Quantum tunneling phenomenon allows a high-barrier reaction to occur even though thermal energy is not sufficient to surmount the barrier. The phenomenon has been suggested to exist in the isomerization of methylhydroxycarbene which occured at 11 K. Here we study the quantum tunneling that occured in the isomerization of methylhydroxycarbene to acetaldehyde and to vinyl alcohol based on density functional calculations. The isomerization pathways are determined under the effect of various solvents which are modeled by polarizable continuum model (PCM). We use Wentzel-Kramers-Brillouin (WKB) approximation to calculate the tunneling probability (T) and extend the usefulness of T into isomerization rate. The results show that the solvents give a significant effect on the isomerization rate of methylhydroxycarbene to acetaldehyde but an insignificant one on the isomerization rate to vinyl alcohol.

Background: health promotion for the elderly mainly targets improvements in physical function, and a lack of strengthening of subjective well-being as well as the creation of interaction-providing opportunities for activity and participation has been reported. Evaluations of positive and negative aspects for subjective well-being are needed. Elderly subjects were intervened through a 6-month complex-type program combining amusement and exercise and its influences on subjective well-being and physical and physiological functions were investigated. Methods: Subjects in this survey were 37 healthy elderly females living in M city, Hiroshima Prefecture (age: 69.7 ± 5.9 years old), and the survey was performed between September 2016 and March 2017. The complex-type program comprised amusement and exercise programs. The survey was performed at 4 time points during the intervention period: before the intervention and 1, 3, and 6 months during the intervention. Subjective well-being (Japanese version of the Subjective Well-being Inventory (SUBI): Mental health scores, Mental fatigue scores), salivary amylase levels, and the locomotive syndrome risk test (two-step test, stand-up test, and 25-question risk assessment) were surveyed at each time point. Regarding the duration of the intervention as a factor, changes in salivary amylase levels were analyzed using repeated measures one-way ANOVA and SUBI was tested using the Friedman test (EZR Ver 1.32). In addition, the relationship between the locomotive syndrome risk level and duration of the intervention was examined using Fisher’s test. Results: SUBI and the locomotive syndrome risk level significantly differed with time during the intervention. Slight changes were observed in salivary amylase levels during the intervention. Standardising the representation of biomedical knowledge among all researchers is an insurmountable task, hindering the effectiveness of many computational methods. To facilitate harmonisation and interoperability despite this fundamental challenge, we propose to standardise the framework of knowledge graph creation instead. We implement this standardisation in BioCypher, a FAIR (findable, accessible, interoperable, reusable) framework to transparently build biomedical knowledge graphs while preserving provenances of the source data. Mapping the knowledge onto biomedical ontologies helps to balance the needs for harmonisation, human and machine readability, and ease of use and accessibility to nonspecialist researchers. We demonstrate the usefulness of this framework on a variety of use cases, from maintenance of task-specific knowledge stores, to interoperability between biomedical domains, to on-demand building of task-specific knowledge graphs for federated learning. The emergence of SARS-CoV in 2002 and SARS-CoV-2 in 2019 led to increased sampling of sarbecoviruses circulating in horseshoe bats. Employing phylogenetic inference while accounting for recombination of bat sarbecoviruses, we find that the closest-inferred bat virus ancestors of SARS-CoV and SARS-CoV-2 existed less than a decade prior to their emergence in humans. Phylogeographic analyses show bat sarbecoviruses traveled at rates approximating their horseshoe bat hosts and circulated in Asia for millennia. We find that the direct ancestors of SARS-CoV and SARS-CoV-2 are unlikely to have reached their respective sites of emergence via dispersal in the bat reservoir alone, supporting interactions with intermediate hosts through wildlife trade playing a role in zoonotic spillover. These results can guide future sampling efforts and demonstrate that viral genomic regions extremely closely related to SARS-CoV and SARS-CoV-2 were circulating in horseshoe bats, confirming their importance as the reservoir species for SARS viruses. © 2025 The Authors

Objectives: We sought to model chronic radiation-associated dysphagia (RAD) in patients given intensity-modulated radiation therapy (IMRT) for oropharyngeal squamous cell cancer (OPSCC) as a function of age and dose to non-target swallowing muscles. Methods: We reviewed 300 patients with T1-T4 N0-3 M0 OPSCC given definitive IMRT with concurrent chemotherapy. Chronic RAD was defined as aspiration or stricture on videoflouroscopy/endoscopy, gastrostomy tube, or aspiration pneumonia at >= 12 months after IMRT. Doses to autosegmented regions of interest (ROIs;inferior, middle and superior constrictors, anterior and posterior digastrics, mylo/geniohyoid complex, intrinsic tongue, and gengioglossus) were obtained from DICOM-RT plans and dose-volume histograms. The probability of chronic RAD as a function of mean ROI dose, stratified by age (<50, 50-59, 60-69, or >70 years), was estimated with logistic probability models and subsequent unsupervised nonlinear curves. Results: Chronic RAD was observed in 34 patients (11%). Age was a significant correlate of chronic RAD, both independently and with dose for all muscle groups examined. Distinct muscle-specific dose-response profiles were observed as a function of age (e.g., 5% of patients in their 50 s [but 20% of those 70 +] who received 60 Gy to the superior constrictor had chronic RAD). This effect was stable across all observed muscle ROIs, with a false discovery rate-corrected p < 0.05, for all dose/muscle/age models, suggesting that including age as a covariate improves modeling of chronic RAD. Conclusions: Age at treatment moderates the probability of chronic RAD after chemo-IMRT for OPSCC, with aging muscles showing lower dose thresholds.
