This paper proposes an executable system architecting paradigm for discrete event system modeling and analysis through integration of a set of architecting tools, executable modeling tools, analytical tools, and visualization tools. The essential step is translating SysML-based specifications into Colored Petri Nets (CPNs) which enables rigorous static and dynamic system analysis as well as formal verification of the behavior and functionality of the SysML-based design. A set of tools have been studied and integrated that enable a structured architecture design process. Some basic principles of executable system architecture for discrete event system modeling that guide the process of executable architecture specification and analysis are discussed. This paradigm is aimed at general system design. Its feasibility was demonstrated with a C4-type network centric system as an example. The simulation results was used to check the overall integrity and internal consistency of the architecture models, refine the architecture design, and, finally, verify the behavior and functionality of the system being modeled.

We report the distance-dependent fluorescence behavior of Au-DNA conjugates prepared by equilibrating the phos-phine-stabilized gold nanoparticles (AuNPs) of 10-nm size with the designed rigid spacer double-stranded DNA consisting of thiol-modified target and Cy3-labelled complementary probe of different lengths (5 to 20 nm). The products were immobilized onto a MPTMS (3- mercaptopropyltrimethoxysilane)-modified glass substrate and were finally excited with a 532-nm laser source. Fluorescence quenching in both solution and solid platforms was assessed via disulfide bond cleavage between Au and DNA-Cy3 using mercaptoethanol. Successful and effective cleavage revealed by UV-vis absorption and fluorescence profiles was achieved in just only a few minutes.

Surface elevation and current records contain non-tidal variance, often dismissed as noise. The processes responsible for the non-tidal component may also modulate the tidal signal, altering its strength and frequency structure. Because of their manner of generation and propagation, internal tides are inherently irregular. The non-stationary character of these and other tidal processes provides an integral and useful property of tidal records, because it provides an opportunity to obtain insights into tidal dynamics and the interaction of tidal and non-tidal processes. It is, moreover, productive to use multiple approaches in analyzing coastal and estuarine tidal processes so that both the time-varying and average frequency content are determined. Only by confronting the causes of non-stationary behaviour in this way can some of the remaining challenges in tidal analysis and prediction be overcome, e.g. shelf and estuarine currents, river tides, internal tides, tide-surge interactions and tidally influenced ecological processes. Several examples illustrate the utility of non-stationary tidal analysis methods. © Springer-Verlag 2003.

Meditation is a family of ancient and contemporary contemplative mind-body practices that can modulate psychological processes, awareness, and mental states. Over the last 40 years, clinical science has manualized meditation practices and designed various meditation interventions that have shown therapeutic efficacy for disorders including depression, pain, addiction, and anxiety. Over the past decade, neuroimaging has been used to examine the neuroscientific basis of meditation practices, effects, states, and outcomes for clinical and nonclinical populations. However, the generalizability and replicability of current neuroscientific models of meditation have not yet been established, because they are largely based on small datasets entrenched with heterogeneity along several domains of meditation (e.g., practice types, meditation experience, clinical disorder targeted), experimental design, and neuroimaging methods (e.g., preprocessing, analysis, task-based, resting-state, structural magnetic resonance imaging). These limitations have precluded a nuanced and rigorous neuroscientific phenotyping of meditation practices and their potential benefits. Here, we present ENIGMA (Enhancing Neuro Imaging Genetics through Meta Analysis)–Meditation, the first worldwide collaborative consortium for neuroscientific investigations of meditation practices. ENIGMA-Meditation will enable systematic meta- and mega-analyses of globally distributed neuroimaging datasets of meditation using shared, standardized neuroimaging methods and tools to improve statistical power and generalizability. Through this powerful collaborative framework, existing neuroscientific accounts of meditation practices can be extended to generate novel and rigorous neuroscientific insights that account for multidomain heterogeneity. Effect of low oxygen conditions on matrix metalloproteinase-9 production of macrophages subjected to cyclic stretching: Involvement of ERK and Rho kinase pathways

Macrophages infiltrated in the walls of abdominal aortic aneurysms (AAA) are subjected to cyclic stretching due to pulsatile deformation of blood vessels and low oxygen conditions caused by intraluminal thrombus. These conditions could induce aberrant changes in macrophage functions and lead local weakening of AAA walls. We previously reported that the combination of 10 % cyclic stretching and 2.2% O2 conditions caused an increase in matrix metalloproteinase-9 (MMP-9) production in macrophages as well as changes in morphological responses to cyclic stretching. In the present study, we investigated the effects of oxygen concentrations on MMP-9 productions and morphological changes of macrophages subjected to cyclic stretching. Macrophages differentiated from THP-1 cells were subjected to 10% cyclic stretching under 5% or 1% O2 conditions for 24 h, and gelatinolytic activity of MMP-9 in the conditioned medium was assessed by zymography. Cells showed spread and rounded shape under static conditions and elongated and oriented to the direction of stretching after exposure to cyclic stretching, and there were no obvious effects of oxygen concentrations in cell morphology. An O2 concentration of 5% did not change MMP-9 productions of macrophages in static culture and subjected to cyclic stretching compared to normal cell culture condition of 20% O2. In contrast, 1% O2 condition stimulated MMP-9 production of macrophages both under static culture and cyclic stretching conditions. We also found that treatments of inhibitors for extracellular signal-regulated kinase (ERK) and Rho associated protein kinase (Rho kinase) suppressed the increased MMP-9 productions of macrophages by 1% O2 condition. These results suggest that lower oxygen conditions such as 1% O2 stimulate MMP-9 production in macrophages through signaling pathways involving ERK as well as Rho kinase-mediated actin cytoskeletal contractility. © 2017 The Japan Society of Mechanical Engineers.

