Histone acetylation is an important epigenetic modification that governs gene expression,chromatin changes in stress response,and cell fate *** biosensors have been developed for various epigenetic events to enable spatiotemporal tracking of sub-cellular signaling *** reported histone H3 acetylation biosensor recognizing two acetyl residues lacked *** this study,using a single bromodomain of the BRD4,we have developed a genetically encoded H3K9ac *** systematically investigated different combinations of the BET family protein as binding domains and performed site-saturated mutagenesis to optimize the biosensor,achieving a dynamic FRET change up to 30%under TSA *** the application of the optimized H3K9ac biosensor,we revealed different basal active chromatin architectures in invasive tumor cells compared to benign tumor ***,we found that H3K9ac level increased dramatically when cancer cells passed through microchannels,which models the physical constraints and mechanical microenvironmental conditions that cancer cells encounter when passing through narrow spaces within the *** result highlights the chromatin plasticity in response to external mechanical *** summary,our H3K9ac biosensor provides a versatile tool for mechanistic investigation of cell fate transition in cancer and mechanotransduction.

One of the applications of data-driven methods in the industry is the creation of real-time, embedded measurements, whether to monitor or replace sensor signals. As the number of embedded systems in products raises over time, the energy efficiency of such systems must be considered in the design. The time (processor) efficiency of the embedded software is directly related to the energy efficiency of the embedded system. Therefore, when considering some embedded software solutions, such as data-driven methods, time efficiency must be taken into account to improve energy efficiency. In this work, the energy efficiency of three data-driven methods: the Sparse Identification of Nonlinear Dynamics (SINDy), the Extreme Learning Machine (ELM), and the Random-Vector Functional Link (RVFL) network were assessed by using the creation of a real-time in-cylinder pressure sensor for diesel engines as a task. The three methods were kept with equivalent performances, whereas their relative execution time was tested and classified by their statistical rankings. Additionally, the space (memory) efficiency of the methods was assessed. The contribution of this work is to provide a guide to choose the best data-driven method to be used in an embedded system in terms of efficiency.

Reliable operation of Cr2O3:Mg/β-Ga2O3 p-n heterojunction diodes at 600◦C

β-Ga2O3-based semiconductor heterojunctions have recently demonstrated improved performance at high voltages and elevated temperatures and are thus promising for applications in power electronic devices and harsh-environment sensors. However, the long-term reliability of these ultra-wide band gap (UWBG) semiconductor devices remains barely addressed and may be strongly influenced by chemical reactions at the p-n heterojunction interface. Here, we experimentally demonstrate operation and evaluate the reliability of Cr2O3:Mg/β-Ga2O3 p-n heterojunction diodes at during extended operation at 600◦C, as well as after 30 repeated cycles between 25-550◦C. The calculated pO3-temperature phase stability diagram of the Ga-Cr-O material system predicts that Ga2O3 and Cr2O3 should remain thermodynamically stable in contact with each other over a wide range of oxygen pressures and operating temperatures. The fabricated Cr2O3:Mg/β-Ga2O3 p-n heterojunction diodes show room-temperature on/off ratios >104 at ±5V and a breakdown voltage (VBr) of -390V. The leakage current increases with increasing temperature up to 600◦C, which is attributed to Poole-Frenkel emission with a trap barrier height of 0.19 eV. Over the course of a 140-hour thermal soak at 600◦C, both the device turn-on voltage and on-state resistance increase from 1.08V and 5.34 mΩ-cm2 to 1.59V and 7.1 mΩ-cm2 respectively. This increase is attributed to the accumulation of Mg and MgO at the Cr2O3/Ga2O3 interface as observed from TOF-SIMS analysis. These findings inform future design strategies of UWBG semiconductor devices for harsh environment operation and underscore the need for further reliability assessments for β-Ga2O3 based devices. © 2024, CC BY.

This study embarked on a rigorous examination of the factors driving user satisfaction and usage behavior in the context of telehealth applications. Utilizing a well- structured Google Form survey, distributed through an array of social media platforms, including WhatsApp, Discord, Instagram, and Line, the research gathered 251 responses. The purposive sampling technique was employed to ensure that the data captured the experiences of individuals who had actively engaged with telehealth services. One hypothesis, relating to the influence of Online Reviews, was found to be unsupported. This surprising result suggests that, contrary to expectations, the opinions and reviews shared online do not significantly affect users’ satisfaction or their behavioral decisions in the context of telehealth applications. This finding highlights the need for a more nuanced understanding of the factors influencing user preferences and choices in this rapidly evolving sector. Conversely, other hypotheses examining factors such as Social Influence, Facilitating Conditions, Perceived Reliability, Price Value, and Purchase Intention were substantiated. The positive influence of these factors on customer usage behavior underscores their significance in shaping user experiences and satisfaction within the telehealth ecosystem. Furthermore, the study posits that facilitating conditions should be closely aligned with technological advancements, as this alignment is conducive to the development of enhanced application features. This insight suggests that telehealth providers should continuously innovate and integrate advanced technologies to ensure that the facilitating conditions meet the ever-evolving needs and expectations of users.

