Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Our findings provide insights into colorectal oncogenesis and highlight potential targets across tissues for new CRC treatment and chemoprevention strategies.

Metabolic flexibility refers to the ability of a tissue to adjust cellular fuel choice in response to conditional changes in metabolic demand and activity. A loss of metabolic flexibility is now recognized as a defining feature of various diseases and cellular dysfunction. In this study, using an inducible, skeletal muscle-specific knockout (KO) mouse, we found microRNA-1 (miR-1), the most abundant microRNA (miRNA) in skeletal muscle, was necessary to maintain whole-body metabolic flexibility. This was demonstrated by a loss of diurnal oscillations in whole-body respiratory exchange ratio and higher fasting blood glucose in miR-1 KO mice. Argonaute 2 enhanced crosslinking and immunoprecipitation sequencing (AGO2 eCLIP-seq) and RNA-seq analyses identified, for the first time, bona fide miR-1 target genes in adult skeletal muscle that regulated pyruvate metabolism. Comprehensive bioenergetic phenotyping combined with skeletal muscle proteomics and metabolomics showed that miR-1 was necessary to maintain metabolic flexibility by regulating pyruvate metabolism through mechanisms including the alternative splicing of pyruvate kinase (Pkm). The loss of metabolic flexibility in the miR-1 KO mouse was rescued by pharmacological inhibition of the miR-1 target, monocarboxylate transporter 4 (MCT4), which redirects glycolytic carbon flux toward oxidation. The maintenance of metabolic flexibility by miR-1 was necessary for sustained endurance activity in mice and in C. elegans. The physiological down-regulation of miR-1 in response to a hypertrophic stimulus in both humans and mice caused a similar metabolic reprogramming necessary for muscle cell growth. Taken together, these data identify a novel post-transcriptional mechanism of whole-body metabolism regulation mediated by a tissue-specific miRNA.

This Research Paper presents the evaluation of an instrument to identify the impact of motivation and engagement factors in undergraduate students in computing. Although researches indicate a direct impact of motivation and engagement on student performance and retention, few studies have been found that address which factors are relevant in this process. The instrument is a questionnaire based on the compilation of several works of the literature containing 48 items divided into 6 groups: personal and demographic data, general perception about motivation, perception about the university, student behavior, perception about program and perception about classes/teachers. The questionnaire evaluation is based on a case study with 112 undergraduate students in Software Engineering. As a result, we found that the questionnaire can be considered reliable (Cronbach's alpha = .8904). Considering the validity of constructs, we found an acceptable degree of correlation between the most pair of items in each group (averaging 63%). We also found that the item-total correlation coefficient was only not adequate for one factor group, indicating satisfactory correlation for all other items. Finally, we found that the number of factors is coherent, but there are several items from different groups strongly correlated, indicating the need for a reorganization.

The blazars B2 1811+31 and GB6 J1058+2817 were found to be in flaring state during 2020 and 2021, respectively. The high states of the sources were registered by the Fermi-LAT at energies below 100 GeV, triggering observations at higher energies with the MAGIC telescopes, in the UV/X rays with the Swift satellite and with ground-based radio and optical telescopes. Image magnification is one of the branches in digital image processing that is often required in various applications such as in the field of medicine, multimedia, and in satellite imagery. As technology grows, more and more methods are used for image enlargement. Deriving enhanced geographical representations via similarity-based spectral analysis: Predicting colorectal cancer survival curves in Iowa

Neural networks are capable of learning rich, nonlinear feature representations shown to be beneficial in many predictive tasks. In this work, we use such models to explore different geographical feature representations in the context of predicting colorectal cancer survival curves for patients in the state of Iowa, spanning the years 1989 to 2013. Specifically, we compare model performance using area between the curves (ABC) to assess (a) whether survival curves can be reasonably predicted for colorectal cancer patients in the state of Iowa, (b) whether geographical features improve predictive performance, (c) whether a simple binary representation, or a richer, spectral analysiselicited representation perform better, and (d) whether spectral analysis-based representations can be improved upon by leveraging geographically-descriptive features. In exploring (d), we devise a similarity-based spectral analysis procedure, which allows for the combination of geographically relational and geographically descriptive features. Our findings suggest that survival curves can be reasonably estimated on average, with predictive performance deviating at the five-year survival mark among all models. We also find that geographical features improve predictive performance, and that better performance is obtained using richer, spectral analysis-elicited features. Furthermore, we find that similaritybased spectral analysis-elicited representations improve upon the original spectral analysis results by approximately 40%. Copyright © 2018, The Authors. This Research to Practice Paper presents the results of the evaluation of pre-university factors that impact the initial motivation of undergraduate students in computing. Although there are studies in the literature that have investigated some previous factors, this paper replicates a previous work that aims to consolidate several pre-university factors and, as the main differential, uses the AMS (Academic Motivation Scale), a scale already consolidated in the literature to measure students' initial motivation, and evaluate the relation between motivation and candidate factors. We applied a questionnaire to 159 students from different computing programs in ten universities, which evaluates 20 factors divided into 4 groups: personal and demographic data, taste and knowledge of the program and area, computing experience, and school performance. To evaluate the correlation between factors and motivation, we used Spearman's coefficient, t-student test, and ANOVA to evaluate the correlation between factors and motivation. As main results, we found significant variation in the initial motivation according to following factors: taste for programming and technology, knowledge about the undergraduate program content, correct perception about computing professionals, knowledge and experience in computer programming, and general school performance.

College wrestling rankings have a major impact upon the sport, as they are one of the criteria used to determine advancement to postseason championships and seeding, thereby benchmarking individual, team, and program success. In addition to ordering athletes based upon past performance, rankings may function as predictors for future match outcomes. This research identifies potential biases in traditional ranking methodologies and offers alternatives based on the network-based PageRank and Elo ratings. Using data from the 2013-2014 NCAA Division I wrestling season, we evaluate both existing and our new rankings on the basis of predictive accuracy. Our new methods significantly improve upon a baseline of 67% accuracy and several of the existing ranking methods. These results and follow-up analyses suggest that Elo presents an especially attractive alternative to current ranking systems for college wrestling, with potential extensions to the prediction of point differentials, ensemble methods, and generalizability to other combat sports.

The launch of commercial off-the-shelf (COTS) software on the market requires careful planning, which incorporates a requirements prioritization process. Although there are numerous prioritization techniques, few satisfy quality criteria such as scalability, efficiency and ease of use. This research presents a survey of COTS software requirements prioritization to be potentially used by thousands of users. The HCV (Hierarchical Cumulative Voting) requirements prioritization technique was addressed and applied through a web questionnaire directly to potential users. At the end, 80 respondents sent the questionnaire and a score of requirements in ratio scale of measurement was obtained. It was found that the technique applied can respond positively to the needs and characteristics of Market-Driven Software Development, in similar situations to those described. © 2016 IEEE.
