In 2023, we proposed a hypothetical knowledge management framework that incorporates science fiction prototyping (SFP) into the traditional roadmapping (RM) method to make RM more innovative and create a future beyond what we can currently imagine. Our main objective is to create a new organizational knowledge co-creation method for generating innovative ideas by incorporating humanistic factors utilizing SFP into RM. In the current article, we present the exploratory study we conducted to examine the hypothetical framework. Experimental workshops were held in November 2023 to compare the integrated method, in which SFP is applied to RM, with the original RM method. We followed a crossover design in the workshops and analyzed the results qualitatively and quantitatively following the perspectives proposed in the hypothetical framework. The qualitative analysis revealed successful integration regarding diverse and broader perspectives. The quantitative analysis utilizing Wilcoxon signed rank tests showed that the scores of the integrated method were significantly higher than those of RM for variables evaluating the effectiveness of the integration, while not for variables related to participants' feelings and satisfaction with the workshops, and sustainability. These findings indicate that the integration was successful in terms of its effectiveness as a method, while participants' subjective feelings toward the workshops were not different in both methods. As for sustainability, which requires a long-term perspective, no apparent differences were observed in these short-term workshops. © 2024 PICMET.

Mitochondrial division inhibitor 1 (Mdivi-1) is a well-known synthetic compound aimed at inhibiting dynamin-related protein 1 (Drp1) to suppress mitochondrial fission, making it a valuable tool for studying mitochondrial dynamics. However, its specific effects beyond Drp1 inhibition remain to be confirmed. In this study, we employed integrative proteomics and phosphoproteomics to delve into the molecular responses induced by Mdivi-1 in SK-N-BE(2)C cells. A total of 3070 proteins and 1945 phosphorylation sites were identified, with 880 of them represented as phosphoproteins. Among these, 266 proteins and 97 phosphorylation sites were found to be sensitive to the Mdivi-1 treatment. Functional enrichment analysis unveiled their involvement in serine biosynthesis and extrinsic apoptotic signaling pathways. Through targeted metabolomics, we observed that Mdivi-1 enhanced intracellular serine biosynthesis while reducing the production of C24:1-ceramide. Within these regulated phosphoproteins, dynamic dephosphorylation of proteasome subunit alpha type 3 serine 250 (PSMA3-S250) occurred after Mdivi-1 treatment. Further site-directed mutagenesis experiments revealed that the dephosphorylation-deficient mutant PSMA3-S250A exhibited a decreased cell survival. This research confirms that Mdivi-1’s inhibition of mitochondrial division leads to various side effects, ultimately influencing cell survival, rather than solely targeting Drp1 inhibition. © 2023 American Chemical Society.

Goal-oriented inference of environment from redundant observations

The agent learns to organize decision behavior to achieve a behavioral goal, such as reward maximization, and reinforcement learning is often used for this optimization. Learning an optimal behavioral strategy is difficult under the uncertainty that events necessary for learning are only partially observable, called as Partially Observable Markov Decision Process (POMDP). However, the real-world environment also gives many events irrelevant to reward delivery and an optimal behavioral strategy. The conventional methods in POMDP, which attempt to infer transition rules among the entire observations, including irrelevant states, are ineffective in such an environment. Supposing Redundantly Observable Markov Decision Process (ROMDP), here we propose a method for goal-oriented reinforcement learning to efficiently learn state transition rules among reward-related"core states" from redundant observations. Starting with a small number of initial core states, our model gradually adds new core states to the transition diagram until it achieves an optimal behavioral strategy consistent with the Bellman equation. We demonstrate that the resultant inference model outperforms the conventional method for POMDP. We emphasize that our model only containing the core states has high explainability. Furthermore, the proposed method suits online learning as it suppresses memory consumption and improves learning speed. Copyright © 2023, The Authors. All rights reserved.

