This paper presents a new deep regression model, which we call DeepDistance, for cell detection in images acquired with inverted microscopy. This model considers cell detection as a task of finding most probable locations that suggest cell centers in an image. It represents this main task with a regression task of learning an inner distance metric. However, different than the previously reported regression based methods, the DeepDistance model proposes to approach its learning as a multi-task regression problem where multiple tasks are learned by using shared feature representations. To this end, it defines a secondary metric, normalized outer distance, to represent a different aspect of the problem and proposes to define its learning as complementary to the main cell detection task. In order to learn these two complementary tasks more effectively, the DeepDistance model designs a fully convolutional network (FCN) with a shared encoder path and end-to-end trains this FCN to concurrently learn the tasks in parallel. DeepDistance uses the inner distances estimated by this FCN in a detection algorithm to locate individual cells in a given image. For further performance improvement on the main task, this paper also presents an extended version of the DeepDistance model. This extended model includes an auxiliary classification task and learns it in parallel to the two regression tasks by sharing feature representations with them. Our experiments on three different human cell lines reveal that the proposed multi-task learning models, the DeepDistance model and its extended version, successfully identify cell locations, even for the cell line that was not used in training, and improve the results of the previous deep learning methods. The microbiome, a complex micro-ecosystem, helps the host with various vital physiological processes. Alterations of the microbiome (dysbiosis) have been linked with several diseases, and generally, differential abundance testing between the healthy and patient groups is performed to identify important bacteria. However, providing a singular species of bacteria to an individual as treatment has not been as successful as fecal microbiota transplant therapy, where the entire microbiome of a healthy individual is transferred. These observations suggest that a combination of bacteria might be crucial for the beneficial effects. Here we provide the framework to utilize topic modeling, an unsupervised machine learning approach, to identify a community of bacteria related to health or disease. Specifically, we used our previously published gut microbiome data of patients with multiple sclerosis (MS), a neurodegenerative disease linked to a dysbiotic gut microbiome. We identified communities of bacteria associated with MS, including genera previously discovered, but also others that would have been overlooked by differential abundance testing. This method can be a useful tool for analyzing the microbiome, and it should be considered along with the commonly utilized differential abundance tests to better understand the role of the gut microbiome in health and disease.

BACKGROUND:Infection is a recognized risk factor for mortality among hemodialysis (HD) population, including infection caused by Enterobacteriaceae. We aimed to investigate Enterobacteriaceae in gut microbiota among HD patients and to analyze associations between microbiota and clinical parameters. METHODS:This prospective study of microbiota analysis in HD patients was conducted in April-May 2019. A control group without recent antibiotic use or hospitalization was used for comparison. Stool samples underwent 16S rRNA sequencing, using Greengenes 16S rRNA database for microbiota analysis. RESULTS:Among 96 hemodialysis (HD) patients, mean age was 61.9 ± 0.8 years and mean duration of HD was 6.5 ± 0.7 years. No significant differences were found in alpha diversity between HD and control groups (HD group 949.5, controls 898; p = 0.16) although significant between-group differences were found in beta diversity (p < 0.001). At phylum level, HD group had a higher abundance of Firmicutes and Proteobacteria, but lower abundance of Bacteriodetes. At genus level, Escherichia-Shigella complex increased among HD patients who had hospitalization with 1 year (median 0.024 vs 0.004, p = 0.054) and Klebsiella was associated with emergency room visit within 1 year among HD patients (p = 0.002). CONCLUSIONS:Alpha diversity in HD patients is not lower than that in healthy controls but significant between-group differences are found in microbiota composition according to beta diversity, due to decreased Bacteriodetes and increased Firmicutes and Proteobacteria. Deeper microbiota analyses for Enterobacteriaceae are necessary. Whether change in dietary components can help to decrease mortality among dialysis population warrants further research.

Technological developments in the current era make use of various media for learning. Learning media can facilitate and support practical activities to improve skills, one of which is by using video tutorial media because the video tutorials show the natural form of real objects to follow the instructions easily. This initial research aims to determine the need to develop video tutorial media used by teachers and students in the learning process to assemble computers at the Muhammadiyah 3 Yogyakarta vocational high school. This study used a descriptive-qualitative method by conducting interviews with one teacher and distributing needs analysis questionnaires to 30 students of class X TKJ as research subjects. The results obtained from the statement questionnaire given to students, 93% agreed to develop a video tutorial to facilitate understanding of the material to assemble a computer used for independent or group study wherever and whenever. This research can be used as a criterion for developing instructional media for video tutorials following the material's characteristics for assembling computers.

