Background: Systemic sclerosis (SSc) is an autoimmune disease characterized by vasculopathy, fibrosis, and immune dysregulation. In SSc pathogenesis, circulating monocytes can be recruited to the skin and differentiate into macrophages. This possibly contributes to the activation and conversion of resident fibroblasts into myofibroblasts, driving fibrosis and skin thickness. However, how these cells exert their functions and via which fibroinflammatory mediator(s) is not fully understood. Objectives: To investigate the role of monocytes in mediating myofibroblast contraction and activation using an innovative 3D collagen hydrogel model. Methods: For our 3D skin model we cocultured human primary dermal fibroblasts with either peripheral blood mononuclear cells (PBMCs) or MACS-sorted CD14+ monocytes in a 3D collagen type 1 hydrogel. Subsequently, monocyte-driven tissue contraction was measured over time. We performed immunostainings for CD68, fibroblast activation protein (FAP), and alpha-smooth muscle actin (α-SMA) to evaluate monocyte/macrophage and fibroblast activation. To investigate the signaling pathways involved in the observed myofibroblast activation, we measured transcription factor-driven luciferase production using reporter constructs in the same dermal fibroblasts. The activity of the following reporter constructs was determined after 24 hours: Sis-Inducible Element (SIE); SMAD-Binding Element (SBE); Nuclear Factor of Activated T-cells 5 Response Element (NFAT-5), and NFκB Response Element (NFκB). Results: After 60 hours of co-culture, hydrogel plugs containing fibroblasts + monocytes displayed a strong spontaneous contraction, seen by a (approx. 80-90%) decrease in the area of the plugs. Hydrogels containing only fibroblasts did not contract. Hydrogels with fibroblasts + monocytes contracted as fast as fibroblasts + PBMCs, but depletion of CD14+ cells from PBMCs slowed down contraction, showing that monocytes strongly activated fibroblasts. Analysis of energy pathways is useful in helping to understand adaptations to specific sports practices, which vary according to the type of effort and individual characteristics of the athletes. In this sense, this study aimed to describe the physiological changes related to lipid markers in amputee athletes who practice para-sport jiu-jitsu. This is an observational study, which included three male amputee athletes, aged between 41 and 46 years old and with more than 24 months of experience at a professional level. The collection procedures included the verification of anthropometric measurements (body mass and height), handgrip strength, body fat and lipid profile, before and after a simulated para-sport jiu-jitsu fight. The main results of the study refer to elevations in the values of total lipids (13.2 %);total cholesterol (11.8 %);and phospholipids (7.7 %) in the three participants. The findings of other indices had more discrepant results, ranging from 14 % (very low density lipoprotein and triacylglycerol) to 65 % (high density lipoprotein). In addition, a high percentage of body fat was observed among the participants. The data presented require attention, since understanding metabolic demands is crucial to provide favorable conditions for sports practice at safe levels and expected sports performance. © 2024 The Authors

Anacardium humile, known as “cajuzinho-do-cerrado,” is a native Brazilian Cerrado fruit similar to the cashew but less studied, particularly its phenolic compound content and profile. This study used a factorial design to examine how A. humile concentration and maturation time affect the liqueur's physicochemical attributes, phenolic content, antioxidant capacity, and sensory appeal. The liqueurs exhibited low antioxidant capacities as determined by the 2,2-diphenyl-1-picrylhydrazyl radical scavenging capacity assay (DPPH), ferric ion reducing antioxidant power (FRAP), and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid). This is likely attributable to a reduction in phenolic compounds within the liqueurs compared to A. humile. The phenolic compound profile of A. humile showed 11 compounds, comprising 5 flavonols, 3 hydroxycinnamic acids, and 2 hydrolyzable tannins. Treatment 8 (500 g of A. humile and 17 days of maturation) showed the highest value for phenolic compound content. However, with regard to sensory analysis, this was the darkest liqueur and the least accepted test, while Treatment 5 (147.5 g of A. humile and 10 days of maturation) was one of the most accepted and chosen as the best liqueur due to the highest antioxidant capacity by FRAP and DPPH methods. The fusion of technology and factorial design proved efficacious in formulating functional liqueurs. © 2023

