Acinetobacter baumannii is a major pathogen of nosocomial meningitis and ventriculitis. Due to very limited antibiotic treatment options, polymyxins are often used as a last-line therapy. To optimise polymyxin use in the intraventricular environment, cerebrospinal fluid (CSF) proteomics was employed to investigate host-pathogen-polymyxin interactions in a 69-year-old patient with multidrug-resistant A. baumannii ventriculitis treated with a combination of intrathecal (ITH;50,000 IU q24h/q48h), intraventricular (IVT;50,000 IU q48h), and intravenous (500,000 IU, q12h) polymyxin B. CSF was collected before the first ITH dose in the ICU (0 h) and at 24 h, Day 7 and Day 26. The proteome was quantified at each time point and proteins with Qvalue <0.05 and fold change >1.2 were considered differentially expressed. Within 24 h of ITH/IVT polymyxin B administration, the innate immune system and neuroimmunity were highly active, evidenced by up-regulation of various pathways related to pathogen invasion, endocytosis and neutrophil degranulation. Blood-brain barrier impairment had worsened at 24 h but signs of repair were evident on Day 7 and Day 26. This is the first CSF proteomic study with polymyxins. Nanometer thin single-walled carbon nanotube (CNT) films collected from the aerosol chemical deposition reactors have gathered attention for their promising applications. Densification of these pristine films provides an important way to manipulate the mechanical, electronic, and optical properties. To elucidate the underlying microstructural level restructuring, which is ultimately responsible for the change in properties, we perform large scale vector-based mesoscopic distinct element method simulations in conjunction with electron microscopy and spectroscopic ellipsometry characterization of pristine and densified films by drop-cast volatile liquid processing. Matching the microscopy observations, pristine CNT films with finite thickness are modeled as self-assembled CNT networks comprising entangled dendritic bundles with branches extending down to individual CNTs. Simulations of the film under uniaxial compression uncover an ultra-soft densification regime extending to a ~75% strain, which is likely accessible with the surface tensional forces arising from liquid surface tension during the evaporation. When removing the loads, the pre-compressed samples evolve into homogeneously densified films with thickness values depending on both the pre-compression level and the sample microstructure. The significant reduction in thickness, confirmed by our spectroscopic ellipsometry, is attributed to the underlying structural changes occurring at the 100 nm scale, including the zipping of the thinnest dendritic branches. A total of 8218 pelagic microplastic samples from the world’s oceans were synthesized to create a dataset composed of raw, calibrated, processed, and gridded data which are made available to the public. The raw microplastic abundance data were obtained by different research projects using surface net tows or continuous seawater intake. Fibrous microplastics were removed from the calibrated dataset. Microplastic abundance which fluctuates due to vertical mixing under different oceanic conditions was standardized. An optimum interpolation method was used to create the gridded data;in total, there were 24.4 trillion pieces (8.2 × 10⁴ ~ 57.8 × 10⁴ tons) of microplastics in the world’s upper oceans. © The Author(s) 2021.

A palladium-based (Pd-based) core@shell catalyst can be modified to achieve the desired oxygen adsorption properties by selecting an appropriate core composition, surface alloying, and compressive strain. Herein, we present the effects of compressive strain, core composition, and surface alloying in Pd3Ni@PdIr(111), Pd3CuNi@PdIr(111), and Pd3Cu@PdIr(111) alloy-core@alloy-shell catalysts on dioxygen adsorption. Using experimental lattice parameters for the unstrained catalysts,-1% to-5%, strain was systematically introduced. The calculated dioxygen-adsorption energies for the surface Pd and surface Ir atoms reveal that the Pd3CuNi@PdIr catalyst has the lowest dioxygen-adsorption energy at a given compressive strain. Bader charge calculations show that the Pd3CuNi@PdIr catalyst surface is the most charge depleted. The d-band model displays an intermediate d-band center downshift for the surface Pd atoms, and the highest downshift for the surface Ir atoms. Due to synergism between charge depletion, the d-band center shift, and the surface alloy effect, the Pd3CuNi@PdIr catalyst has the lowest dioxygen-adsorption energy. The relationship between the experimentally obtained catalyst-surface mass activity and the theoretically calculated d-band center of the surface Pd and the surface Ir is volcano shaped, with the Pd3CuNi@PdIr catalyst at the apex of the volcano. The catalytic activities of these catalysts were observed to follow the order: Pd3CuNi@PdIr >Pd3Cu@PdIr >Pd3Ni@PdIr. This work sheds light on the importance of ligand and strain effects, as well as surface alloying for the fine-tuning of alloy-core@alloy-shell-catalysts during the rational design of catalysts from first principles. Radiation-induced dermatitis (RID) remains a significant and challenging side effect of radiotherapy, with few effective topical interventions. This study investigates ON101 as a therapeutic candidate for RID, focusing on its modulation of the Keap1/Nrf2 signaling pathway. Using a murine model of radiation-induced skin injury, topical application of ON101 substantially reduced skin damage, as evidenced by decreased wound severity scores, preserved body weight, and improved histopathological outcomes. Histological analyses using H&E and Masson's trichrome staining revealed that ON101 and its active component, PA-F4, reversed radiation-associated dermal thickening and fibrosis, thus restoring skin architecture and elasticity. RNA sequencing demonstrated that ON101 treatment downregulated Keap1 expression, facilitating nuclear translocation and activation of Nrf2, a central regulator of antioxidant responses. Consistently, irradiated human keratinocytes (HaCaT) and dermal fibroblasts (HDFs) treated with ON101 exhibited significantly decreased levels of oxidative stress markers and pro-inflammatory cytokines, confirming its combined antioxidant and anti-inflammatory effects. Moreover, ON101 enhanced the expression of key genes involved in DNA damage repair and skin regeneration, including EGFR, VIM, and CTNNB1, highlighting its regenerative potential. Mechanistically, the study identified increased p62 and nuclear Nrf2 levels following ON101 treatment, confirming activation of the p62-Keap1-Nrf2 axis-a pivotal pathway governing cellular defense mechanisms against oxidative damage. Collectively, these results suggest that ON101 provides comprehensive protection against RID by modulating oxidative stress, reducing inflammation, and promoting tissue repair. Through these targeted molecular mechanisms, ON101 emerges as a promising topical therapeutic agent for managing radiation-induced cutaneous toxicity in clinical settings.

