Cellular senescence,a persistent state of cell cycle arrest,accumulates in aged organisms,contributes to tissue dysfunction,and drives age-related *** clearance of senescent cells is expected to decrease chronic,low-grade inflammation and improve tissue repair capacity,thus attenuating the decline of physical function in aged ***,selective and effective clearance of senescent cells of different cell types has proven ***,we developed a prodrug strategy to design a new compound based on the increased activity of lysosomalβ-galactosidase (β-gal),a primary characteristic of senescent *** prodrug SSK1 is specifically activated by β-gal and eliminates mouse and human senescent cells independently of senescence inducers and cell *** aged mice,our compound effectively cleared senescent cells in different tissues,decreased the senescence-and age-associated gene signatures,attenuated low-grade local and systemic inflammation,and restored physical *** results demonstrate that lysosomalβ-gal can be effectively leveraged to selectively eliminate senescent cells,providing a novel strategy to develop anti-aging interventions.

Melanoma is visible unlike other types of cancer, but it is still challenging to diagnose correctly because of the difficulty in distinguishing between benign nevus and melanoma. We conducted a robust investigation of melanoma, identifying considerable differences in local elastic properties between nevus and melanoma tissues by using atomic force microscopy (AFM) indentation of histological specimens. Specifically, the histograms of the elasticity of melanoma displayed multimodal Gaussian distributions, exhibiting the heterogeneous mechanical properties, in contrast with the unimodal distributions of elasticity in the benign nevus. We identified this notable signature was consistent regardless of blotch incidence by sex, age, anatomical site (e.g., thigh, calf, arm, eyelid, and cheek), or cancer stage (I, IV, and V). In addition, we found that the non-linearity of the force-distance curves for melanoma is increased compared to benign nevus. We believe that AFM indentation of histological specimens may technically complement conventional histopathological analysis for earlier and more precise melanoma detection. © 2021, CC BY.

Cancer is the second leading cause of death in the *** major treatments for cancer management include surgery,cytotoxic chemotherapy,targeted therapy,radiation therapy,endocrine therapy and *** the endeavors and achievements made in treating cancers during the past decades,resistance to classical chemotherapeutic agents and/or novel targeted drugs continues to be a major problem in cancer *** resistance,either existing before treatment(intrinsic)or generated after therapy(acquired),is responsible for most relapses of cancer,one of the major causes of death of the *** among patients and tumors,and the versatility of cancer to circumvent therapies make drug resistance more challenging to deal *** understanding the mechanisms of drug resistance is required to provide guidance to future cancer treatment and achieve better *** this review,intrinsic and acquired resistance will be *** addition,new discoveries in mechanisms of drug resistance will be ***,we will highlight roles of ATP in drug resistance by discussing recent findings of exceptionally high levels of intratumoral extracellular ATP as well as intracellular ATP internalized from extracellular *** complexity of drug resistance development suggests that combinational and personalized therapies,which should take ATP into consideration,might provide better strategies and improved efficacy for fighting drug resistance in cancer.

Fundamental metabolic functions depend upon the endoplasmic reticulum (ER), as this organelle plays a central role in maintaining cellular homeostasis. Different physiological and pathological conditions can result in the accumulation of misfolded/unfolded proteins, which this accumulation causes ER stress. These pathological conditions can lead to disease and are concerning for species used for aquaculture. In this comprehensive article, we review studies in cultured species that demonstrate the presence of ER stress to understand conditions and events that may underlie this toxicological outcome. Literature indicates that ER stress can be induced by exposure to pollution, environmental factors (salinity, ammonia, nitrate, hypoxia, and temperature), nutritional changes (quality and quantity of protein, lipid, carbohydrate, vitamins, and minerals), and pathogens. ER stress in aquatic species has been demonstrated through tissue histology and microscopy, gene expression analysis, and other omics approaches. In terms of cell signaling for ER stress, the most common gene indicators identified in aquaculture species include grp78, ire1, perk, chop, erol, atf4, atf6, xbp1, and eif2. The ER stress should be minimized in order to divert more energy for individual growth and achieve sustainable and profitable aquaculture. Here, we provide an overview of ER stress in aquatic species and suggest future directions for research. © 2025 John Wiley & Sons Australia, Ltd.