Background Apolipoprotein E4 (APOE4) has toxic effects on the cerebrovascular system, neuronal function, and functional connectivity of brain networks (e.g., default-mode, frontal-attention, and salience networks). The thalamus, a vital structure that acts as a relay station between many networks, has become of interest in AD research with studies showing atrophy among those with early AD and diminished cortical-thalamic connections among those with vascular impairment. Additionally, CSF soluble platelet-derived growth factor receptor-β (sPDGFRβ), a marker of blood-brain barrier breakdown, has been associated with AD risk, especially in APOE4 carriers. The present study investigated the relationship between CSF sPDGFRβ and functional connectivity in APOE4 carriers and non-carriers. Method One hundred sixty-six older adults from the Los Angeles area were selected from a larger cohort (96 females, mean age±SD = 67.4±10.7, 68 APOE4 carriers) based on availability of rsfMRI data. Image data was processed using CONN-toolbox v19c. Grey matter was parcellated using Harvard/Oxford brain atlas, excluding cerebellum. ROI-to-ROI analyses were conducted selecting the thalamus as seed region averaging mean connectivity between the left and right structure of other parcellated brain regions. Linear regression models evaluated significant associations between blood oxygen level-dependent (BOLD) and network connectivity among seed-target regions and CSF sPDGFRβvalues, while covarying for age and sex. Comparison of averaged functional connectivity values between seed and all target regions among APOE4 carriers and non-carriers was also examined. Result Higher functional connectivity between the thalamus and resting state network hub regions were associated with higher CSF sPDGFRβ (Fig 1A, 1C). An e-Commerce company has been using an Enterprise Resource Planning (ERP) system for several years, but is still constrained in its implementation, this is reflected in the number of issue/change request tickets submitted by users who have not been able to reach ideal conditions. To overcome this problem, we need an appropriate reference model, the ERP system user acceptance model in companies. The study conducted factors that influence the level of acceptance of SAP ERP users in the XYZ group to then determine a model that is able to explain practically the level of ERP system user acceptance, which in turn can produce concrete proposals to increase ERP system user acceptance. Research is quantitative descriptive using factor analysis and regression analysis methods. The research method uses a questionnaire with a total of 107 valid respondents coming from 8 company names under the XYZ group in 6 Southeast Asian countries. The conclusion of the study is that there are 24 indicators and 4 factors that influence user acceptance of the SAP ERP system. These four factors are the low contribution of the ERP system, the quality of ERP systems and data, the development of ERP systems, and also the low quality of ERP system supporters. It is hoped that by optimizing the value of the 4 factors, the XYZ group can increase user acceptance of the SAP ERP system so that the system can provide benefits expected by the company to be competitive advantage.

This full paper of research to practice category describes an experiment using an Intelligent Tutoring System to support teaching and learning that evolve solutions of first-degree equations. The difficulties encountered in solving algebra problems can make students consider math and related science a difficult task. We propose a Tutor to promote the learning of this class of problems. In this article, we describe the functioning and architecture of the tutor composed by the Interface, Student Model, Tutorial Strategies and Domain Model. Also, a Virtual Assistant was included to motivate students in the learning process. The tutoring strategies that comprise the tutor are projected from models found in the literature. To approximate the interactions of the tutor with the actions of the teachers, Lev Vygotsky's Concept of Zone Proximal Development was used. The methodology used in this project was the case study applied in a real-life context, obtaining a combination of quantitative and qualitative evidence about technical and pedagogical usability that show the degree of satisfaction of the students regarding the proposed approach.

Bank as a company that has many customers who conduct transactions every day, of course, has data that is increased continuously. In this paper, customer transaction data has been predicted to find potential customers in deposit offer. Data mining approach has been performed to classify potential customers for marketing through telemarketing. This data analysis can be used as a consideration in determining marketing strategy decisions for marketing managers. 15,713 data with 13 class attributes and 1 target class were obtained from UCI Machine Learning repository. The data was divided into 70% training data and 30% test data. Feedforward method of Artificial Neural Network was used to classify customer data. Multilayer Perceptron Neural Network and Radial Basis Function Neural Network were used to obtain optimal classification results. The result of this classification predicted potential customers to subscribe deposits. The result of this study indicated that the Radial Basis Function Neural Network method with 95.3% accuracy and 96.4% sensitivity was a better method compared to the Multilayer Perceptron Neural Network method with 88.0% accuracy and 99.4% sensitivity.
