Lower back pain (LBP) is included in the top 10 contributors to the overall disease burden. One of the etiologies of LBP is the intervertebral disc (IVD) degeneration, which in some cases, needs to be replaced with an artificial disc, the so-called total disc replacement (TDR). Unfortunately, TDR might provide post-surgical complications, including the failure in the metal-polymer assembly of TDR. The failure was argued due to the rigidity of the metal part and the shape of TDR that was not fully compliant with the anatomical features. Due to these complications, we proposed PU and monolithic construct to be alternative materials and comply with anatomical relevance. This study designed and developed a monolithic TDR made of PU lattices based on anatomical features. We focused on the IVDs in the lumbar section of the spine. The design step consisted of 3 phases: segmentation of DICOM files of IVDs using a 3D Slicer software, alignment using an Autodesk Meshmixer software, and lattice design using an Autodesk Inventor software. The manufacturing step started with a configuration setup in Simplify3D, software for computer-aided manufacturing (CAM) in 3D printing. The lattice features were fabricated using a Creality CP01 3D printer with two types of PU filaments: TPU 87A and TPU 95A. The printing results were able to construct the external curvature of the IVDs. Unfortunately, the microscopic measurement results showed that struts and holes’ dimensions deviated from the design, which must be resolved in the next studies.

At the inaugural Frontiers in Hydrology Meeting in San Juan, Puerto Rico in the summer of 2022, the Hydrology Section organized a poster session and invited our 2020 and 2021 Classes of AGU Fellows, with the initial goal of both celebrating their careers as well as to provide an opportunity for an informal exchange and connection between the section's early career members and our more senior and established scientists and engineers. Due to the challenges of time zones, virtual poster presentations and other logistics, the formal poster session was adjourned but continued as a hybrid “meet-up” with six of our Section's Fellows (Suzanne Anderson, Paul Brooks, Aaron Packman, Remko Uijlenhoet, Andrew Western, and Xubin Zeng) from around the world. As you will see, what started as an informal chat quickly took deep dives into pressing issues in our section and science in general, including thoughts on how our community values (or in some cases doesn't value) multi- and interdisciplinary accomplishments, critiques of our system of rewards and awards including how we assess publication impacts and finally, a frank and honest discussion of our current efforts to diversify our community and where/why are we still failing. We hope that by sharing this open and impromptu dialogue that these discussions can expand to our entire community, and to encourage future Fellows exchanges such as this to reach our entire community of scientists and engineers. Stroke is a neurological disease and is a significant cause of disability throughout the world. Patients with stroke or post-stroke usually still experience interference in walking in the form of a drop foot caused by atrophy of the anterior tibialis muscle, so they cannot perform dorsiflexion. Drop foot disorders can be treated by rehabilitation using Posterior Leaf Spring Ankle-Foot Orthosis (PLS AFO). PLS AFO has the advantage of a higher level of flexibility compared to other AFO types. The primary purpose of this research is to observe the level of resilience based on the influence of AFO thickness. This study uses the finite element analysis of three model variations in the initial contact, midstance, terminal stance, and calf loading phases. The material used in this study was carbon fiber. The simulation will result in the distribution of stress, strain, and deformation values in each AFO model. The simulation results show that AFO resistance is affected by variations in AFO thickness where the increase in strength is proportional to the increase in AFO thickness. The AFO with a thickness of 10 mm is the most suitable for the tibialis anterior atrophy case.

We evaluated the concentration of copper (Cu), zinc (Zn), mercury (Hg), nickel (Ni), chromium (Cr), vanadium (V), and cobalt (Co) in the mangrove tissues (leaf and root) and adjusted sediments, as well as seawater samples in the north coast of the Persian Gulf from three counties (Mahshahr, Bord Khun, Asaluyeh). Assaluyeh and Mahshahr were selected because of their proximity to the oil and petrochemical industries and Bord Khun was considered for being away from pollution sources. The metal detected in highest concentration in mangrove leaves and roots was Cu (Asaluyeh: 1.507 μg/g and 1.198 μg/g, Bord Khun: 1.812 and 1.756 μg/g, and Mahshahr: 0.880 and 1.617 μg/g, for leaves and roots, respectively). For sediments, Cr (among the metals) had the highest concentration (1.325 μg/g) in Asaluyeh, whereas in Bord Khun and Mahshahr Ni was predominant: 2.542 and 2.237 μg/g, respectively. For seawater, Cu was the most abundant metal and Hg was the one with the lowest concentration. The bio-concentration factor for Zn and Cu revealed that mangroves in the study areas have suitable potential for phytoremediation of these two metals. Translocation factors from the roots to leaves also indicated higher transportability for Zn and Cu through the plant tissues, compared to the other metals. Ecological indexes quantified the contamination of sediments and revealed a low level of heavy metal accumulation in sediments of the north of the Persian Gulf. Human risk hazards appeared low in all the areas under study. Gas and petrochemical activities can be considered as a potential source of the discharge of heavy metals into the aquatic environment. © 2021 Elsevier B.V.