Past air sampling detection (ASD) systems had no definitive positioning;most allocation of sampling points was based on experience. This research discovered that sampling points inside the electrical rooms were significantly affected by the ambient airflow, causing the detection system to have difficulties in fire detection. Fire dynamics simulator (FDS) was used to simulate very early smoke detection apparatus (VESDA) effectiveness within the protection area of an electrical room. Results showed that ASD sampling points in electrical rooms should be placed above high-voltage transformers and capacitors, while rearranging the high risk machines together to avoid long detection pipelines, which increases the transportation time of the gas, thus hindering early detection by the ASD system. Field tests using a smoke detector tester yielded transportation times of 33 s, which was in very good agreement with the simulated results of 35 s by ASPIRE, showing the accuracy of using software to simulate transportation times. Therefore, this research combined FDS with the transportation times of different pipeline designs to conduct performance assessment of VESDA. After appropriate rearrangement of machines, VESDA detection times dropped from 106.0-171.0 s during the original setup to 21.0-31.7 s, assuring the facility to have more time for emergency response. However, it was noticed that FDS software did not have the estimation tool for VESDA transportation times;therefore software such as ASPIRE should be used to calculate the gas transportation times inside the detection pipelines, and then compiled with FDS results to avoid underestimation of actuation times. © 2017 The Authors. A series of tertiary amine quinuclidine-capped cadmium selenide (Q-CdSe) quantum dots (QDs) of ∼4 nm in diameter was successfully synthesized via ligand exchange process in which the original hydrophobic trioctylphosphine oxide (TOPO) ligand bound to QDs was replaced with stronger quinuclidine derivatives, quinuclidine (Q1), 3-quinuclidinone (Q2) and 3-quinuclidinol (Q3). The ligand exchange of TOPO by Q probed using the combined fluorescence and absorption spectroscopy was achieved in just only a few minutes. Moreover, disappearance of prominent C-H aliphatic stretching (∼2900 cm-1) and phosphate signal (35 ppm) of TOPO-capped CdSe after replacement with Q as revealed in FT-IR and solid state 31P-NMR spectra was observed indicating efficient fast ligand exchange. © 2012 IEEE.

Genotoxicity assessment of wood-preserving waste-contaminated soil undergoing bioremediation

Bioremediation represents one of the most cost-effective technologies for treatment of petroleum hydrocarbons in contaminated surface soils. A major concern for regulatory agencies when evaluating bioremediation is how to determine acceptable levels for residual organics in soil. Although guidelines have been developed for some organics in soil, limited information is available to define acceptable levels of the metabolites of biological degradation. The products of oxidative degradation are likely to be more water soluble and may also be more toxic. The purpose of the current study was to monitor changes in compound concentration and genotoxicity in soils undergoing bioremediation. The site selected for this study was a former wood-preserving site in the northwestern United States. Soil samples were collected over a 4-year period from two 6075-m2 land treatment units. Conditions for biodegradation were enhanced by the addition of water and nutrients, as well as by frequent tilling to add oxygen. Due to the location of the facility, the temperature was conducive to a more rapid rate of biodegradation for approximately 6 months per year. Soil samples were collected using a grid system and solvent extracted. Polycyclic aromatic hydrocarbons were quantified in soil extracts using gas chromatography-mass spectrometry (GC/MS), and genotoxicity measured using the Salmonella/microsome assay. After 2 years of treatment, concentrations of total and carcinogenic polycyclic aromatic hydrocarbons (PAHs) were reduced to approximately 10% the concentration in the untreated soil. However, the mean weighted activity of the untreated soil was 293 net revertants per g soil, whereas the extracts of soil collected after 2 years induced a mean weighted activity of 325 net revertants per g soil. Thus, although biodegradation clearly reduced the concentration of total and carcinogenic PAHs in the surface soils, the results from the genotoxicity bioassay indicate that there was a lag in th

Microbial growth may occur in carpet dust when exposed to elevated moisture levels, but it is unknown how diurnal variations in equilibrium relative humidity (ERH) affect this growth. Our goal is to apply the "time-of-wetness" (TOW) concept from fungal growth on drywall to model fungal growth in carpet. Used carpet samples were embedded with dust, placed in an experimental chamber and then the ERH within the chambers was varied from 50% to either 85% or 100% for a period of 0, 6, 12, 18, or 24 hours per day over a two-week period. Quantitative polymerase chain reaction (qPCR) measurements of fungi revealed that for the 85% and 100% samples, growth could be modeled with the standard TOW and activation limited growth curve. These measurements demonstrated that the relative growth rate fit the TOW model with the Pearson Correlation Coefficients of 0.958 for 100% TOW and 0.898 for 85% TOW. DNA sequencing revealed increased abundance of species in the genera Aspergillus and Penicillium, among others, at increased moisture. Ultimately, this data can be used to accurately model fungal growth in housing based on moisture and can be utilized in future public health, policy, and epidemiological models. © 2018 15th Conference of the International Society of Indoor Air Quality and Climate, INDOOR AIR 2018.
