The decision to determine whether or not a journal to be published in Majalah Kedokteran Nusantara can be decided by using the Decision Support system with certain methods. One of methods used is fuzzy logic method and is applied to the Microsoft Visual Basic programming language. From the results of the research, the first step that must be done in determining a journal is published was to identify the problem and to determine how to evaluate the quality of the journal which is done by calculating the parameter values of the variables that have been determined by the editorial board. The parameters used in the calculation were title of articles, abstract, background, content, conclusions and bibliography. The conclusions obtained from the research is to building a decision support system to determine a journal published in Majalah Kedokteran Nusantara using fuzzy logic method was not so difficult to do, because it utilized Fis Sugeno Matlab application and by using fuzzy logic methods and utilizing fuzzyfication to get linguistic variables were very helpful in determining how a journal is published. © Published under licence by IOP Publishing Ltd.

Favipiravir (T-705) has been developed as a potent anti-influenza drug and exhibited a strong inhibition effect against a broad spectrum of RNA viruses. Its active form, ribofuranosyl-triphosphate (T-705-RTP), functions as a competitive substrate for the RNA-dependent RNA polymerase (RdRp) of the influenza A virus (IAV). However, the exact inhibitory mechanisms of T-705 remain elusive and subject to a long-standing debate. Although T-705 has been proposed to inhibit transcription by acting as a chain terminator, it is also paradoxically suggested to be a mutagen towards IAV RdRp by inducing mutations due to its ambiguous base pairing of C and U. Here, we combined biochemical assay with molecular dynamics (MD) simulations to elucidate the molecular mechanism underlying the inhibitory functions exerted by T-705 in IAV RdRp. Our in vitro transcription assay illustrated that IAV RdRp could recognize T-705 as a purine analogue and incorporate it into the nascent RNA strand. Incorporating a single T-705 is incapable of inhibiting transcription as extra natural nucleotides can be progressively added. However, when two consecutive T-705 are incorporated, viral transcription is completely terminated. MD simulations reveal that the sequential appearance of two T-705 in the nascent strand destabilizes the active site and disrupts the base stacking of the nascent RNA. Altogether, our results provide a plausible explanation for the inhibitory roles of T-705 targeting IAV RdRp by integrating the computational and experimental methods. Our study also offers a comprehensive platform to investigate the inhibition effect of antivirals and a novel explanation for the designing of anti-flu drugs. © 2021

SARS-CoV-2 infections display tremendous interindividual variability, ranging from asymptomatic infections to life-threatening disease. Inborn errors of, and autoantibodies directed against, type I interferons (IFNs) account for about 20% of critical COVID-19 cases among SARS-CoV-2-infected individuals. By contrast, the genetic and immunological determinants of resistance to infection per se remain unknown. Following the discovery that autosomal recessive deficiency in the DARC chemokine receptor confers resistance to Plasmodium vivax, autosomal recessive deficiencies of chemokine receptor 5 (CCR5) and the enzyme FUT2 were shown to underlie resistance to HIV-1 and noroviruses, respectively. Along the same lines, we propose a strategy for identifying, recruiting, and genetically analyzing individuals who are naturally resistant to SARS-CoV-2 infection.

