Simulation of turbulence mixing in the atmosphere boundary layer and analysis of fractal dimension

In the first part of this paper, a flow model for numerical simulation of turbulent parameters in atmospheric boundary layer (ABL), based on finite volume method and large-eddy simulation is introduced. This model consists of balance equations for mass, momentum and energy (for potential temperature) equations. The Lagrangian dynamic model of Smagorinsky with restriction of size for the coefficient Cs was used for sub grid turbulent viscosity. The second part of this paper is devoted to the numerical aspects of flow model using proper orthogonal decomposition (POD) method. In the final part of this paper, results from numerical studies on flow in ABL for the neutral and stable case and analysis of fractal dimensions are presented. These results constitute important tests for the assessment of the predictive capacity for the stratified flow model in hand. ImaCalc program was used to study the fractal parameter and structure functions. We calculated the maximum value of fractal dimension D selected among all of the velocity intensity and vorticity modules which was a good indicator of flow's complexity. The data of evolution of D(t) in the middle section of the domain of the multifractal spectra along the main downstream axis were also calculated. The reduction in the maximum value of the fractal dimension for intermediate velocity and vorticity values is consistent with laboratory experiments and with wind wane measurements.

Methotrexate elicits pro-respiratory and anti-growth effects by promoting AMPK signaling

One-carbon metabolism fuels the high demand of cancer cells for nucleotides and other building blocks needed for increased proliferation. Although inhibitors of this pathway are widely used to treat many cancers, their global impact on anabolic and catabolic processes remains unclear. Using a combination of real-time bioenergetics assays and metabolomics approaches, we investigated the global effects of methotrexate on cellular metabolism. We show that methotrexate treatment increases the intracellular concentration of the metabolite AICAR, resulting in AMPK activation. Methotrexate-induced AMPK activation leads to decreased one-carbon metabolism gene expression and cellular proliferation as well as increased global bioenergetic capacity. The anti-proliferative and pro-respiratory effects of methotrexate are AMPK-dependent, as cells with reduced AMPK activity are less affected by methotrexate treatment. Conversely, the combination of methotrexate with the AMPK activator, phenformin, potentiates its anti-proliferative activity in cancer cells. These data highlight a reciprocal effect of methotrexate on anabolic and catabolic processes and implicate AMPK activation as a metabolic determinant of methotrexate response.

In the previous publication [1], we described a method for minification of a system composed of dynamic modules under the "closed-world" assumption. Although the presented approach is sound in theory, there are complications with reliably using it in practice. Such complications arise from the peculiarities of existing real-world linkers (symbol elimination relies on proper and consistent implementation of the plugin infrastructure and the--gc sect ions switch). The above approach is also imprecise in that it keeps more symbols than strictly necessary due to not discriminating between build time only dependencies (from configure tests) and regular ones. This design limitation as well as the reliability issues are addressed in the new approach presented in this paper.

The paper presents the possibilities of using vibroacoustic methods in the study of the technical condition of designed multimedia mobile scenes. In particular, the possibility of implementing modal analysis methods in modelling and diagnostic research process has been presented. The use of virtual methods enables diagnostic tests both at the design stage and at the stage of normal operation, whereas modal methods help to explain the nature of the work of the element under investigation.

Aim Premix insulin is commonly used in some regions of the world, despite the higher risk of hypoglycaemia and weight gain compared with basal insulin, based on the premise that it offers a simplified insulin regimen. iGlarLixi is a once-daily titratable fixed-ratio formulation that combines basal insulin glargine 100 units/mL (iGlar) and the GLP-1 RA, lixisenatide, which offers a single-injection option for treatment intensification, with improved HbA1c reductions, similar hypoglycaemia risk and more favourable bodyweight profiles over iGlar alone. This randomized controlled study directly compares, for the first time, treatment intensification with iGlarLixi versus premix insulin analogue biphasic insulin aspart 30 (BIAsp 30) in adults with T2D inadequately controlled on basal insulin in combination with one or two oral antihyperglycaemic drugs. Materials and Methods This was an open-label, active-controlled, comparative, parallel-group, multicentre, phase 3b study. In total, 887 adults with T2D uncontrolled on basal insulin were randomized to switch to either iGlarLixi once daily, or BIAsp 30 twice daily, for 26 weeks. Results Overall, 887 participants were enrolled (mean age 59.8 years, 50.2% female) from 89 centres in 17 countries. At baseline, 65.6% had a duration of T2D of 10 years or longer, and the mean HbA1c at baseline was 8.6%. Conclusions The study directly compared the efficacy and safety of iGlarLixi versus BIAsp 30 in people with T2D uncontrolled on basal insulin and one or more oral antihyperglycaemic agents. These results provide robust clinical data that may inform clinicians in their therapeutic management of people with T2D uncontrolled on basal insulin requiring additional therapy.