Still, Zika virus (ZIKV) infection poses a substantial public health risk, especially for pregnant women and their fetuses, as it can result in congenital abnormalities and fetal mortality during pregnancy. Despite significant advances in understanding and combating ZIKV, considerable challenges remain in the fight against this flavivirus. A crucial component of this effort is the development of vaccines, none of which have yet been licensed for human use. Here, we present a comprehensive study of a novel ZIKV vaccine candidate based on virus-like particles (VLPs), designed to provide broad immunological protection against viral infection combined with safety, without the need for additional adjuvants. A self-adjuvanted VLPs-based vaccine displaying the envelope protein domain III (EDIII) of ZIKV was built. The EDIII protein was expressed in E. coli and chemically conjugated to QβVLPs. Immunization of C57BL/6 mice with two doses of the EDIII-QβVLPs vaccine elicited strong EDIII-specific Th1-based immune response. Notably, the vaccine induced neutralizing antibodies and conferred protection in type I IFN receptor-deficient (G129) mice against ZIKV challenge. Furthermore, vaccinated male mice were protected from ZIKV-induced cerebral and testicular damage, critical concerns for ZIKV pathogenesis. These findings suggest that the EDIII-QβVLP vaccine is a promising candidate for preventing ZIKV infection, with potential applications in combatting this and other emerging flaviviruses.

Background: Diabetes is one of the leading causes of death and disability worldwide, and affects people regardless of country, age group, or sex. Using the most recent evidentiary and analytical framework from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD), we produced location-specific, age-specific, and sex-specific estimates of diabetes prevalence and burden from 1990 to 2021, the proportion of type 1 and type 2 diabetes in 2021, the proportion of the type 2 diabetes burden attributable to selected risk factors, and projections of diabetes prevalence through 2050. Methods: Estimates of diabetes prevalence and burden were computed in 204 countries and territories, across 25 age groups, for males and females separately and combined;these estimates comprised lost years of healthy life, measured in disability-adjusted life-years (DALYs;defined as the sum of years of life lost [YLLs] and years lived with disability [YLDs]). We used the Cause of Death Ensemble model (CODEm) approach to estimate deaths due to diabetes, incorporating 25 666 location-years of data from vital registration and verbal autopsy reports in separate total (including both type 1 and type 2 diabetes) and type-specific models. Other forms of diabetes, including gestational and monogenic diabetes, were not explicitly modelled. Total and type 1 diabetes prevalence was estimated by use of a Bayesian meta-regression modelling tool, DisMod-MR 2.1, to analyse 1527 location-years of data from the scientific literature, survey microdata, and insurance claims;type 2 diabetes estimates were computed by subtracting type 1 diabetes from total estimates. Mortality and prevalence estimates, along with standard life expectancy and disability weights, were used to calculate YLLs, YLDs, and DALYs. Background: Mycobacterium tuberculosis multidrug resistance, especially against rifampicin and isoniazid, places pulmonary tuberculosis in the list of emerging diseases. The misuse of therapeutic regimens is one of its main predisposing factors. Case presentation: Four clinical cases (three were brown and one black) with multidrug-resistant tuberculosis, treated in a reference hospital in the state of Maranhão, Brazil, were reported to evaluate the importance of radiological framework on disease evolution. Conclusion: The clinical framework showed a bad evolution and drug resistance, while radiology showed lung lesions, ranging from exudative infiltrates to lung parenchyma disintegration. © 2016 Maia et al.

Background: Decades of steady improvements in life expectancy in Europe slowed down from around 2011, well before the COVID-19 pandemic, for reasons which remain disputed. We aimed to assess how changes in risk factors and cause-specific death rates in different European countries related to changes in life expectancy in those countries before and during the COVID-19 pandemic. Methods: We used data and methods from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to compare changes in life expectancy at birth, causes of death, and population exposure to risk factors in 16 European Economic Area countries (Austria, Belgium, Denmark, Finland, France, Germany, Greece, Iceland, Ireland, Italy, Luxembourg, the Netherlands, Norway, Portugal, Spain, and Sweden) and the four UK nations (England, Northern Ireland, Scotland, and Wales) for three time periods: 1990–2011, 2011–19, and 2019–21. Changes in life expectancy and causes of death were estimated with an established life expectancy cause-specific decomposition method, and compared with summary exposure values of risk factors for the major causes of death influencing life expectancy. Findings: All countries showed mean annual improvements in life expectancy in both 1990–2011 (overall mean 0·23 years [95% uncertainty interval [UI] 0·23 to 0·24]) and 2011–19 (overall mean 0·15 years [0·13 to 0·16]). The rate of improvement was lower in 2011–19 than in 1990–2011 in all countries except for Norway, where the mean annual increase in life expectancy rose from 0·21 years (95% UI 0·20 to 0·22) in 1990–2011 to 0·23 years (0·21 to 0·26) in 2011–19 (difference of 0·03 years). In other countries, the difference in mean annual improvement between these periods ranged from –0·01 years in Iceland (0·19 years [95% UI 0·16 to 0·21] vs 0·18 years [0·09 to 0·26]), to –0·18 years in England (0·25 years [0·24 to 0·25] vs 0·07 years [0·06 to 0·08]).
