Design and Synthesis of Nanoparticle Contrast Agents for Spectral (color) X-Ray Imaging

Computed tomography (CT) is an important tool in clinical diagnostic imaging enabling three-dimensional anatomic imaging at high spatial resolution with short scan times. However, X-ray attenuation differences in physiological fluids and soft tissues are relatively small, requiring the use of contrast agents to achieve sufficient imaging contrast. Recent advances in energy-sensitive X-ray detectors have made spectral (color) CT commercially feasible by unmixing the energy-dependent attenuation profile of different materials and will potentially enable molecular imaging in CT. In order to leverage these capabilities for diagnostic imaging, we are developing a spectral library of nanoparticle contrast agents with K-shell absorption edges spaced at least 10 keV apart. The objective of this study was to demonstrate the ability of spectral CT to simultaneously detect up to three different contrast agents and unmixed their signals to create color images. Gadolinium oxide (Gd), hafnium oxide (Hf) and gold (Au) were chosen due to exhibiting K-edges spaced 10-20 keV apart. Core-shell nanoparticles of each composition were synthesized by various methods to have a core diameter of 15-20 nm and were coated with a silica shell at least 2-4 nm in thickness to create a common platform for surface functionalization. The contrast agents were imaged in a soft tissue equivalent phantom using source-side method for spectral CT imaging. The source-side approach utilized monochromatic synchrotron radiation at the Argonne National Laboratory which, while not clinically applicable, served as a gold standard due to providing the highest spectral resolution. The nanoparticles designed for this study have broad applications in biomedical imaging due to their modular assembly, potential for enabling multi-modal detection, and surface functionalization with biomolecules (e.g., antibodies, peptides or enzymes) for active targeting.

A pilot scale bioreactor using EMMC for carbon and nitrogen removal

A pilot scale (100 l reactor) of an entrapped mixed microbial cell (EMMC) process was fabricated and tested for simultaneous removal of carbon and nitrogen. Process performance, operational stability, and maintenance requirements were all determined. Two sources of actual agricultural processing wastewater containing a high concentration of chemical oxygen demand (COD) (about 800–1,000 mg/l) and domestic sewage containing a low concentration of COD (about 150–200 mg/l) were investigated in this study. Various HRT (hydraulic retention time) and aeration schedules were operated. It was found that soluble COD (SCOD) and soluble total nitrogen (STN) could be removed in the range of 40–70% and 20–90%, respectively, for domestic sewage depending on the operational conditions provided. For agricultural processing wastewater, removal efficiencies of SCOD and STN are 89–91% and 60–75%, respectively, depending on the HRTs and aeration schedules applied. Economic evaluation for the application of domestic sewage was conducted. It was found that at an HRT of 6 h with 24 h of aeration it costs U.S.$1.75 for the treatment of 1,000 gal/day (3.8 m3/day). It is apparent that the EMMC process is technically feasible for simultaneous removal of carbon and nitrogen under the operation of an alternated schedule of the aeration in one single bioreactor. Ultimately, it can replace or upgrade the existing conventional wastewater treatment plant by combining the secondary and tertiary wastewater treatment plant in one bioreactor and provides simple maintenance and operation. This will also assist in providing the high quality of treated effluent meeting current and future environmental regulation for reuse.

Background: High-resolution estimates of HIV burden across space and time provide an important tool for tracking and monitoring the progress of prevention and control efforts and assist with improving the precision and efficiency of targeting efforts. We aimed to assess HIV incidence and HIV mortality for all second-level administrative units across sub-Saharan Africa. Methods: In this modelling study, we developed a framework that used the geographically specific HIV prevalence data collected in seroprevalence surveys and antenatal care clinics to train a model that estimates HIV incidence and mortality among individuals aged 15–49 years. We used a model-based geostatistical framework to estimate HIV prevalence at the second administrative level in 44 countries in sub-Saharan Africa for 2000–18 and sought data on the number of individuals on antiretroviral therapy (ART) by second-level administrative unit. We then modified the Estimation and Projection Package (EPP) to use these HIV prevalence and treatment estimates to estimate HIV incidence and mortality by second-level administrative unit. Findings: The estimates suggest substantial variation in HIV incidence and mortality rates both between and within countries in sub-Saharan Africa, with 15 countries having a ten-times or greater difference in estimated HIV incidence between the second-level administrative units with the lowest and highest estimated incidence levels. Across all 44 countries in 2018, HIV incidence ranged from 2·8 (95% uncertainty interval 2·1–3·8) in Mauritania to 1585·9 (1369·4–1824·8) cases per 100 000 people in Lesotho and HIV mortality ranged from 0·8 (0·7–0·9) in Mauritania to 676·5 (513·6–888·0) deaths per 100 000 people in Lesotho.
