Background: The sustainable development goals (SDGs) aim to end HIV/AIDS as a public health threat by 2030. Understanding the current state of the HIV epidemic and its change over time is essential to this effort. This study assesses the current sex-specific HIV burden in 204 countries and territories and measures progress in the control of the epidemic. Methods: To estimate age-specific and sex-specific trends in 48 of 204 countries, we extended the Estimation and Projection Package Age-Sex Model to also implement the spectrum paediatric model. We used this model in cases where age and sex specific HIV-seroprevalence surveys and antenatal care-clinic sentinel surveillance data were available. For the remaining 156 of 204 locations, we developed a cohort-incidence bias adjustment to derive incidence as a function of cause-of-death data from vital registration systems. The incidence was input to a custom Spectrum model. To assess progress, we measured the percentage change in incident cases and deaths between 2010 and 2019 (threshold >75% decline), the ratio of incident cases to number of people living with HIV (incidence-to-prevalence ratio threshold <0·03), and the ratio of incident cases to deaths (incidence-to-mortality ratio threshold <1·0). Findings: In 2019, there were 36·8 million (95% uncertainty interval [UI] 35·1–38·9) people living with HIV worldwide. There were 0·84 males (95% UI 0·78–0·91) per female living with HIV in 2019, 0·99 male infections (0·91–1·10) for every female infection, and 1·02 male deaths (0·95–1·10) per female death. Global progress in incident cases and deaths between 2010 and 2019 was driven by sub-Saharan Africa (with a 28·52% decrease in incident cases, 95% UI 19·58–35·43, and a 39·66% decrease in deaths, 36·49–42·36). Elsewhere, the incidence remained stable or increased, whereas deaths generally decreased. Background: High-resolution estimates of HIV burden across space and time provide an important tool for tracking and monitoring the progress of prevention and control efforts and assist with improving the precision and efficiency of targeting efforts. We aimed to assess HIV incidence and HIV mortality for all second-level administrative units across sub-Saharan Africa. Methods: In this modelling study, we developed a framework that used the geographically specific HIV prevalence data collected in seroprevalence surveys and antenatal care clinics to train a model that estimates HIV incidence and mortality among individuals aged 15–49 years. We used a model-based geostatistical framework to estimate HIV prevalence at the second administrative level in 44 countries in sub-Saharan Africa for 2000–18 and sought data on the number of individuals on antiretroviral therapy (ART) by second-level administrative unit. We then modified the Estimation and Projection Package (EPP) to use these HIV prevalence and treatment estimates to estimate HIV incidence and mortality by second-level administrative unit. Findings: The estimates suggest substantial variation in HIV incidence and mortality rates both between and within countries in sub-Saharan Africa, with 15 countries having a ten-times or greater difference in estimated HIV incidence between the second-level administrative units with the lowest and highest estimated incidence levels. Across all 44 countries in 2018, HIV incidence ranged from 2·8 (95% uncertainty interval 2·1–3·8) in Mauritania to 1585·9 (1369·4–1824·8) cases per 100 000 people in Lesotho and HIV mortality ranged from 0·8 (0·7–0·9) in Mauritania to 676·5 (513·6–888·0) deaths per 100 000 people in Lesotho. Background Ambient air pollution exposure has been related to cognitive impairment. Several studies have started to explore the association of particulate matter and nitrogen oxides with cognitive impairment, studies evaluated the effect of long-term exposure to ozone(O 3 ), carbon monoxide (CO) and sulfur dioxide (SO 2 ) are sparse. Method This study is part of the ongoing Taiwan Initiative for Geriatric Epidemiological Research (TIGER). A total of 605 participants (Age 65+) were recruited from a teaching hospital in northern Taiwan in 2011 and followed up biennially. After exclusion and loss to follow-up over 6 years, a total of 292 participants were included for analyses. Global cognition was assessed by Montreal Cognitive Assessment-Taiwanese version; domain-specific cognition (logical memory, attention, verbal fluency, and executive function) were assessed by a battery of neuropsychological tests. The data of three air pollutants (O 3 , CO and SO 2 ) were obtained from 27 monitor stations of Taiwan Environmental Protection Administration (EPA) between 1993 and 2010. Air pollutants were tertiled into low, moderate, and high levels. Spatial-Time Analysis Rendering with Bayesian Maximum Entropy was utilized to establish the spatial-temporal model. Logistic regression model was utilized to assess the association of exposure to air pollutants and global or domain-specific cognition over 6 years adjusting for important covariates. Result The 17-year average level was 39.40ppb for 8-hour O 3 , 1.47ppm for 8-hour CO, and 4.70ppb for daily SO 2 , which were below the EPA standards. Exposure to high-level O 3 (>40.83 ppb) was associated