Diabetes mellitus is one of the most pressing health concerns because so many people are afflicted by its disabling symptoms. Factors such as age, excess body fat, insufficient physical activity, a history of diabetes in one's family, a sedentary lifestyle, an unhealthy diet, hypertension, etc., all increase the likelihood of developing diabetes mellitus. Health complications are more common in people with diabetes, including cardiovascular disease, renal failure, stroke, blindness, and nerve injury. To validate a diagnosis of diabetes, hospitals typically perform a battery of procedures on the patient. Big data analytics has many vital applications in the healthcare sector. Numerous large computer systems are used in the healthcare sector. With the help of big data analytics, researchers can sift through mountains of data in search of previously unseen patterns and insights. Current techniques have a poor degree of precision in classification and forecast. While previous research has focused on factors such as glucose, body mass index, age, insulin, etc., the proposed model takes these into account and also the other factors that may be more relevant to the development of diabetes. The newer sample is superior to the older one based on categorization accuracy. A workflow algorithm for diabetes prognosis is also required to improve the accuracy.

Skin photosensitization is a common challenge following intravenous administration of many photodynamic therapy (PDT) drugs, typically lasting days, weeks, or months in laboratory animals and patients. Symptoms of photosensitivity manifest as erythema and edema on skin exposed to sunlight or bright artificial lighting. Recent efforts using nanocarriers to increase photosensitizer accumulation in tumors have also been shown to reduce skin photosensitivity. We previously developed phototheranostic PORPHYSOME (PS) nanoparticles self-assembled from porphyrin–lipid conjugates and capable of potent anti-tumor PDT. Here, we demonstrate in a nonpigmented rat skin model that PS exhibit less severe and shorter-lasting skin photosensitivity compared with an equivalent drug dose of porfimer sodium (PHO), the canonical first-generation PDT drug. At 2, 4, 8, and 12 days post intravenous injection, depilated skin was exposed to escalating doses of simulated solar light. Light exposure 4 days post-injection showed markedly reduced symptoms of skin photosensitivity with PS than PHO. By Day 8, the minimal dose of light eliciting any kind of skin reaction was significantly higher with PS than PHO, and by Day 12, there was no detectable skin response with PS. These differences were attributed to altered intradermal distribution and faster clearance of PS vs. PHO in rat skin. © 2025 The Author(s). Magnesium and its alloys are biodegradable and biocompatible with elastic modulus close to the natural bone which makes them suitable as an orthopedic implant. Poor corrosion resistance and high hydrogen evolution rate are two factors hindering their use as commercial implants. To overcome these challenges, a biocompatible magnesium phosphate coating was deposited using a single step hydrothermal method. Scanning Electron Microscopy (SEM/EDX) and X-Ray Diffraction (XRD) methods were used to analyze the structural and chemical compositions of these coatings. In addition, investigation on their adhesion, degradation, cellular proliferation, and gene expression were studied extensively. SEM analysis confirmed a uniform and defect-free deposition occurs in hydrothermal conditions. XRD validated the presence of Periclase (MgO), magnesium phosphate (Mg3(PO4)2) and Brucite (Mg (OH)2) phases. Degradation studies revealed the hydrogen evolution rate is remarkably decreased and corrosion properties were considerably improved as compared to the bare magnesium alloy. In-Vitro biocompatibility testing with pre-osteoblast (MC3T3-E1) via cell viability assay (Alamar blue reduction test) portrayed almost 100% host cell proliferation both in coated and positive control samples after 8 days. Comparing coated and untreated magnesium alloy in SEM images, greater cell adhesion was seen to the coated surface. It is concluded that magnesium phosphate coatings can be used commercially as they are compact, highly crystalline, defect-free, bioresorbable, cytocompatibility, well-adhered, and protective in nature. Hence, phosphate-coated magnesium alloy is qualified to be used to protect biodegradable metallic implants. © 2022, The Authors. All rights reserved.

Diffractive optical elements (DOEs) have a wide range of applications in optics and photonics, thanks to their capability to perform complex wavefront shaping in a compact form. However, widespread applicability of DOEs is still limited, because existing fabrication methods are cumbersome and expensive. Here, we present a simple and cost-effective fabrication approach for solid, high-performance DOEs. The method is based on conjugating two nearly refractive index-matched solidifiable transparent materials. The index matching allows for extreme scaling up of the elements in the axial dimension, which enables simple fabrication of a template using commercially available 3D printing at tens-of-micrometer resolution. We demonstrated the approach by fabricating and using DOEs serving as microlens arrays, vortex plates, including for highly sensitive applications such as vector beam generation and super-resolution microscopy using MINSTED, and phase-masks for three-dimensional single-molecule localization microscopy. Beyond the advantage of making DOEs widely accessible by drastically simplifying their production, the method also overcomes difficulties faced by existing methods in fabricating highly complex elements, such as high-order vortex plates, and spectrum-encoding phase masks for microscopy. © 2023, CC BY-NC-SA.