We present ViPRA-Haplo, a de novo strain-specific assembly workflow for reconstructing viral haplotypes in a viral population from paired-end next generation sequencing (NGS) data. The proposed Viral Path Reconstruction Algorithm (ViPRA) generates a subset of paths from a De Bruijn graph of reads using the pairing information of reads. The paths generated by ViPRA are an over-estimation of the true contigs. We propose two refinement methods to obtain an optimal set of contigs representing viral haplotypes. The first method clusters paths reconstructed by ViPRA using VSEARCH Deorowicz et al. 2015 based on sequence similarity, while the second method, MLEHaplo, generates a maximum likelihood estimate of viral populations. We evaluated our pipeline on both simulated and real viral quasispecies data from HIV (and real data from SARS-COV-2). Experimental results show that ViPRA-Haplo, although still an overestimation in the number of true contigs, outperforms the existing tool, PEHaplo, providing up to 9% better genome coverage on HIV real data. In addition, ViPRA-Haplo also retains higher diversity of the viral population as demonstrated by the presence of a higher percentage of contigs less than 1000 base pairs (bps), which also contain k-mers with counts less than 100 (representing rarer sequences), which are absent in PEHaplo. For SARS-CoV-2 sequencing data, ViPRA-Haplo reconstructs contigs that cover more than 90% of the reference genome and were able to validate known SARS-CoV-2 strains in the sequencing data. © 2004-2012 IEEE.

In the realm of research, the global health challenge posed by lung cancer remains pronounced, contributing substantially to annual cancer-related fatalities. The critical imperative lies in the early identification of pulmonary nodules, frequently indicative of impending lung cancer, to enhance patient outcomes and diminish mortality rates. Computed Tomography (CT) imaging stands out as a pivotal diagnostic instrument for the timely detection of these nodules. The swift proliferation of medical imaging data has underscored the pressing necessity for precise and efficient methodologies dedicated to nodule segmentation and measurement. These approaches are crucial in assisting radiologists in their diagnostic and clinical decision-making endeavors. In this study, we introduced a thorough method for analyzing lung nodules, leveraging dataset from Far Eastern Memorial Hospital (FEMH) comprising original CT images and manually annotated ground truth masks obtained with the assistance of radiologists at FEMH. This dataset is utilized for the segmentation of nodules. We employed advanced deep learning models, specifically the U-Net architecture, identified as the optimal model through our training process. We made substantial progress in nodule segmentation, attaining an Intersection over Union (IoU) score of 0.824 and a Dice Coefficient of 0.903 for the FEMH dataset. Furthermore, our performance improved when utilizing the merged dataset comprising FEMH and Luna16, yielding an IoU score of 0.862 and a Dice Coefficient of 0.926. Luna16 has been extensively utilized in numerous studies related to nodule detection and segmentation. In the next phase of the study, the best-performing model from our segmentation phase was utilized to predict nodule masks on the merged dataset. Subsequently, we measured the size of each predicted nodule by comparing it with the size ground truth mask in millimeters. The design and construction of delivering announcements using a web-based SMS gateway aims to help overcome problems that often occur at pioneer airports; this is because when passengers want to travel, many passengers still arrive late to check-in, even though the passenger ticket has a notification for checking. -in 1 hour before Departure. If a flight's Departure is delayed or accelerated, passengers will only receive this information when they arrive at the airport, whereas if at the time the departure notification is delayed or accelerated, the passenger is still outside the airport or still on their way to the airport, as a result, they will definitely not receive the information. Testing voice announcements on parking, arrival, departure, delay, check-in, and waiting room services, as well as testing SMS broadcasts (30 user numbers and one airline), shows that the announcement application provides information through voice data recording and broadcasts SMS to passengers and families. Testing was carried out at Indonesian Pioner airport for 30 passengers for a week. The percentage of Ease of obtaining test voice information was 81.98%, and the percentage of Ease of obtaining broadcast SMS information was 83.1%. Implementing the announcement is feasible at pioneer airports with a percentage of 82.54%.

