All human diseases involve proteins, yet our current tools to characterize and quantify them are limited. To better elucidate proteins across space, time, and molecular composition, we provide provocative projections for technologies to meet the challenges that protein biology presents. With a broad perspective, we discuss grand opportunities to transition the science of proteomics into a more propulsive enterprise. Extrapolating recent trends, we offer potential futures for a next generation of disruptive approaches to define, quantify and visualize the multiple dimensions of the proteome, thereby transforming our understanding and interactions with human disease in the coming decade. © 2021, CC0.

Background Diarrhoeal diseases claim more than 1 million lives annually and are a leading cause of death in children younger than 5 years. Comprehensive global estimates of the diarrhoeal disease burden for specific age groups of children younger than 5 years are scarce, and the burden in children older than 5 years and in adults is also understudied. We used results from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to assess the burden of, and trends in, diarrhoeal diseases overall and attributable to 13 pathogens, as well as the contributions of associated risk factors, in children and adults in 204 countries and territories from 1990 to 2021. Methods We used the Cause of Death Ensemble modelling strategy to analyse vital registration data, verbal autopsy data, mortality surveillance data, and minimally invasive tissue sampling data. We used DisMod-MR (version 2.1), a Bayesian meta-regression tool, to analyse incidence and prevalence data identified via systematic reviews, population-based surveys, and claims and inpatient data. We calculated diarrhoeal disability-adjusted life-years (DALYs) as the sum of years of life lost (YLLs) and years lived with disability (YLDs) for each location, year, and age-sex group. For aetiology estimation, we used a counterfactual approach to quantify population-attributable fractions (PAFs). Additionally, we estimated the diarrhoeal disease burden attributable to the independent effects of risk factors using the comparative risk assessment framework. Findings In 2021, diarrhoeal diseases caused an estimated 1.17 million (95% uncertainty interval 0.793-1.62) deaths globally, representing a 60.3% (50.6-69.0) decrease since 1990 (2.93 million [2.31-3.73] deaths). The most pronounced decline was in children younger than 5 years, with a 79.2% (72.4-84.6) decrease in diarrhoeal deaths. The modeling and simulation community in aviation is in search of standardization in simulation scenario definition. While the system Entity Structure (SES) ontology promises an effective formal means of domain modeling, computational representation is still a key challenge to tackle for fostering wide-acceptance and standardization. This paper presents an eXtended Markup Language (XML) based approach. It first gives the relational specification and the schema definition of the SES ontology. Mobility-on-demand versus fixed-route transit systems: An evaluation of traveler preferences in low-income communities

Emerging transportation technologies, such as ride-hailing and autonomous vehicles, are disrupting the transportation sector and transforming public transit. Some transit observers envision future public transit to be integrated transit systems with fixed-route services running along major corridors and on-demand ridesharing services covering lower-density areas. A switch from a conventional fixed-route service model to this kind of integrated mobility-on-demand transit system, however, may elicit varied responses from local residents. This paper evaluates traveler preferences for a proposed integrated mobility-on-demand transit system versus the existing fixed-route system, with a particular focus on disadvantaged travelers. We conducted a survey in two low-resource communities in the United States, namely, Detroit and Ypsilanti, Michigan. A majority of survey respondents preferred a mobility-on-demand transit system over a fixed-route one. Based on ordered logit model outputs, we found a stronger preference for mobility-on-demand transit among males, college graduates, individuals who have never heard of or used ride-hailing before, and individuals who currently receive inferior transit services. By contrast, preferences varied little by age, income, race, or disability status. The most important benefit of a mobility-on-demand transit system perceived by the survey respondents is enhanced transit accessibility to different destinations, whereas their major concerns include the need to actively request rides, possible transit-fare increases, and potential technological failures. Addressing the concerns of female riders and accommodating the needs of less technology-proficient individuals should be major priorities for transit agencies that are considering mobility-on-demand initiatives. We outline emerging opportunities and challenges to enhance the utility of AI for scientific discovery. The distinct goals of AI for industry versus the goals of AI for science create tension between identifying patterns in data versus discovering patterns in the world from data. If we address the fundamental challenges associated with "bridging the gap" between domain-driven scientific models and data-driven AI learning machines, then we expect that these AI models can transform hypothesis generation, scientific discovery, and the scientific process itself. © 2021, CC BY.