Bridging particle deformability and collective response in soft solids

Soft, amorphous solids such as tissues, foams, and emulsions are composed of deformable particles. However, the effect of single-particle deformability on the collective behavior of soft solids is still poorly understood. We perform numerical simulations of two-dimensional jammed packings of explicitly deformable particles to study the mechanical response of model soft solids. We find that jammed packings of deformable particles with excess shape degrees of freedom possess low-frequency quartic vibrational modes that stabilize the packings even though they possess fewer interparticle contacts than the nominal isostatic value. Adding intra-particle constraints can rigidify the particles, but these particles undergo a buckling transition and gain an effective shape degree of freedom when their preferred perimeter is above a threshold value. We find that the mechanical response of jammed packings of deformable particles with shape degrees of freedom differs significantly from that of jammed packings of rigid-shape particles, which emphasizes the importance of particle deformability in modelling soft solids. © 2020, CC BY.

Void distributions reveal structural link between jammed packings and protein cores

Dense packing of hydrophobic residues in the cores of globular proteins determines their stability. Recently, we have shown that protein cores possess packing fraction ϕ≈0.56, which is the same as dense, random packing of amino-acid-shaped particles. In this article, we compare the structural properties of protein cores and jammed packings of amino-acid-shaped particles in much greater depth by measuring their local and connected void regions. We find that the distributions of surface Voronoi cell volumes and local porosities obey similar statistics in both systems. We also measure the probability that accessible, connected void regions percolate as a function of the size of a spherical probe particle and show that both systems possess the same critical probe size. We measure the critical exponent τ that characterizes the size distribution of connected void clusters at the onset of percolation. We find that the cluster size statistics are similar for void percolation in packings of amino-acid-shaped particles and randomly placed spheres, but different from that for void percolation in jammed sphere packings. We propose that the connected void regions are a defining structural feature of proteins and can be used to differentiate experimentally observed proteins from decoy structures that are generated using computational protein design software. This work emphasizes that jammed packings of amino-acid-shaped particles can serve as structural and mechanical analogs of protein cores, and could therefore be useful in modeling the response of protein cores to cavity-expanding and -reducing mutations.

Previous research has explored theories regarding the vertical transmission of human papillomavirus (HPV) infection and its association with adverse pregnancy and perinatal outcomes. However, the impact of maternal HPV infection on congenital anomalies (CAs) in offspring remains relatively understudied. We conducted a population-based cohort study linking the Taiwan Birth Registry, Taiwan Death Registry, and National Health Insurance Research Database, in which newborns born in Taiwan between 2009 and 2015 were included. We established a maternal HPV infection cohort comprising 37 807 newborns and matched them with a comparison group of 151 228 newborns at a 1:4 ratio based on index year, age, and sex. The study examined a composite outcome and subgroups of different types of congenital malformations. Differences in cumulative incidence of CAs were assessed using Kaplan–Meier curves and log-rank tests. Adjusted hazard ratios (aHRs) were estimated using Cox proportional hazard regressions. No significant association was found between HPV infection and the broad spectrum of CAs (aHR: 1.04, 95% confidence interval [CI]: 0.98–1.10;log-rank test p = 0.14). However, we observed a 19% increased risk of musculoskeletal CAs in the maternal HPV infection group (aHR: 1.19;95% CI: 1.05–1.34) compared to those without maternal HPV exposure. Other factors, including the type of HPV (aHR: 0.65;95% CI: 0.16–2.63), the timing of exposure (during or before pregnancy), and maternal age (aHR for <30 years: 1.02, 95% CI: 0.94–1.1;aHR for 30–39 years: 1.05, 95% CI: 0.99–1.11;aHR for ≥40 years: 0.88, 95% CI: 0.67–1.17), did not significantly affect the risk for any CA. In conclusion, gestation detection of HPV infection was associated with musculoskeletal CAs but not other major CAs. Prospective studies are warranted to elucidate the necessity of prenatal screening in populations at risk. © 2024 Wiley Periodicals LLC.