with poor executive function [adjusted odds ratio (aOR)=2.14, 95% confidence interval (CI)=1.05-4.35] compared with low-level O 3 . No significant association was observed between O 3 or CO and global cognition or other cognitive domains. SO 2 were not associated with cognitive function over 6 years. Purpose: There are major gaps in our knowledge of hereditary ocular conditions in the Asia-Pacific population, which comprises approximately 60% of the world's population. Therefore, a concerted regional effort is urgently needed to close this critical knowledge gap and apply precision medicine technology to improve the quality of lives of these patients in the Asia-Pacific region. Design: Multi-national, multi-center collaborative network. Methods: The Research Standing Committee of the Asia-Pacific Academy of Ophthalmology and the Asia-Pacific Society of Eye Genetics fostered this research collaboration, which brings together renowned institutions and experts for inherited eye diseases in the Asia-Pacific region. The immediate priority of the network will be inherited retinal diseases (IRDs), where there is a lack of detailed characterization of these conditions and in the number of established registries. Results: The network comprises 55 members from 35 centers, spanning 12 countries and regions, including Australia, China, India, Indonesia, Japan, South Korea, Malaysia, Nepal, Philippines, Singapore, Taiwan, and Thailand. The steering committee comprises ophthalmologists with experience in consortia for eye diseases in the Asia-Pacific region, leading ophthalmologists and vision scientists in the field of IRDs internationally, and ophthalmic geneticists. Conclusions: The Asia Pacific Inherited Eye Disease (APIED) network aims to (1) improve genotyping capabilities and expertise to increase early and accurate genetic diagnosis of IRDs, (2) harmonise deep phenotyping practices and utilization of ontological terms, and (3) establish high-quality, multi-user, federated disease registries that will facilitate patient care, genetic counseling, and research of IRDs regionally and internationally. © 2023 The Authors

Cytokines mediate and control immune and inflammatory responses. Complex interactions exist between cytokines, inflammation and the adaptive responses in maintaining homeostasis, health, and well being. On-demand, local delivery of anti-inflammatory drug molecules to target tissues provides a means for effective drug dosing while reducing the adverse effects of systemic drug delivery. This work demonstrates a proof-of-concept theranostic approach for inflammation based on analyte-kissing induced signaling, whereby a drug (in this report, aspirin) can be released upon detection of a target level of a proinfammatory cytokine(i.e. Interferon-γ (IFN-γ)) in real time. The structure-switching aptamer based biosensor described here is capable of quantitatively and dynamically detecting IFN-γ both in vitro and in vivo with a sensitivity of 10 pg mL-1. Moreover, the released aspirin triggered by the inflammatory cytokine IFN-γ is able to inhibit inflammation in the inflammatory rat model, and the releasing of aspirin can be quantitatively controlled. Herein research provides a novel and promising strategy for in vivo detection of proinflammatory cytokines and the subsequent therapeutic delivery of anti-inflammatory molecules. This universal theranostic platform is expected to achieve great potentials on personalized medicine towards treatment of various of diseases and health conditions. © 2019, The Authors. The COVID-19 pandemic triggered unparalleled political, economic, and social ramifications, exacerbating global food insecurity (FI). To understand the overall impact of the pandemic and how different socio-economic groups were affected, we assessed prevalence and severity of FI in a sample of 18,997 households across seven countries in South America. We employed the Food Insecurity Experience Scale developed by the FAO. Our results showed that pre-pandemic, 4.5% of the sampled population across the entire continent faced Moderate FI, while 0.6% experienced severe FI. During the pandemic, Moderate FI increased to 16.9% (+ 12.4%), and Severe FI to 2.7% (+ 2.1%). By country, pre-pandemic households in Venezuela had the highest prevalence of Moderate FI (9.7%), with Peru experiencing the highest Severe FI frequency (1.1%). Peru had the greatest rise in Moderate (+ 23.9%) and Severe FI (+ 4.6%) during the pandemic. Low-income households, defined as those earning < 2 minimum wages per month, were most susceptible to FI. Uruguayan low-income families exhibited the most significant rise (+ 40.4%) in Moderate FI, while those in Peru experienced an increase of + 9.1% in Severe FI. This study measures the profound and far-reaching impact of the COVID-19 pandemic on FI in South America. Our findings also emphasise the critical importance of implementing effective public policy interventions to improve resilience against future shocks. This would enable policymakers to develop targeted strategies that address the immediate challenges posed by pandemics as well as laying the groundwork for a more resilient and sustainable food security landscape in the region. © The Author(s) 2025.