New Combinatorial Insights for Monotone Apportionment

The apportionment problem constitutes a fundamental problem in democratic societies: How to distribute a fixed number of seats among a set of states in proportion to the states’ populations? This—seemingly simple—task has led to a rich literature and has become well known in the context of the US House of Representatives. In this paper, we connect the design of monotone apportionment methods to classic problems from discrete geometry and combinatorial optimization and explore the extent to which randomization can enhance proportionality. We first focus on the well-studied family of stationary divisor methods, which satisfy the strong population monotonicity property, and show that this family produces only a slightly superlinear number of different outputs as a function of the number of states. While our upper and lower bounds leave a small gap, we show that—surprisingly—closing this gap would solve a long-standing open problem from discrete geometry, known as the complexity of k-levels in line arrangements. The main downside of divisor methods is their violation of the quota axiom, i.e., every state should receive ⌊qi⌋ or ⌈qi⌉ seats, where qi is the proportional share of the state. As we show that randomizing over divisor methods can only partially overcome this issue, we propose a relaxed version of divisor methods in which the total number of seats may slightly deviate from the house size. By randomizing over these methods, we can simultaneously satisfy population monotonicity, quota, and ex-ante proportionality. Finally, we turn our attention to quota-compliant methods that are house-monotone, i.e., no state may lose a seat when the house size is increased. We provide a polyhedral characterization based on network flows, which implies a simple description of all ex-ante proportional randomized methods that are house-monotone and quota-compliant. Kidney stones are primarily crystals formed from ion oversaturation in urine. Currently, the diagnosis of kidney stones involves experienced professionals manually interpreting images of urinary crystals under a microscope. In recent years, with the rapid development of artificial intelligence (AI), many researchers have proposed various methods to assist in diagnosis, hoping to improve medical quality. This study explored the application of AI in the medical field, aiming to automate the interpretation of urinary crystal images, such as red blood corpuscles (RBCs), calcium oxalate monohydrate (COM), calcium oxalate dihydrate octahedron (COD-OCT), calcium oxalate dihydrate dodecahedron (COD-DOD), debris, and others, and to use image processing techniques to classify urinary crystal types. This research is expected to assist medical personnel in quickly determining the categories of urinary crystals. Ultimately, our model achieved an F1-score of 0.93, indicating that this architecture has significant potential. © 2024 Copyright ACM.

Following recent advances on parameterized hypercomplex multiplication [21], we explore the usefulness of hypercomplex convolutions and deconvolutions in a document labeling task. We show that the proposed Hypercomplex Generative Adversarial Networks achieve excellent results while requiring significantly less independent parameters than real-valued models. © 2022, Springer Nature Switzerland AG.

Fermented foods, such as kombucha, have gained popularity due to their rich content of beneficial bioactive compounds. Papaya leaves, known for their abundance of bioactive substances such as alkaloids, flavonoids, and phenolic compounds, have garnered attention for their potential to improve immune function by neutralising free radicals. This study investigates the suitability of papaya leaves and petioles as substrates for kombucha fermentation. Our findings indicate that papaya leaves and petioles were suitable substrates for kombucha fermentation, indicated by the SCOBY mass accumulation during fermentation. The gradual decrease in brix and pH values over a 14 days fermentation period corresponds to sugar metabolism and the accumulation of acetic acid. Additionally, fermentation increases total polyphenol (PPK: 250.07 ± 3.00 mg/L, PLK: 330.87 ± 15.56 mg/L) and total flavonoid content (PPK: 26.67 ± 6.31 mg/L, PLK: 34.44 ± 5.55 mg/L). Moreover, papaya-derived kombucha demonstrates enhanced antibacterial properties and inhibits ⍺-amylase linked to diabetes. Sensory evaluations confirm that papaya kombucha achieves a well-balanced and appealing sensory profile, catering to diverse palates. In summary, papaya leaves and petiole exhibit promise as substrates for kombucha production, offering elevated nutritional value and potential health advantages. © 2024 institute of Food, science and Technology (IFSTTF).
