areas communicate via macroscopic oscillations across frequency bands, generating nonlinear amplitude modulation (AM) in the signal. Traditionally, AM is expressed as the coupling strength between the signal and a prespecified modulator at a lower frequency. Therefore, the idea of AM and coupling cannot be studied separately. In this study, 33 participants completed a color recall task while their brain activity was recorded through EEG. The AM of the EEG data was extracted using the Holo-Hilbert spectral analysis (HHSA), an adaptive method based on the Hilbert-Huang transforms. The results showed that WM load modulated parieto-occipital alpha/beta power suppression. Furthermore, individuals with higher frontal theta power and lower parieto-occipital alpha/beta power exhibited superior WM precision. In addition, the AM of parieto-occipital alpha/beta power predicted WM precision after presenting a target-defining probe array. The phase-amplitude coupling (PAC) between the frontal theta phase and parieto-occipital alpha/beta AM increased with WM load while processing incoming stimuli, but the PAC itself did not predict the subsequent recall performance. These results suggest frontal and parieto-occipital regions communicate through theta-alpha/beta PAC. However, the overall recall precision depends on the alpha/beta AM following the onset of the retro cue. © 2023, Springer Nature Limited.

Polylactic acid (PLA) ranks among the most extensively produced bioplastics, raising waste management concerns globally. This study explored dual pretreatment strategies, specifically photolysis by Ultraviolet-C (UVC) and hydrolysis by subcritical water extraction (SWE), for PLA waste treatment. UVC irradiation reduced the mechanical strength and viscosity average molecular weight (M v ) of PLA, but the effects were more significant in thin PLA films than in thick PLA beverage cups. Thus, the PLA cups were selected for SWE treatment, which reduced their M v from 190,000 to 5300. The pulverized SWE-treated PLA was later used for inducing protease and esterase in PLA-degrading bacterial consortium EAc. This active inoculum was applied as a bioaugmentation agent in a food composter for degrading UVC-treated PLA cups with food waste. PLA weight loss in the bioaugmented food composter (42 %) after 56 days was greater than the naturally attenuated composter (1 %). The residual PLA in the final bioaugmented compost had a relatively smaller size and lower M v with prominent surface erosion. In addition, the 16S rRNA gene amplicon sequencing revealed that the bioaugmentation promoted bacterial diversity and community interactions. Conclusively, PLA waste can be treated by UVC irradiation followed by composting with food waste using an active EAc inoculum.

Fish handling conditions on the market nowadays mainly in the traditional market is less a principle handling with caution, fast, and clean, making the selling fish polluted with chemical, physical, and microbiological pollutants. The fish handling on the modern market is generally better than the traditional markets. Puncak Permai and Simo Gunung Market provide fresh fish more than any other market in Surabaya, making these markets become the main destination for consumers, therefore this study was conducted to observe the fish freshness in both markets. The purpose of this study was to compare the freshness and observe the handling way of traders situating in both markets. This research used a survey method that was conducted for seven times sampling with three days of interval sampling. This study took three types of marine fish. The main parameters observed were pH and organoleptic values, while the supporting parameters were interviewed data. The result showed that the handling method had a significant effect (p0.05) on the pH level. © Published under licence by IOP Publishing Ltd.

Enteric pathogens and Carbapenem resistance genes are widespread in the fecal contaminated soils of cattle farms in the United States

Cattle farming is considered a major contributor to antibiotic resistant (AR) genes and bacteria in nearby environments, predominantly due to the large amount of manure released by cattle to soils. This study sought to compare soil within and outside of cattle pens from seven farms in the US. Soil microbial community differences are described, as well as differences in AR gene and enteric pathogen concentrations in the soils. Sources of fecal loading are quantified via microbial source tracking (MST) methods. The family Peptostreptococcaceae was more abundant in pen soils, and Rhodanobacteraceae was more abundant in non-pen soils. There were no differences in microbial community diversity between soils within and outside of cattle pens. The MST result showed that ruminant-specific fecal contamination (Rum2Bac) was abundant both in pen and non-pen soils, likely indicating heavy cattle fecal loading. However, the soil within cattle pens had higher levels of Campylobacter, Salmonella, and AR genes for resistance to sulfonamides (sul1) and carbapenems (KPC) than the soil outside of pens (p<0.05). This is the first study identifying KPC in livestock-associated soils in the US. This study supports increased hazard associated with livestock soils for animal and human health, as well as environmental quality. © 2021

BackgroundSystematic reviews (SR), an evidence-synthesis tool, have been increasingly used in toxicology, for regulatory decision-making and allocation of research funding resources. A critical domain of methodological quality is publicly available in the SR protocol that describes the review methods prior to the review. Earlier work showed that only a small number of published SRs in environmental health had identifiable protocols, but did not evaluate their completeness of reporting. Thus, there is a critical need to assess the number of protocols published in peer-reviewed literature related to environmental/occupational toxicology and how much those SR protocols adhere to current reporting standards such as *** overarching objective of this review is to assess the reporting quality of SR protocols in environmental and occupational toxicology, published in peer-reviewed *** and analysisThis protocol has been reported following PRISMA-P and PRISMA-S checklist and study selection, data extraction, and reporting quality assessment were piloted by independent reviewers. Four electronic databases (PubMed, Scopus, Web of Science, EMBASE) were selected and will be searched for SR protocols using pre-defined strings. Both title/abstract and full-text screening will follow detailed eligibility criteria based on the population concept context (PCC) framework. We will include peer-reviewed, self-identified SR protocols that aim to assess the adverse effects of environmental and occupational exposures. We will perform data extraction using the form that includes general bibliographic information, exposure, adverse effect information, evidence streams, and guidelines/checklists used in protocol reporting or development. A reporting assessment form consisting of 15 PRISMA-P-based items, of which eleven with identical binary responses (reported vs not reported) address critical elements of SR protocol.
