The COVID-19 pandemic is one of the unprecedented devastating catastrophes with severe public health threat globally. In Brazil, the use of Eucalyptus is focused on the production of wood or pulp for the paper industry but without any general recovery of waste, with leaves and branches being left on the ground. One possibility is to use these residues as raw materials in the production of industrially relevant and value-added compounds such as essential oil. The aim of the present study was to investigate the chemical composition, yield, anti-inflammatory/antinociceptive activities, and acute toxicity in mice, as well as the antimicrobial effects of essential oils from the leaves of 7 varieties of Eucalyptus and hybrids against Escherichia coli , Staphylococcus aureus, and Candida albicans . The extraction of oils was carried out using hydrodistillation, and they were analyzed by gas chromatography coupled to mass spectrometry. Urocam and Grancam were the plants that obtained the highest oil yield, with yields of 3.32 and 2.30%, respectively. The main chemical components identified in these plants were 1.8 cineole and α -pinene. The antinociceptive effect of the 7 oils (50 mg/kg, p.o.) was initially assessed in the acetic acid-induced writhing test. In this assay, a significant ( p < 0.05) antinociceptive/anti-inflammatory effect was observed from 4 tested essential oils ( E. benthamii, E. saligna, and the hybrids Urocam and Grancam) when compared to the vehicle-treated group. This effect was then confirmed in the formalin-induced paw licking test. No toxicological effects or alterations were observed in motor coordination after the administration of the studied oils to the animals. In the antimicrobial evaluation, the seven essential oils inhibited the growth of S. aureus, E. coli , and C. albicans at different concentrations. Collectively, these results demonstrate that the essential oil from the leaves and branches of Eucalyptus species and varieties present potential biomedical applications and represent a source of antimicrobial and/or anti-inflammatory compounds.

Feline infectious peritonitis (FIP) is a lethal, viral-induced immune-mediated disease that remains a challenge for diagnosis and treatment in cats. Proteomic profiling, which analyzes the protein content of biological samples, offers the potential to identify novel biomarkers that could improve the diagnosis and management of FIP. This study aims to assess the serum proteome and identify proteins that differentiate healthy cats from cats diagnosed with effusive FIP using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). A total of 30 cats diagnosed with effusive FIP and 27 clinically normal cats were enrolled. Twenty-three proteins were significantly (p < 0.01, ≥ fivefold change in abundance) differentially expressed between cats with effusive FIP and controls. Among these, the P2X purinoceptor, DNA topoisomerase, Notch receptor 2, and cadherin-17 were identified as key proteins of interest in cats with effusive FIP. Our findings suggest that these differentially expressed proteins could serve as potential diagnostic biomarkers and therapeutic targets for FIP. However, further studies are needed to validate these findings and explore their potential applications.

The interactions of charged particles moving paraxially in multi-walled carbon nanotubes may excite electromagnetic modes. This wake effect has recently been proposed as a potential novel method of short-wavelength high-gradient particle acceleration and for obtaining brilliant radiation sources. In this work, the excitation of wakefields in double-walled carbon nanotubes is studied by means of the linearized hydrodynamic theory. General expressions have been derived for the excited longitudinal and transverse wakefields and related to the resonant wavenumbers which can be obtained from the dispersion relation. In the absence of friction, the stopping power of the wakefield driver, modelled here as a charged macroparticle, can be written solely as a function of these resonant wavenumbers. The dependencies of the wakefields on the radii of the double-walled carbon nanotubes and the driving velocity have been studied. Double-walled carbon nanotubes with inter-wall distances much smaller than the internal radius may be a potential option to obtain higher wakefields for particle acceleration compared to single-walled carbon nanotubes. © 2024 The Authors

Cholera is a highly contagious and lethal waterborne disease induced by an infection with Vibrio cholerae (V. cholerae) secreting cholera toxin (CTx). Cholera toxin subunit B (CTxB) from the CTx specifically binds with monosialo-tetra-hexosyl-ganglioside (GM1) found on the exterior cell membrane of an enterocyte. Owing to the pathological process of CTx, a biomimetic electrochemical biosensor (CCB) was developed by applying GM1-expressing Caco-2 cell membrane (CCM) on the electrode surface of the biosensor for CTx detection. Briefly, the electrode surface was functionalized with CCM using the vesicle fusion method. The specific binding between GM1 and CTxB on the electrode surface was verified using confocal laser scanning microscopy and atomic force microscopy. The CTxB detection performances of the CCB, including sensitivity, selectivity, and long-term stability, were determined using electrochemical impedance spectroscopy (EIS). Based on the results, the CCB had an excellent limit of detection of ~11.46 nM and a detection range spanning 100 ng/mL - 1 mg/mL. Further, the CCB had high selectivity for the CTxB compared to different interfering molecules, which are abundant constituents of intestinal fluid and fecal matter in the human body. The long-term stability of the CCBs was also verified for 3 weeks using EIS. Overall, the CCB was found to have an excellent potential for application to point-of-care and cost-effective testing for CTxB detection in developing countries. © 2022, The Authors. The clinical engineer is the professional inside the hospitals that uses the knowledge of engineering in favor the resolutions of questions from the health environment. In addition to equipment management, clinical engineers have a number of diversified assignments and they need ensure the safety and satisfaction of human beings during them. One way to promote this is through hospital ergonomics, the observation of ergonomic parameters can help identify ergonomic problems that can be solved, promoting health, satisfaction and safety in the health units. Methods: The present study intended to establish a relationship between the attributions of clinical engineering and ergonomics, in order to demonstrate the ability of promoting ergonomics that clinical engineers possess, using bibliographic research as a method to provide information and data for the development of the research. Results: Comparing ergonomics parameters and clinical engineering attributions found in the literature it was possible to identify several convergences between them. Discussion: With the comparison, this relationship was perceptible, demonstrating the opportunity that clinical engineers have in ergonomics of offering better health services. © 2018 IEEE.