Mesenchymal stromal/stem cell (MSC)-derived exosomes are nanoscale extracellular vesicles that have emerged as promising candidates for therapeutic and diagnostic applications because of their unique bioactive cargo, including proteins, lipids, and nucleic acids. These vesicles mitigate concerns of immunogenicity and tumorigenicity associated with MSC-based therapies and offer enhanced stability, higher scalability, and ease of modification. However, the use of MSC-derived exosomes in clinical practice is associated with challenges, including difficulties in isolation, characterization, and standardization. This review explores the biogenesis and structural properties of MSC-derived exosomes and discusses the molecular mechanisms underlying their therapeutic effects. It also discusses ongoing clinical trials on their applications in cancer, cardiovascular, neurological, and regenerative medicine. Preclinical and clinical data have demonstrated the potential of MSC-derived exosomes in enhancing tissue repair, reducing inflammation, and modulating immune responses. Despite these advancements, gaps in scalable production methods, regulatory guidelines, and therapeutic consistency must be addressed. Future innovations in bioengineering, manufacturing, and regulatory frameworks are essential to realize the full potential of MSC-derived exosomes in mainstream medicine. © 2025 Elsevier Inc.

Rare earth elements (REEs) are exploited for global use in manufacturing. Such activities result in their release into the environment and the transformation into more stable phosphate deposition. The objective of this study was to evaluate molecular and behavioral changes of zebrafish exposed to the synthesized terbium phosphate (TbPO4) at concentrations of 10, 20, and 50 mg/L and to determine its potential for neurotoxicity. Metabolomics related to neurotransmitters, and assessment of transcripts and proteins were conducted to uncover the molecular mechanisms underlying TbPO4 with emphasis on neurotransmitter systems. Exposure to 20 mg/L TbPO4 induced larval hyperactivity and perturbed the cholinergic system in zebrafish. Based on metabolomics related to neurotransmitters, dopamine (DA), serotonin (5-HT), and many of their precursors and metabolites were decreased in abundance by TbPO4. In addition, the expression levels of transcripts related to the synthesis, transport, receptor binding, and metabolism of DA and 5-HT were analyzed at the mRNA and protein levels. Transcript and protein levels for tyrosine hydroxylase (TH), the rate-limiting enzyme for DA synthesis, were down-regulated in larval fish. Monoamine oxidase (MAO), an enzyme that catabolizes monoamines DA and 5-HT, was also reduced in mRNA abundance. We hypothesize that DA synthesis and monoamine metabolism are associated with behavioral alterations. This study elucidates putative mechanisms and exposure risks to wildlife and humans by characterizing phosphatic REE-induced neurotoxicity in developing zebrafish. © 2023 Elsevier Ltd

Cholera is a highly contagious and lethal waterborne disease induced by an infection with Vibrio cholerae (V. cholerae) secreting cholera toxin (CTx). Cholera toxin subunit B (CTxB) from the CTx specifically binds with monosialo-tetra-hexosyl-ganglioside (GM1) found on the exterior cell membrane of an enterocyte. Owing to the pathological process of CTx, a biomimetic electrochemical biosensor (CCB) was developed by applying GM1-expressing Caco-2 cell membrane (CCM) on the electrode surface of the biosensor for CTx detection. Briefly, the electrode surface was functionalized with CCM using the vesicle fusion method. The specific binding between GM1 and CTxB on the electrode surface was verified using confocal laser scanning microscopy and atomic force microscopy. The CTxB detection performances of the CCB, including sensitivity, selectivity, and long-term stability, were determined using electrochemical impedance spectroscopy (EIS). Based on the results, the CCB had an excellent limit of detection of ~11.46 nM and a detection range spanning 100 ng/mL - 1 mg/mL. Further, the CCB had high selectivity for the CTxB compared to different interfering molecules, which are abundant constituents of intestinal fluid and fecal matter in the human body. The long-term stability of the CCBs was also verified for 3 weeks using EIS. Overall, the CCB was found to have an excellent potential for application to point-of-care and cost-effective testing for CTxB detection in developing countries. © 2022, The Authors. Neuroimmune diseases are a group of disorders that occur due to the dysregulation of both the nervous and immune systems, and these illnesses impact tens of millions of people worldwide. However, patients who suffer from these debilitating conditions have very few FDA-approved treatment options. Neuroimmune crosstalk is important for controlling the immune system both centrally and peripherally to maintain tissue homeostasis. This review aims to provide readers with information on how natural products modulate neuroimmune crosstalk and the therapeutic implications of natural products, including curcumin, epigallocatechin-3-gallate (EGCG), ginkgo special extract, ashwagandha, Centella asiatica, Bacopa monnieri, ginseng, and cannabis to mitigate the progression of neuroimmune diseases, such as Alzheimer's disease, multiple sclerosis, amyotrophic lateral sclerosis, Parkinson's disease, depression, and anxiety disorders. The majority of the natural products based clinical studies mentioned in this study have yielded positive results. To achieve the expected results from natural products based clinical studies, researchers should focus on enhancing bioavailability and determining the synergistic mechanisms of herbal compounds and extracts, which will lead to the discovery of more effective phytomedicines while averting the probable negative effects of natural product extracts. Therefore, future studies developing nutraceuticals to mitigate neuroimmune diseases that incorporate phytochemicals to produce synergistic effects must analyse efficacy, bioavailability, gut-brain axis function safety, chemical modifications, and encapsulation with nanoparticles. © 2023 Gayathri Wijeweera et al.
