Restriction of YWHAB-mediated YAP cytoplasmic retention is a novel mechanism underlying stemness maintenance and chemoresistance in ovarian cancer peritoneal metastasis

Ovarian cancer (OC) peritoneal metastasis (OCPM) is a major cause of high mortality of OC, in which cancer cells incubated in ascites evolve various mechanisms to survive. Hippo/YAP singling plays multiple roles in carcinogenesis, however, its roles in OCPM have remained elusive. Here, we report that restriction of YWHAB-mediated YAP cytoplasmic retention is a critical mechanism underlying OCPM stemness maintenance. Combined tandem mass tag- and tissue microarray-based proteomic studies revealed YWHAB down-regulation in post-neoadjuvant chemotherapy OCPM tissues, which was confirmed in no-neoadjuvant-chemotherapy-response tissues, isolated OCPM stem cells, and induced cisplatin-resistant cells. Knockdown of YWHAB promoted stemness and resistance in parental complete or near-complete primary OCPM and OVCAR3 cells in vitro and in vivo . Mechanistic study showed that YWHAB directly bound to YAP and promoted YAP cytoplasmic retention and thus YWHAB restriction promoted YAP activity and stemness in OCPM in the cells in which the Hippo/YAP signaling was constitutively activated by overloaded constitutively active YAP (YAP5SA), and the effect of YWHAB knockdown was significantly abolished. The SH3 binding domain in YAP is critical for YWHAB-YAP binding. Alteration in the 5mc methylation level in the YWHAB promoter was observed in OCPM stem cells. In summary, our results reveal that restriction of YWHAB-mediated YAP cytoplasmic retention is a critical mechanism underlying OCPM stemness maintenance. Our findings suggest that YAP would be a therapeutic target for suppressing OCPM stemness caused by YWHAB restriction.

Now everything is connected and because of the data explosion, data generation per year will exceed 160ZB in 2025. Non-volatile memory is a key component because store, manage and utilize those data are very important. The NAND flash is an inevitable memory device as a storage and continue to improve its density and performance even it faces many challenges. Breakthrough in computing system is required to manage big data and new non-volatile memory called storage class memory is expected to be one of solutions. There is an opportunity for emerging non-volatile memories to realize SCM. 3D Flash also stretches its performance towards SCM area and competes with emerging memories. Tough challenges continue in coming years.

Background Tau positron emission tomography (PET) is increasingly used in the clinical evaluation of patients and as an outcome measure in Alzheimer’s disease (AD) clinical trials. Due to differences in tracer properties, instrumentation, and methods of analysis, however, tau-PET outcome data cannot currently be meaningfully compared or combined. Here, we tested i) the feasibility of adapting the Centiloid method—an approach originally developed to standardize amyloid PET—to harmonize tau-PET quantification (CenTauRs); ii) the performance of a non-linear mixed model-based approach (Joint Propagation Model) that does not require the use of a reference tracer. Methods Head-to-head tau-PET data (Table 1) was included from two cohorts ([ 18 F]RO948 vs [ 18 F]flortaucipir, n = 37 [BioFINDER-2]; [ 18 F]flortaucipir [Avid A05] vs [ 18 F]MK-6240, n = 15, University of Pittsburgh) in which each participant was scanned with two tau-PET tracers. Standardized uptake value ratio (SUVR) values were calculated using the inferior cerebellar cortex as the reference region. Anchor point data (Table 1) was derived for each tracer using the following criteria: CenTauR-0, cognitively unimpaired (CU), amyloid PET negative (<10 Centiloids); CenTauR-100, amyloid PET positive (>50 Centiloids), typical (temporoparietal) AD pattern on tau-PET visual read, age<65 andMMSE>20. Regions-of-interest (ROIs) included a universal tau-PET ROI—based on the intersection of tracer specific ([ 18 F]flortaucipir, [ 18 F]MK-6240, [ 18 F]PI-2620, [ 18 F]PM-PBB3, [ 18 F]GTP1 and [ 18 F]RO948) masks that had been derived by subtracting average of amyloid-negative CU images from the average AD image—as well as four subregions delineated within this ROI (medial temporal, meta-temporal, temporoparietal and frontal) (Figure 1A). An overview of the adapted Centiloid-like approach and the joint propagation model is shown in Figure 1B. Results High R 2 values were observed between tracers across all ROIs: [ 18 F]RO948