Ethereum is one of the most popular blockchain platforms with a high number of adoption in the blockchain world today. Ethereum token (ERC-20) can tokenize any real-world object while it is also possible to exchange the token across the network participants. This utility from the token drives many blockchain enthusiasts to create decentralized applications. ERC-20 token could be used to collect project funds, decision voting, or even can be used to provide investment platforms for people that want to hedge the volatility of the currency with buying an asset that is back to back with the crypto-token. This research shows the main characteristics of asset-backed token and its implementation on a complete end-to-end "digitalization assets system" (assets buy and assets redemption). NOMAD: Nonlinear Manifold Decoders for Operator Learning

Supervised learning in function spaces is an emerging area of machine learning research with applications to the prediction of complex physical systems such as fluid flows, solid mechanics, and climate modeling. By directly learning maps (operators) between infinite dimensional function spaces, these models are able to learn discretization invariant representations of target functions. A common approach is to represent such target functions as linear combinations of basis elements learned from data. However, there are simple scenarios where, even though the target functions form a low dimensional submanifold, a very large number of basis elements is needed for an accurate linear representation. Here we present NOMAD, a novel operator learning framework with a nonlinear decoder map capable of learning finite dimensional representations of nonlinear submanifolds in function spaces. We show this method is able to accurately learn low dimensional representations of solution manifolds to partial differential equations while outperforming linear models of larger size. Additionally, we compare to state-of-the-art operator learning methods on a complex fluid dynamics benchmark and achieve competitive performance with a significantly smaller model size and training cost. © 2022, CC BY-NC-SA.

A transient stability enhancement method for Grid-Forming converters enabling operation across the four-quadrant range of Power Angle

Grid-forming (GFM) converters are a promising solution for integrating converter-based resources into power grids and can be broadly categorized into (i) Synchronous Machine (SM)-inspired methods, which emulate SM control at different abstraction levels, and (ii) Virtual Oscillator Control (VOC) methods, based on oscillator network synchronization principles. Both behave as voltage sources, making them prone to transient angle instability under severe disturbances, despite differences in their phase synchronization dynamics. Existing transient stability enhancement methods for GFM converters restrict operation of power angle δ within the range [-90, 90°], limiting converter utilization and hence reducing its ability to support grid during contingencies. This paper analyzes the large-signal synchronization stability of two GFM methods—dispatchable VOC (dVOC) and Virtual Synchronous Generator (VSG)—and proposes a mode-adaptive power-angle control strategy to enhance their transient stability in ultra-weak grids, i.e., with a Short Circuit Ratio (SCR) ≈ 1 across varying X/R ratios. Assuming constant grid voltage and known line X/R ratio, the strategy estimates δ using only local measurements while maintaining a constant voltage amplitude at converter’s output. By regulating active power to control δ, it enables GFM converters to operate with SCR lower than unity across the four-quadrant range of power angle during high-impedance faults. Experimental validation confirms enhanced converter power controllability for both dVOC and VSG systems. Controller Hardware-in-the-Loop (CHIL) results demonstrate the proposed solution’s superiority over existing methods in terms of transient stability and voltage support, while exhibiting robustness against line X/R ratio measurement errors. Therefore, the proposed solution has shown improved voltage support and better utilization of the full controllability of converter output power for both dVOC and VSG-based systems. © 2013 IEEE.

Systemic evaluation of spatio-temporal variations in travel time reliability due to a toll road over time

This paper evaluates the effectiveness of toll roads in reducing congestion and improving travel time reliability on streets within its vicinity, over time. The data, from the year 2013 to 2017, for the Triangle Expressway in the city of Raleigh, North Carolina was used in the systemic evaluation process. The spatial and temporal variations in the travel time distributions on the toll road, parallel route, and near-vicinity cross-streets (in a 2-mile buffer) were evaluated. The average travel time (ATT), the 95th percentile travel time (or planning time, PT), the buffer time (BT), and the buffer time index (BTI) were computed for each link by the day-of-the-week and the time-of-the-day. Though there was an improvement in travel time after the speed limit change in the year 2014, the travel time reliability got reduced on the Triangle Expressway. Over time, there was a gradual improvement in BT and BTI. The parallel route reliability decreased significantly over time, whereas the travel time reliability of cross-streets showed a consistent trend. The stabilization of travel time distributions and the reliability measures in the year 2017 indicate that further reduction in performance measures may not be expected in the next few years on the near-vicinity links. © 2020 The Author(s). In this paper, we propose a new method to estimate the traveling direction for pedestrian by applying IMU (Inertial Measurement Unit) and PCA (Principal Component Analysis) to sensor information from a smartwatch. Because a result of PCA do not have orientation information, an estimate of the traveling direction includes an indeterminacy problem. We show that the proposed method can solve this problem and provide high accuracy in the traveling direction estimation by walking experiments. © 2020 IEEE.

Transition to a carbon-free energy system is crucial for global decarbonization and underpins Circular Economy (CE) goals. Photovoltaic (PV) technology is required for Energy Transition, but manufacturing and circular pathways can be material, energy, and carbon-intensive. Therefore, we need a prioritization of sustainability strategies for PV evolution and lifecycle management in the context of Energy Transition. This study employs a suite of quantitative metrics to compare different proposed sustainability strategies for PV modules on their ability to achieve Energy Transition. Proposals for sustainable PV range from high-yield, high-efficiency paradigms, to short-lived and fully recyclable, to long-lasting, indestructible modules. We leverage a global decarbonization deployment schedule through 2100 with the open-source PV in Circular Economy (PV ICE) tool to quantify the impacts of different evolving module design scenarios covering the range of proposed sustainability strategies. First, modules are compared on effective capacity and required replacements to meet and maintain decarbonization capacity targets through 2100. We demonstrate the effects of lifetime, degradation, and reliability on effective capacity. Next, we quantify and compare virgin material demands and lifecycle wastes, examining the impacts of lifetime and recycling rates. Finally, and critically for renewable energy technologies, we quantify the energy demands required to achieve the decarbonization capacity targets and calculate energy balance metrics (net energy, energy return on investment). These results are then summarized into a metric matrix, demonstrating tradeoffs and the importance of longevity. Our suite of mass and energy metrics provides stakeholders and decision-makers with quantitative data on circular economy choices for PV in the energy transition, enabling informed evaluation of tradeoffs of different PV module designs and CE pathways.