Multipath transmission scheduling in millimeter wave cloud radio access networks

Millimeter wave (mmWave) communications provide great potential for next-generation cellular networks to meet the demands of fast-growing mobile data traffic with plentiful spectrum available. However, in a mmWave cellular system, the shadowing and blockage effects lead to the intermittent connectivity, and the handovers are more frequent. This paper investigates an "all-mmWave" cloud radio access network (cloud-RAN), in which both the fronthaul and the radio access links operate at mmWave. To address the intermittent transmissions, we allow the mobile users (MUs) to establish multiple connections to the central unit over the remote radio heads (RRHs). Specifically, we propose a multipath transmission framework by leveraging the "all-mmWave" cloud-RAN architecture, which makes decisions of the RRH association and the packet transmission scheduling according to the time-varying network statistics, such that a MU experiences the minimum queueing delay and packet drops. The joint RRH association and transmission scheduling problem is formulated as a Markov decision process (MDP). Due to the problem size, a low-complexity online learning scheme is put forward, which requires no a priori statistic information of network dynamics. Simulations show that our proposed scheme outperforms the state-of-art baselines, in terms of average queue length and average packet dropping rate. Copyright © 2018, The Authors. Increasingly more mobile Internet traffic is produced which contains ample personal information related to user mobility and website browsing behavior. Prior research has attempted to recommend friends based on Global Position system (GPS) in location based social networks (LBSN). However, the study of friend recommendation in general social network according to position from the base station is relatively understudied. This paper introduces a novel feature set extracted from mobile Internet traffic according to base station location and Uniform Resource Locator (URL). We train classification models using these features to predict friendship between pairs of Weibo users. Results show that both base station location and URL when acted alone can already effectively reflect friendships even in general social network. We further show that by fusing the two features together, the model obtains even better performance. Finally, we demonstrate that the location and URL features can improve prediction performance than only using the common friends.

A joint learning and communications framework for federated learning over wireless networks

In this paper, the problem of training federated learning (FL) algorithms over a realistic wireless network is studied. In particular, in the considered model, wireless users execute an FL algorithm while training their local FL models using their own data and transmitting the trained local FL models to a base station (BS) that will generate a global FL model and send it back to the users. Since all training parameters are transmitted over wireless links, the quality of the training will be affected by wireless factors such as packet errors and the availability of wireless resources. Meanwhile, due to the limited wireless bandwidth, the BS must select an appropriate subset of users to execute the FL algorithm so as to build a global FL model accurately. This joint learning, wireless resource allocation, and user selection problem is formulated as an optimization problem whose goal is to minimize an FL loss function that captures the performance of the FL algorithm. To address this problem, a closed-form expression for the expected convergence rate of the FL algorithm is first derived to quantify the impact of wireless factors on FL. Then, based on the expected convergence rate of the FL algorithm, the optimal transmit power for each user is derived, under a given user selection and uplink resource block (RB) allocation scheme. Finally, the user selection and uplink RB allocation is optimized so as to minimize the FL loss function. Simulation results show that the proposed joint federated learning and communication framework can reduce the FL loss function value by up to 10% and 16%, respectively, compared to: 1) An optimal user selection algorithm with random resource allocation and 2) a standard FL algorithm with random user selection and resource allocation. Major depressive disorder (MDD) is highly heterogeneous, posing challenges for effective treatment due to complex interactions between clinical symptoms and neurobiological features. To address this, we apply contrastive principal-component analysis to fMRI-based resting-state functional connectivity, isolating disorder-specific variations by contrasting data from 233 MDD patients and 285 healthy controls. Subsequently, we use sparse canonical correlation analysis to identify two significant dimensions linking distinct brain circuits with clinical profiles. One dimension relates to an internalizing-externalizing symptom spectrum involving visual and limbic networks and is associated with cognitive task reaction times. The other dimension, linked to personality traits protective against depression (e.g., extraversion), is driven by dorsal attention network connections and correlates with cognitive control and psychomotor performance. This approach illuminates stable symptom dimensions and their neurophysiological underpinnings, aiding in precision phenotyping for MDD and supporting the development of targeted, individualized therapeutic strategies for mental health care.