Some problems connected to vortex methods development for 2D incompressible flow simulation around airfoils are discussed. In the numerical schemes and algorithms which are normally used in the vortex methods, the airfoil is approximated by a polygon consists of rectilinear panels, and the the vortex sheet intensity, which is placed on the airfoil surface line and simulates the airfoil's influence the flow, is assumed to be piecewise-constant or piecewise-linear function. The most accurate mathematical models and numerical algorithms, based on such approaches, provide the estimation for the order of the error between O(h) and O(h(2)) for the average vortex sheet intensity over the panels (his the maximal panel length). In the present research a new approach is developed, where the vortex sheet intensity is assumed to be discontinuous piecewise-linear or piecewise-quadratic function and the curvature of the panels is taken into account. In order to compute the coefficients of the main system of the algebraic equations, the least squares method is used instead of commonly used no-through or no-slip conditions at separate control points or over the panels in the average. It is shown, that the developed approach is much more accurate in comparison with previously known ones: for some test problems (flows around a circular airfoil, an elliptical airfoil and Zhukovsky airfoil) it permits to obtain numerical solution which has error of order O(h(4)).

The article describes an open source C++ code we have developed for 2D incompressible flow simulation using vortex methods. The code has a modular structure, and allows users to simulate flows around airfoils (also, around a system of airfoils) and compute unsteady hydrodynamic loads acting the airfoils. It is also possible to simulate hydroelastic regimes of airfoil motion in the flow by using weakly and strongly coupling strategies. The software implements well known algorithms and also original numerical schemes developed by the authors, thereby significantly increasing the accuracy of simulations compared to traditional algorithms. The software makes it possible to run simulations in parallel mode;OpenMP and MPI technologies are supported. The VM2D source code is available on GitHub under GNU GPL license.

Augmented T cell function leading to host damage in autoimmunity is supported by metabolic dysregulation, making targeting immunometabolism an attractive therapeutic avenue. Canagliflozin, a type 2 diabetes drug, is a sodium glucose co-transporter 2 (SGLT2) inhibitor with known off-target effects on glutamate dehydrogenase and complex I. However, the effects of SGLT2 inhibitors on human T cell function have not been extensively explored. Here, we show that canagliflozin-treated T cells are compromised in their ability to activate, proliferate, and initiate effector functions. Canagliflozin inhibits T cell receptor signaling, impacting on ERK and mTORC1 activity, concomitantly associated with reduced c-Myc. Compromised c-Myc levels were encapsulated by a failure to engage translational machinery resulting in impaired metabolic protein and solute carrier production among others. Importantly, canagliflozin-treated T cells derived from patients with autoimmune disorders impaired their effector function. Taken together, our work highlights a potential therapeutic avenue for repurposing canagliflozin as an intervention for T cell-mediated autoimmunity.

A component model enabling to construct new software components from existing ones dynamically, at runtime, without their bytecodes generation is presented with supporting it software framework. The framework is implemented using JavaBeans component model, but is aimed to eliminate its drawback - the inability to create user-defined components without bytecodes generation. To construct user-defined component dynamically, a composed prototype object is built using predefined (hardcoded and/or composed) component instances;that prototype object can provide functionality required and can be transformed at runtime into a new component (instantiable type) whose instances are able to provide the same functionality, but more efficiently. The prototype object is composed using meta-components - the framework provided components to produce user-defined components dynamically.

In the literature, it is well known that there is a bidirectional causality between economic growth and energy consumption. This is why it is crucial to forecast energy consumption. In this study, four deep learning models, i.e., Long Short-Term Memory (LSTM), stacked LSTM, bidirectional LSTM, and Gated Recurrent Unit (GRU), were used to forecast energy consumption in Brazil, Canada, and France. After a training test period, the performance evaluation criterion, i.e., R2, mean square error, root mean square error, mean absolute error, and mean absolute percentage error, was performed for the performance measure. It showed that GRU is the best model for Canada and France, while LSTM is the best model for Brazil. Therefore, the energy consumption prediction was made for the 12 months of the year 2017 using LSTM for Brazil and GRU for Canada and France. Based on the selected model, it was projected that the energy consumption in Brazil was 38 597.14-38 092.88, 63 900-4 800 000 GWh in Canada, and 50 999.72-32 747.01 GWh in France in 2017. The projected consumption in Canada was very high due to the country's higher industrialization. The results obtained in this study confirmed that the nature of energy production will impact the complexity of the deep learning model.
