Hitting two birds with one stone: The unforeseen consequences of nested gene knockouts in Caenorhabditis elegans

Nested genes represent an intriguing form of non-random genomic organization in which the boundaries of one gene are fully contained within another, longer host gene. The C. elegans genome contains over 10,000 nested genes, 92% of which are ncRNAs, which occur inside 16% of the protein coding gene complement. Host genes are longer than non-host coding genes, owing to their longer and more numerous introns. Indel alleles are available for nearly all of these host genes that simultaneously alter the nested gene, raising the possibility of nested gene disruption contributing to phenotypes that might be attributed to the host gene. Such dual-knockouts could represent a source of misinterpretation about host gene function. Dual-knockouts might also provide a novel source of synthetic phenotypes that reveal the functional effects of ncRNA genes, whereby the host gene disruption acts as a perturbed genetic background to help unmask ncRNA phenotypes.

High blood pressure is a highly heritable and modifiable risk factor for cardiovascular disease. We report the largest genetic association study of blood pressure traits (systolic, diastolic and pulse pressure) to date in over 1 million people of European ancestry. We identify 535 novel blood pressure loci that not only offer new biological insights into blood pressure regulation but also highlight shared genetic architecture between blood pressure and lifestyle exposures. Our findings identify new biological pathways for blood pressure regulation with potential for improved cardiovascular disease prevention in the future.

In this paper we explore the application of a novel data collection scheme for multi-sensory information to the question of whether different sensory domains tend to show similar relations between objects (along with some unique variance). Our analyses-hierarchical clustering, MDS mapping, and other comparisons between sensory domains-support the existence of common representational schemes for food items in the olfactory, taste, visual, and tactile domains. We further show that the similarity within different sensory domains is a predictor for Rosch (1975) typicality measures. We also use the relative importance of sensory domains to predict the overall similarity between pairs of words, and compare subjective similarities to objective similarities based on physical sensory properties of the foods, showing a reasonable match. © 2014 Proceedings of the 36th Annual Meeting of the Cognitive Science Society, CogSci 2014. Young people's social practices online and use of digital media have been conceptualized both as an opportunity and a threat to society. While the mass media has often focused on moral panic related to dramatic examples of social media-related harm to young people, research within Library and Information Science has taken multiple methodological approaches to investigate the ways digital youth navigate and negotiate privacy in online spaces. Findings from such studies offer insight into youth cultures and technology design that supports online privacy. This panel will demonstrate a variety of projects and approaches that have been taken to investigate privacy issues related to young people's online interactions and practices, and the social and design implications of emerging findings for our understanding of young people and online privacy. We will also lead audience members in an interactive brainstorming session centered around various research scenarios, asking participants to consider the strengths and weaknesses of specific research questions and methods in addressing adults’ concerns related to youth and online privacy. We will conclude with an interactive discussion of recommended future research directions for enhancing our understanding of young people's privacy practices.

Background: Decades of steady improvements in life expectancy in Europe slowed down from around 2011, well before the COVID-19 pandemic, for reasons which remain disputed. We aimed to assess how changes in risk factors and cause-specific death rates in different European countries related to changes in life expectancy in those countries before and during the COVID-19 pandemic. Methods: We used data and methods from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to compare changes in life expectancy at birth, causes of death, and population exposure to risk factors in 16 European Economic Area countries (Austria, Belgium, Denmark, Finland, France, Germany, Greece, Iceland, Ireland, Italy, Luxembourg, the Netherlands, Norway, Portugal, Spain, and Sweden) and the four UK nations (England, Northern Ireland, Scotland, and Wales) for three time periods: 1990–2011, 2011–19, and 2019–21. Changes in life expectancy and causes of death were estimated with an established life expectancy cause-specific decomposition method, and compared with summary exposure values of risk factors for the major causes of death influencing life expectancy. Findings: All countries showed mean annual improvements in life expectancy in both 1990–2011 (overall mean 0·23 years [95% uncertainty interval [UI] 0·23 to 0·24]) and 2011–19 (overall mean 0·15 years [0·13 to 0·16]). The rate of improvement was lower in 2011–19 than in 1990–2011 in all countries except for Norway, where the mean annual increase in life expectancy rose from 0·21 years (95% UI 0·20 to 0·22) in 1990–2011 to 0·23 years (0·21 to 0·26) in 2011–19 (difference of 0·03 years). In other countries, the difference in mean annual improvement between these periods ranged from –0·01 years in Iceland (0·19 years [95% UI 0·16 to 0·21] vs 0·18 years [0·09 to 0·26]), to –0·18 years in England (0·25 years [0·24 to 0·25] vs 0·07 years [0·06 to 0·08]). In an attempt to assist proctors to prevent test takers from academic dishonesty in remotely administrated exams, this study investigated the ability of test takers' behaviors during online assessments to predict their cheating decisions. Specifically, this experimental study focused on the role of students' time delay and certainty rating during lab based online testing sessions. The analysis of hierarchical logistic regression indicated that not only time delay but also certainty rating had significantly statistical relation to test takers' cheating decisions. The critical infrastructures of a nation including the power grid and the communication network are highly interdependent. Recognizing the need for a deeper understanding of the interdependency in a multi-layered network, significant efforts have been made by the research community in the last few years to achieve this goal. Accordingly a number of models have been proposed and analyzed. Unfortunately, most of the models are over simplified and, as such, they fail to capture the complex interdependency that exists between entities of the power grid and the communication networks involving a combination of conjunctive and disjunctive relations. To overcome the limitations of existing models, we propose a new model that is able to capture such complex interdependency relations. Utilizing this model, we provide techniques to identify the K most 'vulnerable' nodes of an interdependent network. We show that the problem can be solved in polynomial time in some special cases, whereas for some others, the problem is NP-complete. We establish that this problem is equivalent to computation of a fixed point of a multilayered network system and we provide a technique for its computation utilizing Integer Linear programming. Finally, we evaluate the efficacy of our technique using real data collected from the power grid and the communication network that span the Maricopa County of Arizona. © 2014 IEEE.

High Performance computing (HPC) aggregates computing power in order to solve large and complex problems in different knowledge areas. Nowadays, HPC users can utilize virtualized infrastructures as a low-cost alternative to deploy their applications. However, virtualization brings some challenges for HPC, specially in regard to overhead caused by hyper visors. In this work, our main goal is to analyze the performance of two hyper visors (KVM and Virtual Box) under HPC activities, considering full virtualization, and Para virtualization approaches. We used the HPC Challenge Benchmark (HPCC) to evaluate processor, RAM, inter-process communication and network communication performance. Our results show KVM in Para virtualization mode has a similar performance of a native cluster.