Soybean isoflavones have been linked to potential cardiovascular protective effects. This study investigated changes in isoflavone content during germination in five Taiwanese soybean cultivars. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was established to quantify 12 individual isoflavones within 15 min, using a Kinetex F5 column and Agilent 6470 mass spectrometer in MRM mode. The method showed good linearity (3–800 ng/mL, r² > 0.99), sensitivity (LLOQ = 3 ng/mL), and precision (RSD < 13.6 %). Total isoflavone concentrations increased progressively during the germination period and peaked on day 4. Malonyl-genistin was the dominant component, contributing 55–70 % of the total isoflavone content, and exhibited a 1.8- to 2.5-fold increase from day 1 to day 4. Glycoside forms also increased during germination, while aglycones and acetyl-glucosides remained relatively constant. These results indicate active isoflavone transformation during germination and suggest that day 4 may be a favorable time point for harvesting to maximize bioactive isoflavone yield. This analytical approach can support future studies on soybean sprout quality and processing optimization aimed at enhancing functional compound accumulation.

Diallyl trisulfide (DATS), an organosulfide compound derived from garlic, is renowned for its potent antioxidant properties, particularly in countering the generation of reactive oxygen species (ROS). It has also gained recognition as a potential agent for preventing heart-related conditions. Doxorubicin (Dox), a commonly used chemotherapeutic drug, is known to induce severe cardiac complications by promoting ROS production. Therefore, it was imperative to investigate whether DATS possesses cardioprotective capabilities against Dox-induced cardiac apoptosis and elucidate the underlying mechanisms. In this study, we observed that the intracellular ROS levels and cardiac apoptosis were heightened in H9c2 cells exposed to Dox (1 μM). However, treatment with 10 μM DATS effectively mitigated the Dox-induced ROS generation and apoptotic signaling, concurrently activating the PI3K/Akt pathway. Notably, the anti-apoptotic effects of DATS were attenuated when PI3K siRNA and the LY294002 PI3K inhibitor were employed. Furthermore, the TUNEL assay results demonstrated a significant reduction in Dox-induced apoptosis with DATS treatment. In summary, our findings indicate that DATS can activate the PI3K/Akt pathway, reducing ROS production in cardiac cells exposed to Dox, and subsequently rescue cardiac cells from apoptosis. © 2023 The Authors

Over 150 000 Americans are diagnosed with colorectal cancer (CRC) every year, and annually over 50 000 individuals will die from CRC, necessitating improvements in screening, prognostication, disease management, and therapeutic options. Tumor metastasis is the primary factor related to the risk of recurrence and mortality. Yet, screening for nodal and distant metastasis is costly, and invasive and incomplete resection may hamper adequate assessment. Signatures of the tumor-immune microenvironment (TIME) at the primary site can provide valuable insights into the aggressiveness of the tumor and the effectiveness of various treatment options. Spatially resolved transcriptomics technologies offer an unprecedented characterization of TIME through high multiplexing, yet their scope is constrained by cost. Meanwhile, it has long been suspected that histological, cytological, and macroarchitectural tissue characteristics correlate well with molecular information (e.g., gene expression). Thus, a method for predicting transcriptomics data through inference of RNA patterns from whole slide images (WSI) is a key step in studying metastasis at scale. In this work, we collected tissue from 4 stage-III (pT3) matched colorectal cancer patients for spatial transcriptomics profiling. The Visium spatial transcriptomics (ST) assay was used to measure transcript abundance for 17 943 genes at up to 5000 55-micron (i.e., 1–10 cells) spots per patient sampled in a honeycomb pattern, co-registered with hematoxylin and eosin (H&E) stained WSI. The Visium ST assay can measure expression at these spots through tissue permeabilization of mRNAs, which are captured through spatially (i.e., x–y positional coordinates) barcoded, gene specific oligo probes. WSI subimages were extracted around each co-registered Visium spot and were used to predict the expression at these spots using machine learning models.
