In the current industrial revolution, additive manufacturing (AM) embodies a promising technology that can enhance the effectiveness, adaptability, and competitiveness of supply chains (SCs). Moreover, it facilitates the development of distributed SCs, thereby enhancing product availability, inventory levels, and lead time. However, the wide adoption of AM in industrial SCs creates various challenges, leading to new difficulties for SC design. In this context, this paper proposes a new design approach to AM SCs using optimization methods. More specifically, the proposed approach, comprising the pmedian and mixed-integer linear programming models, considers the decision of deploying productive resources (3D printers) in specific locations of generic spare part SCs. The approach was evaluated in a real-world use case of an elevator maintenance service provider. The obtained results demonstrated the promising capabilities of the proposed design approach in managing the challenges arising from the forthcoming widespread use of 3D printers in manufacturing SCs. © 2020, CC-BY-NC-ND.

Benefit for clinical melanoma treatments, the transdermal neoadjuvant therapy could reduce surgery region and increase immunotherapy efficacy. Using lipoplex (Lipo-PEG-PEI-complex, LPPC) encapsulated doxorubicin (DOX) and carrying CpG oligodeoxynucleotide;the transdermally administered nano-liposomal drug complex (LPPC-DOX-CpG) would have high cytotoxicity and immunostimulatory activity to suppress systemic metastasis of melanoma. LPPC-DOX-CpG dramatically suppressed subcutaneous melanoma growth by inducing tumor cell apoptosis and recruiting immune cells into the tumor area. Animal studies further showed that the colonization and growth of spontaneously metastatic melanoma cells in the liver and lung were suppressed by transdermal LPPC-DOX-CpG. Furthermore, NGS analysis revealed IFN-γ and NF-κB pathways were triggered to recruit and activate the antigen-presenting-cells and effecter cells, which could activate the anti-tumor responses as the major mechanism responsible for the therapeutic effect of LPPC-DOX-CpG. Finally, we have successfully proved transdermal LPPC-DOX-CpG as a promising penetrative carrier to activate systemic anti-tumor immunity against subcutaneous and metastatic tumor. © 2022

A system for automatic detection of road damages is essential for logistics management. At the core of the system lies an algorithm for classification of damages. This work intends to establish such an algorithm. In the current proposal, the algorithm receives data of the probe vehicle rate of rotations, extracts statistics descriptive from the data to produce a feature vector, and finally, feeds the vector to a simple-three-layer neural network to classify the damages. The types of damages are limited to four cases: normal, pothole, speed bump, and expansion joint. For the development, a probe vehicle is used to produce 400 empirical data involving those four damage cases. By using Monte Carlo approach, the established neural network model is evaluated. The results show that the approach is able to classify the cases with 85% accuracy. The study also finds that the rates of pitch and roll to be the determining factors. © 2018 Institute of Physics Publishing. All rights reserved.

The CALorimetric Electron Telescope (CALET), developed and operated by Japan in collaboration with Italy and the United States, is a high-energy astroparticle physics experiment installed on the International Space Station (ISS). Its mission goals include investigating the possible presence of nearby sources of high-energy electrons, performing direct measurements of observables sensitive to the details of the acceleration and propagation of galactic particles, and detecting potential dark matter signatures. CALET measures cosmic-ray electron+positron flux up to 20 TeV, gamma rays up to 10 TeV, and nuclei up to 1,000 TeV. Charge measurements cover from Z=1 to 40 allowing to study the more abundant elements and to extend the range of long-term observations above iron. CALET is collecting science data on the International Space Station since October 2015 with excellent and continuous performance with no major interruptions. Approximately 20 million triggered events per month are recorded with energies > 10 GeV. Here, we present the highlights of CALET observations carried out during the first 5.5 years of operation, including the electron+positron energy spectrum, the spectra of protons and other nuclei, gamma-ray observations, as well as the characterization of on-orbit performance. Some results on the electromagnetic counterpart search for LIGO/Virgo gravitational wave events and the observations of solar modulation and gamma-ray bursts are also included. © Copyright owned by the author(s).

Control of industrial plants is an important engineering field of study, where the optimization of such processes can lead to better product quality and higher profit. The present work deals with the optimization of an industrial gasifier, proposed as a benchmark challenge by ALSTOM Power Technologies in 2002. Researchers from around the globe proposed different methods for optimizing such system, using linear and nonlinear system identification techniques, different controller schemes, such as proportional-integral-derivative and model predictive controllers, and search methods for controller tuning. The authors care for the study of a methodology that can generalize the controller optimization process of real industrial systems, where linear system identification can lead to simpler systems, which are used to design more robust controllers. The proposed work deals with linear system identification and multiobjective optimization for the gasifier's controller design. To do so, the authors divide the problem in two steps, the linear system identification, where step functions are fed to the gasifier in order to detect the output response of the system, and the design of a proportional-integral-derivative controller, where spherical pruned multi-objective differential evolution algorithm is used in order to optimize the controller gains. The proposed method, unfortunately, does not provide an optimal controller, which can be explained by the complexity of the benchmark system. With such results, the authors believe a better multi-objective problem definition is necessary. Also, it is concluded that such benchmark presents a real challenge for future controller design methndologies.