Introduction: The Cognitive Ageing, Nutrition and Neurogenesis trial hypothesizes that a combined intervention with long-chain n-3 polyunsaturated fatty acids (n-3) and cocoa flavan-3-ols (FLAV) will mitigate the cognitive decline anticipated to naturally occur over 1 year in older adults. Methods: In a double-blinded, placebo-controlled parallel design, 259 individuals with mild cognitive impairment or subjective memory impairment were randomized to a control or n-3 FLAV group (1.5 g docosahexaenoic acid + eicosapentaenoic acid and 500 mg n-3 FLAV daily) for 12 months. Cognition was measured at 0, 3, and 12 months. The primary end-point is hippocampus-sensitive cognitive function (e.g., number of false-positives on the Picture Recognition Task of the Cognitive Drug research test battery). Secondary outcomes include additional cognitive measures, brain atrophy and blood flow (assessed by magnetic resonance imaging), vascular function, circulating biomarkers of cardiovascular and cognitive health, gut microflora, red blood cell fatty acid status, and urine flavan-3-ol metabolites. Results: Screening began in 2015, with all baseline visits completed in March 2017. The intervention was finished in March 2018. Discussion: Cognitive Ageing, Nutrition and Neurogenesis aims to identify an effective diet-based intervention to prevent or delay cognitive impairment in cognitively at-risk individuals, which could ultimately contribute to a reduced population burden of dementia. ***: NCT02525198. © 2018

The Technical Program Committee worked very hard to have all manuscripts reviewed before the review deadline. In graphene superlattices, bulk topological currents can lead to long-range charge-neutral flow and non-local resistance near Dirac points. A ballistic version of these phenomena has never been explored. Here, we report transport properties of ballistic graphene superlattices. This allows us to study and exploit giant non-local resistances with a large valley Hall angle without a magnetic field. In the low-Temperature regime, a crossover occurs toward a new state of matter, referred to as a quantum valley Hall state (qVHS), which is an analog of the quantum Hall state without a magnetic field. Furthermore, a non-local resistance plateau, implying rigidity of the qVHS, emerges as a function of magnetic field, and the collapse of this plateau is observed, which is considered as a manifestation of valley/pseudospin magnetism. The alloying of wide-band-gap ZnO and GaN causes band-gap reduction, enabling visible-light-driven photocatalysis for high-efficiency water splitting. However, challenges in single-crystal (GaN)1−x(ZnO)x solid-solution synthesis prevent a better understanding of the optical properties and electronic structures. Here, low-temperature epitaxial growth of (GaN)1−x(ZnO)x thin films with a wide tunability of chemical composition is demonstrated by using a multitarget pulsed-laser-deposition (PLD) system. The phase pure (GaN)1−x(ZnO)x solid solution is obtained by alternately depositing GaN and ZnO with the thickness of each GaN/ZnO pair set within one or two unit cells. The band gap of the solid-solution thin films as a function of systematically controlled chemical composition shows asymmetric bowing with a minimum at approximately 2.0 eV for x = 0.65. Furthermore, a large absorption coefficient (>104cm−1) in the visible-light region is observed. The shape of the absorption edge is not consistent with that of a direct-transition semiconductor. First-principles calculation suggests that this inconsistency originates from localization of the valence-band maximum on N atoms bonded with Zn. A technique for fabricating high-quality epitaxial (GaN)1−x(ZnO)x solid solutions is essential for acquiring a deep understanding of the fundamental properties of this system.

BACKGROUND:Structural brain differences in the thalamus and the cortex have been widely reported in schizophrenia (SCZ) relative to neurotypical controls (NC). Most prior studies examined the thalamus as a whole, single region-of-interest. Additionally, findings in individuals at familial high-risk for schizophrenia (FHR) remain inconclusive. Here, we investigated whether local and network-wide thalamic-related structural alterations vary as a function of familial risk for schizophrenia. METHODS:Structural MRI scans were obtained from 5,197 participants (3,409 NC, 257 FHR, 1,531 SCZ) across 32 cross-sectional samples within the ENIGMA Consortium. Random-effects meta-analyses, and network analyses were conducted on (i) local thalamic alterations (volume estimates of seven thalamic subdivisions), and (ii) network-wide thalamic alterations (thickness and surface-related thalamo-cortical/cortico-cortical co-variation patterns) across groups (NC, FHR, SCZ). RESULTS:Individuals with SCZ showed significantly lower gray matter volume estimates in the anterior, pulvinar, medial, posterior, and ventral thalamic subdivisions compared to NC (q<0.05). FHR did not differ from NC. At the network-wide level, thalamo-cortical co-variations discriminated FHR from NC (q<0.05), with FHR showing intermediate co-variation between SCZ and NC. Cortico-cortical co-variation patterns revealed that SCZ and FHR shared similarly disconnected clustering configurations, distinct from NC (q<0.05). CONCLUSIONS:Results revealed lower thalamic volume estimates in SCZ but not in FHR, hence yielding no evidence of a familial risk trait, whereas thalamo-cortical and cortico-cortical co-variation estimates were associated with familial risk for SCZ These findings suggest that, once the thalamus is parsed into subdivisions, network-wide thalamo-cortical features may identify trait-dependent, neurobiological correlates of genetic risk for SCZ.
