The K-means algorithm is a widely used clustering algorithm that offers simplicity and efficiency. However, the traditional K-means algorithm uses the random method to determine the initial cluster centers, which make clustering results prone to local optima and then result in worse clustering performance. Many initialization methods have been proposed, but none of them can dynamically adapt to datasets with various characteristics. In our previous research, an initialization method for K-means based on hybrid distance was proposed, and this algorithm can adapt to datasets with different characteristics. However, it has the following drawbacks: (a) When calculating density, the threshold cannot be uniquely determined, resulting in unstable results. (b) Heavily depending on adjusting the parameter, the parameter must be adjusted five times to obtain better clustering results. (c) The time complexity of the algorithm is quadratic, which is difficult to apply to large datasets. In the current paper, we proposed an adaptive initialization method for the K-means algorithm (AIMK) to improve our previous work. AIMK can not only adapt to datasets with various characteristics but also obtain better clustering results within two interactions. In addition, we then leverage random sampling in AIMK, which is named as AIMK-RS, to reduce the time complexity. AIMK-RS is easily applied to large and high-dimensional datasets. We compared AIMK and AIMK-RS with 10 different algorithms on 16 normal and six extra-large datasets. The experimental results show that AIMK and AIMK-RS outperform the current initialization methods and several well-known clustering algorithms. Furthermore, AIMK-RS can significantly reduce the complexity of applying it to extra-large datasets with high dimensions. The time complexity of AIMK-RS is O(n). Copyright © 2019, The Authors. BACKGROUND:Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is the most common monogenic form of hereditary cerebral small vessel disease (CSVD). While studies have demonstrated heterogeneous alterations in cerebrovascular reactivity (CVR) and gray matter structure in CADASIL patients, the diagnostic utility of CVR and brain structural imaging in identifying CADASIL remains uncertain. METHODS:This study investigated CVR and gray matter density (GMD) in 22 CADASIL patients compared to 21 age- and sex-matched healthy controls. Resting-state functional magnetic resonance imaging (fMRI) assessed regional CVR changes, while voxel-based morphometry (VBM) analyzed GMD differences. The study further explored the correlation between CVR and GMD alterations with clinical scores in CADASIL patients. Finally, receiver operating characteristic (ROC) curve analysis, including the area under curve (AUC) calculation, determined the diagnostic accuracy of CVR and morphological indices for CADASIL. RESULTS:Compared to controls, CADASIL patients exhibited significantly lower CVR in the left substantia nigra and higher CVR in the bilateral proximal anterior cerebral artery (ACA). VBM revealed reduced GMD in bilateral thalamic subregions in CADASIL patients, significantly correlating with several clinical measures. CVR indicators demonstrated superior diagnostic accuracy for CADASIL (AUC = 0.853) compared to GMD (AUC = 0.654). CONCLUSION:The significant changes in CVR indices observed in CADASIL patients suggest that these indices may serve as potential biomarkers for CADASIL diagnosis.

Paroxysmal dyskinesias are a group of neurological diseases characterized by intermittent episodes of involuntary movements with different *** kinesigenic dyskinesia(PKD)is the most common type of paroxysmal dyskinesia and can be divided into primary and secondary types based on the ***,PKD is characterized by recurrent and transient attacks of involuntary movements precipitated by a sudden voluntary *** major cause of primary PKD is genetic abnormalities,and the inheritance pattern of PKD is mainly autosomal-dominant with incomplete *** proline-rich transmembrane protein 2(PRRT2)was the first identified causative gene of PKD,accounting for the majority of PKD cases *** increasing number of studies has revealed the clinical and genetic characteristics,as well as the underlying mechanisms of *** seeking the views of domestic experts,we propose an expert consensus regarding the diagnosis and treatment of PKD to help establish standardized clinical evaluation and therapies for *** this consensus,we review the clinical manifestations,etiology,clinical diagnostic criteria and therapeutic recommendations for PKD,and results of genetic analyses in PKD patients performed in domestic hospitals.

Accumulating evidences have shown gut microbial dysbiosis in patients with bipolar disorder(BD),but little is known how the microbiota-gut-brain axis operates in *** this study,we aimed to characterize the gut microbiota with metagenomic sequencing and explored its linkage to tetratricopeptide repeat and ankyrin repeat containing 1(TRANK1),a risk gene of *** samples were obtained from 60 healthy individuals and 62 patients in acute depressive episodes,who received over one-month quetiapine treatment *** analysis of BD microbiota revealed specific alterations in microbial diversity and compositions and the impacts of quetiapine treatment on microbial *** neural oscillation in hippocampus was positively associated with the abundance of Clostridium *** transplanted with feces from depressed patients presented depression-like *** mRNA expressions of inflammatory cytokines,brain-derived neurotrophic factor,kynurenine aminotransferase Ⅱ,and TRANK1 were detected in mice hippocampus and prefrontal ***3.1(+) vector encoding human TRANK 1 was constructed to explore the impact of TRANK1 overexpression on neuronal morphology,which manifested as decreased dendritic spine density in primary cortical *** findings add new evidences to microbiota-gut-brain regulation in BD,indicating that microbiota is possibly involved in the neuropathogenesis of BD by modulating the expression of TRANK1.