Real-world classification problems generally deal with imbalanced data, where one class represents the majority of the data set. The present work deals with event detection on a drinking-water quality time series, where the presence of a quality event is the minority class. In order to solve such problems, supervised learning algorithms are recommended. Researchers have also used multi-objective optimization (MOO) in order to generate diverse models to build ensembles of classifiers. Although MOO has been used for ensemble member generation, there is a lack on it's application for member selection, which is usually done by selecting a specific subset from the resulting models, or by using meta-algorithms, such as boosting. The proposed work comprises the application of MOO design in the whole process of ensemble generation. To do so, one multi-objective problem (MOP) is defined for the creation of a set of non-dominated solutions with Pareto-optimal support vector machines (SVM). After that, a second MOP is defined for the selection of such SVMs as members of an ensemble. Such methodology is compared to other member selection methods, such as: the single best classifier, an ensemble composed of the full set of non-dominated solutions, and the selection of a specific subset from the Pareto front. Results show that the proposed method is suitable for the creation of ensembles, achieving the highest classification scores.

Tumor hypoxia leads to radioresistance and markedly worse clinical outcomes for pediatric malignant rhabdoid tumors (MRTs). Our transcriptomics and bioenergetic profiling data reveal that mitochondrial oxidative phosphorylation is a metabolic vulnerability of MRT and can be exploited to overcome consumptive hypoxia by repurposing an FDA-approved antimalarial drug, atovaquone (AVO). We then establish the utility of oxygen-enhanced-multispectral optoacoustic tomography, a label-free, ionizing radiation-free imaging modality, to visualize and quantify spatiotemporal changes in tumor hypoxia in response to AVO. We show a potent but transient increase in tumor oxygenation upon AVO treatment that results in complete elimination of tumors in all tested mice when combined with 10-gray radiotherapy, a dose several times lower than the current clinic standard. Last, we use translational mathematical modeling for systematic evaluation of dosing regimens, administration timing, and therapeutic synergy in a virtual patient cohort. Together, our work establishes a framework for safe and pediatric patient-friendly image-guided metabolic radiosensitization of rhabdoid tumors.

Organizational models of production systems that focus on services and servitization are experiencing an increase in an application. These models emphasize the value proposition for the customer and a better understanding of the value creation process. Additionally, the emergence of new technologies such as those associated with Industry 4.0 also requires well-structured and results-oriented processes. In this perspective, although there are several tools for measuring both operations and organizational performance, the literature still lacks tools that integrate and align these two perspectives, accounting for activities at the operational level that reflect objectives at the strategic level, and vice-versa. Therefore, a procedure for aligning process performance (operations' performance) and strategic performance (organizational performance) in a service context is proposed. This procedure is based on three main components identified through a literature review: (1) guidelines associated with enterprise engineering that focus on processes, developed by Deschamps (2013);(2) a set of critical success factors for implementation of performance measurement systems that focus on organizational performance, developed by Keathley (2016);and (3) ten determinants of service quality, proposed by Parasuraman (1985). The alignment procedure is presented as being mediated by Parasuraman's model, relating enterprise engineering guidelines and critical success factors. © 2018 Institute of industrial Engineers (IIE). In this paper, we address a decision-making problem related to the requirement of costly equipment by medical diagnostic services in a segmented public healthcare system comprising several institutions and private providers. The problem is motivated by a real-world case of the Mexican healthcare system. The aim of this study is to determine which hospitals can provide the service, their capacity levels, the allocation of demand in each institution, and the referral of patients to other institutions or private providers while minimizing annual investment costs and operating costs required to satisfy demand. A mixed-integer linear programming model that takes into account different characteristics such as patient acuity levels, types of equipment, and demand variation through time is introduced. The model was empirically assessed to evaluate its impact on the decision-making process. A sensitivity analysis to evaluate solution behavior for variations of critical parameters was performed. The results showed that some values could generate a significant effect on the total costs for the service coverage and in the efficiency of the service, whereas overall results indicated the usefulness of the model. While this model is valuable to aid this decision-making problem, it is limited to medium-size instances of up to 90 facilities. To solve the problems with larger instances, a two-phase heuristic algorithm is proposed. In the first phase, the method uses a greedy construction mechanism, and in the second phase, it attempts to improve the solution. Empirical evidence on large instances shows that good solutions with low computing times are reached in comparison with the exact method. © 2019 World Scientific Publishing Company.

Background: Despite major advances in artificial intelligence (AI) research for healthcare, the deployment and adoption of AI technologies remain limited in clinical practice. In recent years, concerns have been raised about the clinical, technical, ethical and legal risks associated with healthcare AI. To increase adoption in the real world, it is essential that AI technologies are trusted and accepted by patients, clinicians, health organisations and authorities. This paper describes the FUTURE-AI framework as the first international consensus guideline for trustworthy AI in healthcare. Methods: The FUTURE-AI consortium was founded in 2021 and now comprises 117 interdisciplinary experts from 50 countries representing all continents, including AI scientists, clinical researchers, biomedical ethicists, and social scientists. Over a two-year period, the consortium established guiding principles and best practices for trustworthy and deployable AI through an iterative process comprising an in-depth literature review, a modified Delphi survey, and online consensus meetings. Findings: The FUTURE-AI framework was established based on six guiding principles for trustworthy AI in healthcare, i.e. Fairness, Universality, Traceability, Usability, Robustness and Explainability. Through consensus, a set of 30 best practices were defined, addressing technical, clinical, socio-ethical and legal dimensions of trustworthy AI. The recommendations cover the entire lifecycle of healthcare AI, from design, development and validation to regulation, deployment, and monitoring. Interpretation: FUTURE-AI is a structured, risk-informed framework which provides guidance for constructing healthcare AI tools that will be trusted, deployed and adopted in real-world clinical practice. Researchers are encouraged to take the recommendations into account in proof-of-concept stages to facilitate future translation towards clinical practice of healthcare AI.