Lithium is regarded as the first-line treatment for bipolar disorder (BD), a severe and disabling mental health disorder that affects about 1% of the population worldwide. Nevertheless, lithium is not consistently effective, with only 30% of patients showing a favorable response to treatment. To provide personalized treatment options for bipolar patients, it is essential to identify prediction biomarkers such as polygenic scores. In this study, we developed a polygenic score for lithium treatment response (Li) in patients with BD. To gain further insights into lithium's possible molecular mechanism of action, we performed a genome-wide gene-based analysis. Using polygenic score modeling, via methods incorporating Bayesian regression and continuous shrinkage priors, Li was developed in the International Consortium of Lithium Genetics cohort (ConLiGen: N = 2367) and replicated in the combined PsyCourse (N = 89) and BipoLife (N = 102) studies. The associations of Li and lithium treatment response - defined in a continuous ALDA scale and a categorical outcome (good response vs. poor response) were tested using regression models, each adjusted for the covariates: age, sex, and the first four genetic principal components. Statistical significance was determined at P < 0.05. Li was positively associated with lithium treatment response in the ConLiGen cohort, in both the categorical (P = 9.8 × 10, R = 1.9%) and continuous (P = 6.4 × 10, R = 2.6%) outcomes. Compared to bipolar patients in the 1 decile of the risk distribution, individuals in the 10 decile had 3.47-fold (95%CI: 2.22-5.47) higher odds of responding favorably to lithium. The results were replicated in the independent cohorts for the categorical treatment outcome (P = 3.9 × 10, R = 0.9%), but not for the continuous outcome (P = 0.13). Salivary uric acid is regarded as a noninvasive biomarker of metabolic syndrome and renal disorder. Although many enzyme based assays have been developed for quantitation of uric acids, most of the biological reagents are only stable for 30 days after opening even stored at 2-8 °C. To overcome this short-lasting problem, long-lasting (more than two years), easily stored and cost-effective nonenzymatic C-dots prepared from PEI through a simple hydrothermal method were developed. Experimental results reveal that the sensitivity of the PEI C-dot assay for uric acid is higher than that of reported fluorescece and enzyme based sensors. Background Sedentary behaviors (ST) are linked to children's mental health, including internalizing and externalizing problems. Research on these associations in children with overweight/obesity is limited. To this end, we examined cross-sectional and longitudinal associations between sedentary time (ST) and behavioral problems in children with overweight/obesity, followed by an investigation of potential sex-related differences. Methods We included 2273 children with overweight/obesity (49.9 % boys) from the UK Millennium Cohort Study. Accelerometer-measured sedentary time (ST) and MVPA were collected at age 7, and behavioral problems were assessed at ages 7 and 11 via the Strengths and Difficulties Questionnaire (SDQ). General linear models were used to examine how ST at 7 years of age was cross-sectionally and longitudinally associated with externalizing and internalizing, while ethnicity, SDQ scores, parental psychological distress, highest parental education, household poverty, and moderate-to-vigorous physical activity (MVPA) were statistically controlled. These analyses are followed by an analysis investigating sex-related differences. Results In the cross-sectional analyses, more ST was associated with fewer externalizing problems at 7 years among boys (β: −0.008, 95 % CI: −0.013, −0.003) and girls (β: −0.006, 95 % CI: −0.011, −0.001) without controlling for MVPA. More ST at 7 years was associated with fewer externalizing problems (β: −0.010, 95 %CI: −0.015, −0.004) only among boys when adjusting for MVPA. In the longitudinal analysis, more ST at 7 years was associated with fewer externalizing problems at 11 years for boys (β: −0.006, 95 %CI: −0.010, −0.001) and girls (β: −0.007, 95 %CI: −0.012, −0.002) without controlling for MVPA. When adjusting for MVPA at 7 years, more ST at 7 years was associated with fewer externalizing problems at 11 years for girls (β: −0.006, 95 %CI: −0.013, 0.000). In any organization, the appropriate use of information systems will have an impact on the effectiveness of day-to-day operations. To find out how the performance of an information system is necessary to do an evaluation of the implementation of which has been run. This study evaluates the implementation of information systems in the Maternity Hospital St. Yosef. By adopting PIECES framework as an evaluation, primary data collection is done through questionnaires distributed. Of the some respondents who collected the data used factor analysis to get factors and indicators affect the successful implementation of system and get right model for evaluation of the Maternity Hospital in St. Joseph. The results showed that the factor of ease use and suitability to needs of a positive effect on the success, while reliability of system negatively affect the success. On a scale of 0-100 obtained an un-expected value of system performance is at 45.75 and the expected value of performance of the system is equal to 83.77.
