Analysis of growth phase regulated KatA and CatE and their physiological roles in determining hydrogen peroxide resistance inAgrobacterium tumefaciens

Agrobacterium tumefaciens possesses two catalases, a bifunctional catalase-peroxidase, KatA and a homologue of a growth phase regulated monofunctional catalase, CatE. In stationary phase cultures and in cultures entering stationary phase, total catalase activity increased 2-fold while peroxidase activity declined. katA and catE were found to be independently regulated in a growth phase dependent manner. KatA levels were highest during exponential phase and declined as cells entered stationary phase, while CatE was detectable at early exponential phase and increased during stationary phase. Only small increases in H 2 O 2 resistance levels were detected as cells entering stationary phase. The katA mutant was more sensitive to H 2 O 2 than the parental strain during both exponential and stationary phase. Inactivation of catE alone did not significantly change the level of H 2 O 2 resistance. However, the katA catE double mutant was more sensitive to H 2 O 2 during both exponential and stationary phase than either of the single catalase mutants. The data indicated that KatA plays the primary role and CatE acts synergistically in protecting A. tumefaciens from H 2 O 2 toxicity during all phases of growth. Catalase-peroxidase activity (KatA) was required for full H 2 O 2 resistance. The expression patterns of the two catalases in A. tumefaciens reflect their physiological roles in the protection against H 2 O 2 toxicity, which are different from other bacteria.

Linkage between knowledge management and R&D management

Purpose - The relationship between knowledge management (KM) and research and development (R&D) management is intrinsically close. However, little attention has been paid to the development and implementation of KM systems for R&D organizations. The main objective of this current research is, from the perspective of evolutionary ecology, to suggest a framework of system design to link KM and R&D management. Design/methodology/approach - The chronological profile of R&D management systems over the past few decades is reviewed and the salient features of respective generations are identified. Then, the list of requirements for KM systems is suggested for each generation. Findings - Finds that these requirements constitute crucial variables for the design of KM systems and, as a whole, provide the building-blocks for integrating R&D management and KM. Originality/value - The proposed architecture is considered as a starting-point and serves as a milestone in implementing KMS in accordance with the R&D characteristics of a particular firm.

Effective Project Sponsorship an Evaluation of the Role of the Executive Sponsor in Complex Infrastructure Projects by Senior Project Managers

Project management literature frequently refers to the role of the project executive sponsor, but does not address in any great depth the factors contributing to effective project sponsorship, a role increasingly associated with project success. Most research about key project roles addresses either structural or behavioral factors with a definite emphasis on the former. This research attempts to bridge an emerging structural-behavioral schism using an approach based on Grounded Theory, involving in-depth interviews with senior project managers and project directors, all of whom were involved with the management of internal infrastructure projects. Project managers’ assessments of the effectiveness of the role of the executive sponsor are analyzed. The projects under scrutiny have been described by business owners as complex and of medium-to-high risk to the organizations concerned. Analysis of 28 interviews identifies key attributes of successful project sponsorship. Significantly, project managers appear to be exercising a complex range of behavior patterns to compensate for inadequate sponsor support in some projects. Important consequences arise for the organizations concerned if, in the process, inadequate sponsor performance is masked and opportunities for problem-focused conversations between project managers and sponsors are reduced or eliminated.

The process of writing in a new field is always challenging;however, developing a new paradigm and then communicating it to the international business community is a process that encompasses research into multiple fields and employs multiple tasks. Service Leadership is a new paradigm that incorporates a multidisciplinary approach to embedding empowered leadership within a service organization's culture. This paper gives an overview of the process of developing the paradigm itself and then proposing a process for its implementation within an existing corporate culture. © 2005 IEEE.

What makes some firms more innovative than others and what determines the source of these innovations are questions that are still not adequately answered due to the complex, often esoteric, nature of the innovation process. This paper considers the effect of one externally oriented strategy (extent of formal inter-organizational linkages) and one internally oriented strategy (degree of knowledge intensity) on overall levels of innovativeness and the source of these innovations. Using data collected from firms operating in the bioscience-technology industries in both New York and Utah, our results suggest that both of these strategies have a significant effect upon innovation levels and the source of innovation (internally versus externally stimulated). Interestingly, there were few firms that undertook both of these strategies simultaneously and thus the proposed interaction effect where increased absorptive capacity (on the basis of knowledge intensity) would make external linkages even more valuable could not be discerned.

The Engineering Games at UnicenP began in 1999 and, in 2004, the 6 th edition had the participation of approximately 600 students, freshmen and sophomore, from the Civil, Computer, Electric and Mechanical Engineering programs at Centro Universitário Positivo. In the last two editions of the games there has been a change in the format, with the students now being made responsible for the acquisition of the materials to be employed in the games. This is now possible due to advanced announcement of team composition, which gives the students enough time to contact fellow team members and get organized prior to Engineering Games day. The new format has helped improve team organization, and also made the games easier to set up, since more time can be devoted by the faculty to task creation and evaluation as opposed to the former assemblage of kits of materials to be handed to the teams. © 2005 IEEE.

AIRPET, an Asian regional air pollution research network, which is coordinated by the Asian Institute of technology, involves six cities: Bandung, Bangkok, Beijing, Chennai, Manila, and Hanoi. One of the objectives of the network is to provide a comprehensive assessment of particulate matter (PM) air quality though monitoring and receptor modeling tools jointly by six research teams in these cities. The results within the scope of this monitoring and modeling objective in phase 1 (2001-2003) were presented. In total, the network collected over 2500 PM2.5 and PM10 samples from characteristic urban sites. In all these cities the levels of PM10 and PM2.5 were found high especially during the dry season which the frequently exceeded the EPA standard for PM10 and PM2.5, especially at the traffic sites. Traffic had the highest PM2.5 contribution in most cities with the exceptional high share (> 70%) in Manila. Traffic and coal combustion had high contribution in Beijing while NaNO3 was found high in Hanoi. Knowledge management (KM) has become an important term in the construction industry. Knowledge management involves creating, securing, capturing, coordinating, combining, retrieving and distributing knowledge. The knowledge can be reused and shared among the involved engineers and experts to improve the construction process and reduce the time and cost of solving problems. Sharing and reusing knowledge depends on acquiring and preserving both tacit knowledge and explicit knowledge as the property of a corporation. This study addresses application of knowledge management in the construction phase of construction projects and proposes a construction map-based knowledge management (MBKM) concept and system for general contractors. This paper proposes a practical methodology to capture and represent construction project knowledge by using knowledge map. Using knowledge maps, users can get an overview of available and missing knowledge in core project areas and take appropriate management in tacit and explicit knowledge. Also, a portal system is developed to assist and present project-related knowledge by providing keyword and map search. The MBKM system is then applied in a case study of a construction precast office building in Taiwan to verify our proposed methodology and demonstrate the effectiveness of sharing knowledge special in the construction phase. By effectively using information and web technologies during the construction phase of a project, knowledge can be captured and managed to benefit future projects. The combined results demonstrate that, an MBKM-like system can be an effective tool for all experts and engineers participating for construction projects by utilizing the knowledge management concept and web technology.

We review the evidence from a number of surveys in which our NetLab has been involved about the extent to which the Internet is transforming or enhancing community. The studies show that the Internet is used for connectivity locally as well as globally, although the nature of its use varies in different countries. Internet use is adding on to other forms of communication, rather than replacing them. Internet use is reinforcing the pre-existing turn to societies in the developed world that are organized around networked individualism rather than group or local solidarities. The result has important implications for civic involvement.
