Endosomal escape remains a central issue limiting the high protein expression of mRNA therapeutics. Here, we present second near-infrared (NIR-II) lipid nanoparticles (LNPs) containing pH activatable NIR-II dye conjugated lipid (Cy-lipid) for potentiating mRNA delivery efficiency via a s timulus-responsive p hotothermal-promoted e ndosomal e scape d elivery (SPEED) strategy. In acidic endosomal microenvironment, Cy-lipid is protonated and turns on NIR-II absorption for light-to-heat transduction mediated by 1064 nm laser irradiation. Then, the heat-promoted LNPs morphology change triggers rapid escape of NIR-II LNPs from the endosome, allowing about 3-fold enhancement of enhanced green fluorescent protein (eGFP) encoding mRNA translation capacity compared to the NIR-II light free group. In addition, the bioluminescence intensity induced by delivered luciferase encoding mRNA in the mouse liver region shows positive correlation with incremental radiation dose, indicating the validity of the SPEED strategy.

Background: Recent studies have reported that air pollution is related to kidney diseases. However, the global evidence on the risk of death from acute kidney injury (AKI) owing to air pollution is limited. Therefore, we investigated the association between short-term exposure to air pollution—particulate matter ≤ 2.5 μm (PM2.5), ozone (O3), and nitrogen dioxide (NO2)—and AKI-related mortality using a multi-country dataset. Methods: This study included 41,379 AKI-related deaths in 136 locations in six countries during 1987–2018. A novel case time-series design was applied to each air pollutant during 0–28 lag days to estimate the association between air pollution and AKI-related deaths. Moreover, we calculated AKI deaths attributable to non-compliance with the World Health Organization (WHO) air quality guidelines. Results: The relative risks (95% confidence interval) of AKI-related deaths are 1.052 (1.003, 1.103), 1.022 (0.994, 1.050), and 1.022 (0.982, 1.063) for 5, 10, and 10 µg/m3 increase in lag 0–28 days of PM2.5, warm-season O3, and NO2, respectively. The lag-distributed association showed that the risk appeared immediately on the day of exposure to air pollution, gradually decreased, and then increased again reaching the peak approximately 20 days after exposure to PM2.5 and O3. We also found that 1.9%, 6.3%, and 5.2% of AKI deaths were attributed to PM2.5, warm-season O3, and NO2 concentrations above the WHO guidelines. Conclusions: This study provides evidence that public health policies to reduce air pollution may alleviate the burden of death from AKI and suggests the need to investigate the several pathways between air pollution and AKI death. © 2023 The Author(s)

Evaluation of hemiplegic gait using accelerometer

The study aims to distinguish hemiplegic gait and normal gait using simple wearable device and classification algorithm. Thus, we developed a wearable system equipped three axis accelerometer and three axis gyroscope. The developed wearable system was verified by clinical experiment. In experiment, twenty one normal subjects and twenty one patients undergoing stroke treatment were participated. Based on the measured inertial signal, a random forest algorithm was used to classify hemiplegic gait. Four-fold cross validation was applied to ensure the reliability of the results. To select optimal attributes, we applied the forward search algorithm with 10 times of repetition, then selected five most frequently attributes were chosen as a final attribute. The results of this study showed that 95.2% of accuracy in hemiplegic gait and normal gait classification and 77.4% of accuracy in hemiplegic-side and normal gait classification. © 2017 The Korean Institute of Electrical Engineers.

Sensitivity analysis of gas distribution pipeline risk assessment methodology in Indonesia

Sensitivity is the effect of changes in any variables on changes in the risk results. The most common Risk Assessment Methodology in Indonesia is Muhlbauer, Modified Muhlbauer & Risk-Based Inspection. This study evaluated six natural gas distribution pipeline segments. In general, there are two objectives, firstly comparing the risk result between Risk Assessment Methodology. The following objectives are to compare the sensitivity. The Risk Assessments methodology is translated into a mathematic model and computed in Monte Carlo based simulation software. The risk value result shows that in high risk & medium risk pipeline segments, all methodology is valid with 24% and 13% deviation, respectively. The sensitivity analysis result shows that Muhlbauer & Modified Muhlbauer’s methodology is sensitive to third party activity. Risk-Based Inspection is sensitive to intrinsic damage mechanisms. This sensitivity data can be adopted to develop a more sensitive Risk Assessment Methodology for gas pipeline in the future.

In nature, many mysterious creatures capable of deformation camouflage, color camouflage, and self-healing have inspired scientists to develop various biomimetic soft robots. However, the systematic integration of all the above functionalities into a single soft actuator system still remains a challenge. Here we chemically introduce a multi-stimuli-responsive tetraarylsuccinonitrile (TASN) chromophore into a liquid crystal elastomer (LCE) network through a facile thiol-ene photoaddition method. The obtained TASN-LCE soft actuators not only exhibit reversible shape-morphing and reversible color-changing behavior in response to heat and mechanical compression, but also show excellent self-healing, reprogramming and recycling characteristics. We hope that such a TASN-LCE actuator system endowed with dynamic distortion, thermo- and mechano-chromic camouflage, and self-healing functionalities would pave the way for further development of multifunctional biomimetic soft robotic devices.

Penaeid shrimps are widely distributed from Indian to western Pacific Oceans and some which are economically important. In this study, we reported full mitochondrial genome of an endemic shrimp species,Penaeus acehensis, which inhabits exclusively in the coastal water of Aceh, Indonesia. Full length of circular mitogenome ofP. acehensiswas 15,991 bp in length, which contained 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and a control region. Start codons of all protein-coding genes were ATN except for COX1 in which ACG was used. Incomplete stop codon (T- -) was found in five genes including COX2, COX3, NAD5, NAD4, and NAD4L. Among its relatives,P. acehensiswas most closely related toPenaeus monodonshowing 89% sequence identity in its mitogenome, which was corresponding to morphological analysis. Phylogenetic tree result showed thatP. acehensiswas clustered together with those were distributed in Indo-West Pacific region (clade II), which is distinct from Eastern Pacific region (clade I).

Recently,the potential role of vitamins in cancer therapy has attracted considerable research ***,the reported findings are inconsistent,with limited information on the biochemical and molecular interactions of different vitamins in various cancer ***,the presence of vitamin receptors in tumor cells suggests that vitamins play a significant role in the molecular and biochemical interactions in ***,studies on the efficacy of vitamin supplementation and dosage levels on tumor progression and mortality risk have yielded inconsistent ***,molecular and biochemical investigations have reported the function of vitamins in the proliferation,growth,and invasiveness of tumor cells,as well as in cell cycle arrest and inflammatory ***,different vitamins may regulate the cancer microenvironment by activating various molecular *** significantly affect immunological function,antioxidant defense,inflammation,and epigenetic control,and can improve treatment outcomes by affecting cell behavior and combating stress and DNA ***,further research is necessary to confirm the efficacy of vitamins,establish ideal dosages,and develop effective cancer prevention and treatment *** supplementation plans guided by medical knowledge are crucial to achieving optimal results in clinical and preclinical *** this review,we critically evaluated the effects of different vitamins on the risk and development of ***,we examined the potential of vitamin supplements to enhance the efficacy of drug therapy and counteract resistance mechanisms that often arise during cancer treatment.