Surface Modification of (001) Facets Dominated TiO2 with Ozone for Adsorption and Photocatalytic Degradation of Gaseous Toluene

This study investigated the positive effect of surface modification with ozone on the photocatalytic performance of anatase TiO2 with dominated（001） facets for toluene *** performance of photocatalyst was tested on a home-made volatile organic compounds degradation system. The ozone modification, toluene adsorption and degradation mechanism were established by a combination of various characterization methods, in situ diffuse reflectance infrared fourier transform spectroscopy, and density functional theory *** surface modification with ozone can significantly enhance the photocatalytic degradation performance for toluene. The abundant unsaturated coordinated 5 c-Ti sites on（001）facets act as the adsorption sites for ozone. The formed Ti–O bonds reacted with H2O to generate a large amount of isolated Ti5 c-OH which act as the adsorption sites for toluene,and thus significantly increase the adsorption capacity for toluene. The outstanding photocatalytic performance of ozone-modified TiO2 is due to its high adsorption ability for toluene and the abundant surface hydroxyl groups, which produce very reactive OH·radicals under irradiation. Furthermore, the O2 generated via ozone dissociation could combine with the photogenerated electrons to form superoxide radicals which are also conductive to the toluene degradation.

Network analysis of signals originating from different parts of brain during motor imagery (MI) has gained lots of interest recently. In this paper, we used EEG to construct the brain network during MI, and analyzed the topological characteristics of the EEG function network. It is found that the node degree and clustering coefficient of the right hand MI is higher than the left hand MI on the nodes of right sensorimotor cortex, and the characteristic path length of the network is shorter for right hand MI than left hand MI. As for the right hand MI task, most subjects showed higher node degree and clustering coefficient on the right sensorimotor cortex than the left sensorimotor cortex. We conclude that EEG network based measures, which captures the brain information integrating characteristics during MI, may serve as useful features for classification in MI-BCI application, especially for BCI inefficiency subject. © 2017 IEEE.

BACKGROUND: Dysfunctional reward processing is implicated in multiple mental disorders. Novelty seeking (NS) assesses preference for seeking novel experiences, which is linked to sensitivity to reward environmental cues. METHODS: A subset of 14-year-old adolescents (IMAGEN) with the top 20% ranked high-NS scores was used to identify high-NS-associated multimodal components by supervised fusion. These features were then used to longitudinally predict five different risk scales for the same and unseen subjects (an independent dataset of subjects at 19 years of age that was not used in predictive modeling training at 14 years of age) (within IMAGEN, n z 1100) and even for the corresponding symptom scores of five types of patient cohorts (non-IMAGEN), including drinking (n = 313), smoking (n = 104), attention-deficit/hyperactivity disorder (n = 320), major depressive disorder (n = 81), and schizophrenia (n = 147), as well as to classify different patient groups with diagnostic labels. RESULTS: Multimodal biomarkers, including the prefrontal cortex, striatum, amygdala, and hippocampus, associated with high NS in 14-year-old adolescents were identified. The prediction models built on these features are able to longitudinally predict five different risk scales, including alcohol drinking, smoking, hyperactivity, depression, and psychosis for the same and unseen 19-year-old adolescents and even predict the corresponding symptom scores of five types of patient cohorts. Furthermore, the identified reward-related multimodal features can classify among attention-deficit/hyperactivity disorder, major depressive disorder, and schizophrenia with an accuracy of 87.2%. CONCLUSIONS: Adolescents with higher NS scores can be used to reveal brain alterations in the reward-related system, implicating potential higher risk for subsequent development of multiple disorders. HCV is a hepatotropic RNA virus recognized for its frequent virulence and fatality worldwide. Despite many vaccine development programs underway, researchers are on a quest for natural bioactive compounds due to their multivalent efficiencies against viral infections, considering which the current research aimed to figure out the target-specificity and therapeutic potentiality of α, β, and δ subunits of amyrin, as novel bioactive components against the HCV influx mechanism. Initially, the novelty of amyrin subunits was conducted from 203 pharmacophores, comparing their in-silico pharmacokinetic and pharmacodynamic profiles. Besides, the best active site of CD81 was determined following the quantum tunneling algorithm. The molecular dynamic simulation was conducted (100 ns) following the molecular docking steps to reveal the parameters- RMSD (Å); Cα; RMSF (Å); MolSA (Å); Rg (nm); PSA (Å); SASA (Å), and the MM-GBSA dG binding scores. Besides, molecular strings of CD81, along with the co-expressed genes, were classified, as responsible for encoding CD81-mediated protein clusters during HCV infection, resulting in the potentiality of amyrins as targeted prophylactics in HCV infection. Finally, in vivo profiling of the oxidative stress marker, liver-specific enzymes, and antioxidant markers was conducted in the DMN-induced mice model, where -amyrin scored the most significant values in all aspects.
