Over the past few years, different computer-aided diagnosis (CAD) systems have been proposed to tackle skin lesion analysis. Most of these systems work only for dermoscopy images since there is a strong lack of public clinical images archive available to design them. To fill this gap, we release a skin lesion benchmark composed of clinical images collected from smartphone devices and a set of patient clinical data containing up to 22 features. The dataset consists of 1,373 patients, 1,641 skin lesions, and 2,298 images for six different diagnostics: three skin diseases and three skin cancers. In total, 58.4% of the skin lesions are biopsy-proven, including 100% of the skin cancers. By releasing this benchmark, we aim to aid future research and the development of new tools to assist clinicians to detect skin cancer. Copyright © 2020, The Authors. All rights reserved.

Background Epidermal growth factor receptor (EGFR) is upregulated in prostate cancer (PCa). However, suppression of EGFR did not improve the patient outcome, possibly due to the activation of PI3K/Akt signaling in PCa. Compounds able to suppress both PI3K/Akt and EGFR signaling may be effective for treating advanced PCa. Purpose We examined if caffeic acid phenethyl ester (CAPE) simultaneously suppresses the EGFR and Akt signaling, migration and tumor growth in PCa cells. Methods Wound healing assay, transwell migration assay and xenograft mice model were used to determine the effects of CAPE on migration and proliferation of PCa cells. Western blot, immunoprecipitation, and immunohistochemistry staining were performed to determine the effects of CAPE on EGFR and Akt signaling. Results CAPE treatment decreased the gene expression of HRAS, RAF1, AKT2, GSK3A , and EGF and the protein expression of phospho-EGFR (Y845, Y1069, Y1148, Y1173), phospho-FAK, Akt, and ERK1/2 in PCa cells. CAPE treatment inhibited the EGF-induced migration of PCa cells. Combined treatment of CAPE with EGFR inhibitor gefitinib showed additive inhibition on migration and proliferation of PCa cells. Injection of CAPE (15 mg/kg/3 days) for 14 days suppressed the tumor growth of prostate xenografts in nude mice as well as suppressed the levels of Ki67, phospho-EGFR Y845, MMP-9, phospho-Akt S473, phospho-Akt T308, Ras, and Raf-1 in prostate xenografts. Conclusions Our study suggested that CAPE can simultaneously suppress the EGFR and Akt signaling in PCa cells and is a potential therapeutic agent for advanced PCa.

Spoof-Fluid-Spoof Acoustic Waveguide and its Applications for Sound Manipulation

We explore the implementation of a spoof–fluid-spoof (SFS) acoustic waveguide, which is the acoustic analogue of a metal-insulator–metal waveguide in plasmonics. The proposed structure consists of two corrugated rigid surfaces separated by a fluid layer. The properties of the guided acoustic modes between corrugated rigid parallel plates are analytically and numerically studied by deriving their dispersion relation and through a careful analysis of symmetric and antisymmetric modes supported by this structure. Similar to plasmonic waveguides, it is shown that the dispersion relation of the proposed SFS acoustic waveguide, and the supported group and phase velocities, can be largely controlled by changes in the geometrical parameters of the waveguide, opening the possibility for application of this technology for acoustic delay lines, modulators, and sensors. The analytical and numerical results are validated through experiments in a controlled environment.

HIV-specific CD8 + T-cells partially control viral replication and delay disease progression, but rarely provide lasting protection – largely due to immune escape. We show that engrafting mice with memory CD4+ T-cells from HIV + donors uniquely allows for the in vivo evaluation of autologous T-cell responses, while avoiding graft-versus-host disease and the need for human fetal tissues that limit other models. Treating HIV-infected mice with clinically-relevant HIV-specific T-cell products resulted in substantial reductions in viremia, which were augmented by engineering with IL-15 superagonist-containing nanogels. Ultimately, in vivo activity was limited by the selection of diverse escape mutations, recapitulating patterns seen in humans. By applying mathematical modeling, we show that the kinetics of the CD8 + T-cell response have a profound impact on the emergence and persistence of escape mutations. This ‘participant-derived xenograft’ model of HIV provides a powerful tool for studying HIV-specific immunological responses and facilitating the development of effective cell-based therapies. © 2020, The Authors. Currently, the growth of information and technology is rapid. It makes a lot of things in various fields including education becoming more effective and efficient. In education, one of its implementation which is a game is proven to be a useful tool to support conventional teaching methods and bring a more natural understanding of learning materials. Moreover, as a product of the popular culture in modern society, video game mirrors the general culture practice in real life and reflects it via its own culture inside the game. This makes a video game can give a contribution to the social construction of reality as it affects the player's view towards learning in real life and vice versa. From there, we see that there is an opportunity for learning about religion to be supported by its utilization to provide an interactive and fun learning experience. In this paper, we discuss how a video game is implemented to support religious learning. The game was developed with the scrum method where we surveyed to gather the user requirements before the design step. The game design was made by using the use case diagram and storyboard, and it was built using Unity version 2017.3.0f3.

In this paper, a new optical technique, two movable slit plate modules and the subjective optometry method was developed to examine Interpupillary distance. The evaluation of binocular vision are dependent on the three-dimensional structure of natural scenes. The eye position and the fusion image of the eyes are fine-tuned and the alignment is assured by through the vision of subject with the trial frame and a movable slit plate module after full optometry correction procedure. This novel method is to success and replace of new optical lenses, the error of prismatic imbalance estimated has an accuracy of 0.12Δ. It also changes the refraction test from an objective method to a subjective method in order to meet the subject's most comfortable binocular visual fusion.

Eight previously unreported glycosides, designated gomphandranosides I–VIII, along with nine known compounds, were isolated from the roots of Gomphandra mollis Merr. Their structures were elucidated through comprehensive spectroscopic analyses, including NMR, HR-ESI-MS, CD, DP4+ probability, and ECD calculations. The anti-inflammatory activities of the isolated compounds were assessed using nitric oxide inhibition assays in LPS-stimulated RAW264.7 macrophages. Compounds 5 , 10 , and 16 exhibited significant anti-inflammatory activity with IC 50 values of 7.56 ± 0.9, 15.0 ± 1.2, and 14.2 ± 0.9 μM, respectively. Network pharmacology, gene ontology, Kyoto Encyclopedia of Genes and Genomes analysis, and molecular docking studies identified IL-6, TNF-α, MMP9, PTGS2, and HIF1A as key targets, with potentially involved in the NF- κ B and IL-17 signaling pathways. These findings highlight G. mollis as a promising candidate for the treatment of inflammation-related diseases.

The reproducibility crisis in bioscience, characterized by inconsistent study results, impedes our understanding of biological processes. Global collaborative studies offer a unique solution to this problem. Here, we present a global collaboration using the zebrafish (Danio rerio) novel tank test, a popular behavioral assay for anxiety-like responses. We analyzed data from 20 laboratories worldwide, focusing on housing conditions and experimental setups. Our study included 488 adult zebrafish, tested for 5 min, focusing on a variety of variables. Key findings show that female zebrafish exhibit more anxiety-like behavior than males, highlighting sex as a critical variable. Housing conditions, including higher stocking densities and specific feed types, also influenced anxiety levels. Optimal conditions (5 fish/L) and nutritionally rich feeds (for example, rotifers) mitigated anxiety-like behaviors. Environmental stressors, such as noise and transportation, significantly impacted behavior. We recommend standardizing testing protocols to account for sex differences, optimal stocking densities, nutritionally rich feeds and minimizing stressors to improve the reliability of zebrafish behavioral studies.