We establish an open framework for developing plan optimization models for knowledge-based planning (KBP) in radiotherapy. Our framework includes reference plans for 100 patients with head-and-neck cancer and high-quality dose predictions from 19 KBP models that were developed by different research groups during the OpenKBP Grand Challenge. The dose predictions were input to four optimization models to form 76 unique KBP pipelines that generated 7600 plans. The predictions and plans were compared to the reference plans via: dose score, which is the average mean absolute voxel-by-voxel difference in dose a model achieved;the deviation in dosevolume histogram (DVH) criterion;and the frequency of clinical planning criteria satisfaction. We also performed a theoretical investigation to justify our dose mimicking models. The range in rank order correlation of the dose score between predictions and their KBP pipelines was 0.50 to 0.62, which indicates that the quality of the predictions is generally positively correlated with the quality of the plans. Arterial stiffening is a significant predictor of cardiovascular disease development and mortality. In elastic arteries, stiffening refers to the loss and fragmentation of elastic fibers, with a progressive increase in collagen fibers. Type VIII collagen (Col-8) is highly expressed developmentally, and then once again dramatically upregulated in aged and diseased vessels characterized by arterial stiffening. Yet its biophysical impact on the vessel wall remains unknown. The purpose of this study was to test the hypothesis that Col-8 functions as a matrix scaffold to maintain vessel integrity during extracellular matrix (ECM) development. These changes are predicted to persist into the adult vasculature, and we have tested this in our investigation. Through our in vivo and in vitro studies, we have determined a novel interaction between Col-8 and elastin. Mice deficient in Col-8 (Col8^−/−) had reduced baseline blood pressure and increased arterial compliance, indicating an enhanced Windkessel effect in conducting arteries. Differences in both the ECM composition and VSMC activity resulted in Col8^−/− carotid arteries that displayed increased crosslinked elastin and functional distensibility, but enhanced catecholamine-induced VSMC contractility. In vitro studies revealed that the absence of Col-8 dramatically increased tropoelastin mRNA and elastic fiber deposition in the ECM, which was decreased with exogenous Col-8 treatment. These findings suggest a causative role for Col-8 in reducing mRNA levels of tropoelastin and the presence of elastic fibers in the matrix. Moreover, we also found that Col-8 and elastin have opposing effects on VSMC phenotype, the former promoting a synthetic phenotype, whereas the latter confers quiescence. These studies further our understanding of Col-8 function and open a promising new area of investigation related to elastin biology. © 2021 The Author(s)

Multivalent interactions between protein‐oligomer‐fused binding pairs allowed highly monodisperse assembly of two protein oligomers, where spacing between oligomers can be widely varied. Two different oligomers or three oligomers can also be discretely assembled into protein layer structures via this *** approaches to design monodisperse protein assemblies require rigid, tight, and symmetric interactions between oligomeric protein units. Herein, we introduce a new multivalent‐interaction‐driven assembly strategy that allows flexible, spaced, and asymmetric assembly between protein oligomers. We discovered that two polygonal protein oligomers (ranging from triangle to hexagon) dominantly form a discrete and stable two‐layered protein prism nanostructure via multivalent interactions between fused binding pairs. We demonstrated that protein nano‐prisms with long flexible peptide linkers (over 80 amino acids) between protein oligomer layers could be discretely formed. Oligomers with different structures could also be monodispersely assembled into two‐layered but asymmetric protein nano‐prisms. Post-inflammatory hyperpigmentation (PIH) occurs when excess melanin is produced in response to skin irritation or injury, resulting in uneven pigmentation. This study evaluates the cosmeceutical potential of ethanolic extracts from eight Thai herbs— Curcuma xanthorrhiza (CX), Quercus infectoria (QI), Calophyllum inophyllum (CI), Caesalpinia sappan (CS), Garcinia hanburyi (GH), Punica granatum (PG), Cotylelobium lanceolatum (CL), and Strychnos nux-vomica (SN)—to treat PIH through antioxidant, anti-inflammatory, antibacterial, and anti-melanogenesis properties. Phytochemical analysis revealed that CX and QI contained high levels of phenolics and flavonoids, correlating with their strong antioxidant activity. CX also protected against hydrogen peroxide (H₂O₂)-induced oxidative stress in a yeast-based model and prevented DNA damage in vitro, helping to mitigate cellular injury that can lead to melanin overproduction. Anti-inflammatory assays showed that both CX and QI significantly reduced nitric oxide and interleukin-6 (IL-6) levels without causing cytotoxicity, supporting their role in treating inflammation-related PIH. The extracts also demonstrated selective antibacterial activity against Staphylococcus aureus (MSSA and MRSA) and Cutibacterium acnes , while sparing Staphylococcus epidermidis , an important member of the skin microbiome. CI and CX significantly inhibited C. acnes , a key contributor to acne-induced PIH. Finally, anti-melanogenesis assays revealed that QI and SN effectively inhibited α-MSH-induced melanin production. These findings suggest that CX, QI, and CI are promising natural cosmeceutical agents for addressing PIH by modulating oxidative stress, inflammation, microbial imbalance, and melanin overproduction, while maintaining skin microbiota integrity.
